    Notes: The effect of acute O3 exposure (5 h at 150 ppb) on photosynthetic electron transport and the thylakoid polypeptide pattern has been studied in 2-month-old cuttings of two poplar clones (Populus deltoides×maximowiczii clone Eridano and P.×euramericana clone I-214), known for their differential response to O3 in terms of visible symptoms. The more sensitive Eridano developed severe blackish bifacial necrosis concentrated in the interveinal areas of the basal and intermediate leaves, while in the less sensitive I-214 clone, visible symptoms were slight or entirely absent. A generalized, significant decrease in leaf pigment content was shown by both of the stressed clones, although the decline in β-carotene content was much more marked in Eridano than in I-214. Activation of the xanthophyll cycle was not detected in either clone. In both clones, immunoblot analysis of the thylakoid proteins showed a decrease in the levels of all the PSII polypeptides tested following O3 exposure, whereas the PsaA/B protein in the PSI core was more severely affected in Eridano than in I-214. No changes in the chloroplast structure could be seen on electron microscopic study in either clone. The only visible difference was a great accumulation of starch grains in Eridano. All of these results suggest that O3 exposure induces alterations in the photosynthetic apparatus in both poplar clones, Eridano being affected to a greater extent than I-214.

    Notes: The photosynthetic response was studied in two clones (Populus deltoides × maximowiczii Eridano and Populus × euramericana I-214), known for their differential response to ozone (O3) in terms of visible symptoms, when exposed to O3 (60 nl l−1 5 h day−1, 7 and 15 days). The photosynthetic ability was tested using gas exchange and chlorophyll fluorescence analysis. O3 caused a decrease in the CO2 assimilation rate at light saturation level in mature leaves of both clones. Alterations of Chl fluorescence parameters, in particular the Fv/Fm ratio and non-photochemical quenching were also observed. The effects were similar for both clones and it could not be concluded that differential effects on electron transport capacity were responsible for the observed reduction in photosynthesis. The reduction of photosynthetic rate in Eridano was due mainly to a reduced mesophyll activity, as evidenced by the increase in intercellular CO2 concentration and the minimal changes in stomatal conductance. In contrast, in I-214, stomatal effects were primarily responsible, although effects on the mesophyll cannot be excluded. Data obtained indicate that the effects observed at the mesophyll level may be attributed to indirect effects caused by membrane disorders.

    Notes: CO2 assimilation, xanthophyll cycle pigments and PSII efficiency were analyzed in two different ages of pumpkin leaves (Cucurbita pepo L. cv. Ambassador) exposed to 150 nmol mol-1 of ozone (5 days, 5 h day-1). Gas-exchange measurements revealed a reduction in CO2 assimilation and stomatal conductance, accompanied by an increase in the intercellular CO2 concentration both in young and in mature leaves as compared to their respective controls. In both leaves, F0 remained unchanged, while Fm and the Fv/Fm ratio decreased after O3 fumigation, indicating that ozone may induce an alteration in the capability of photosystem II (PSII) to reduce the primary acceptor QA. In the mature leaves the photochemical quenching (qp) was significantly lowered by the pollutant, but this was not the case in the young leaves where qp did not change. In both mature and young ozonated pumpkin leaves, the development of non-photochemical quenching caused a decrease in the PSII photochemical rate, as shown by the correlation between Fv/Fm and the de-epoxidation state of dark-adapted leaves. Decreases in the Fv/Fm ratio are generally attributed to damage to the PSII reaction centre, apart from the down-regulation of the capacity of PSII electron transport. While in young ozonated leaves the decrease in the Fv/Fm ratio was not associated with damage to the D1 protein, in mature ozonated pumpkin leaves, the decrease in the Fv/Fm was accompanied by a significant decline in the D1 content. In conclusion, ozone exposure induces alterations in the light reactions of photosynthesis in both young and mature leaves. However, in young leaves the engagement of the xanthophyll cycle appears to counteract ozone effects against the photosynthetic apparatus as demonstrated by the absence of damage to the D1 protein. On the other hand, the loss of D1 protein in mature fumigated leaves suggests that the activation of the xanthophyll cycle is not sufficient to prevent photoinhibition, probably because a physiological state of senescence adds to the oxidative stress.

    Notes: Abstract An automatic system was installed for continuous analyses of ozone, sulphur dioxide, nitrogen monoxide and nitrogen dioxide in an experimental orchard with a canopy ofPrunus cerasifera plants in summer 1993. Air samples from three elevations (0.8 m, 1.6 m and 3 m above ground) were sequentially analyzed. Ozone concentrations above the canopy were usually higher than within the canopy; their relationships with stomatal resistance have been investigated. Sulphur dioxide levels were negligible. Nitrogen oxides showed a complex profile, with no particular trend, likely due to a reciprocal exchange between the atmosphere and the ground surface.

    Notes: Abstract An innovative miniaturized kit based on the use of 2-week-old ozone-supersensitive tobacco germlings (Nicotiana tabacum L. cv. Bel-W3) raised in tissue culture plates was utilized, in conjunction with four calibrated automatic analyzers, to monitor the distribution of phytotoxic ground level ozone in Tuscany during the summer of 1993 at 27 sites differing in nature. Germlings of ozone-resistant Bel-B tobacco were also included in the protocol. The intensity of visible injury on the cotyledons of Bel-W3 was linearly correlated with several ozone statistical descriptors. The occurrence of phytotoxic levels of photochemical ozone was detected in all the monitoring sites, which included rural and remote areas, whose local sources of pollution were negligible. The suitability of the new methodology for low-cost, space-saving, user-friendly monitoring of ozone on a large geographical scale is discussed.
