    Description: The Qinghai-Tibet Plateau (QTP) is characterized by a vast number of frozen and unfrozen freshwater reservoirs, which is why it is also called “the third pole” of the Earth or “Asian Water Tower”. We analyzed testate amoeba (TA) biodiversity and corresponding protozoic biosilicification in lake sediments of the QTP in relation to environmental properties (freshwater conditions, elevation, and climate). As TA are known as excellent bio-indicators, our results allowed us to derive conclusions about the influence of climate warming on TA communities and microbial biogeochemical silicon (Si) cycling. We found a total of 113 TA taxa including some rare and one unknown species in the analyzed lake sediments of the QTP highlighting the potential of this remote region for TA biodiversity. 〉1/3 of the identified TA taxa were relatively small (〈30 μm) reflecting the relatively harsh environmental conditions in the examined lakes. TA communities were strongly affected by physico-chemical properties of the lakes, especially water temperature and pH, but also elevation and climate conditions (temperature, precipitation). Our study reveals climate-related changes in TA biodiversity with consequences for protozoic biosilicification. As the warming trend in the QTP is two to three times faster compared to the global average, our results provide not only deeper insights into the relations between TA biodiversity and environmental properties, but also predictions of future developments in other regions of the world. Moreover, our results provide fundamental data for paleolimnological reconstructions. Thus, examining the QTP is helpful to understand microbial biogeochemical Si cycling in the past, present, and future.

    Description: Background: FABP1 has been reported to possess strong antioxidant properties. Upon successful transfection of the Chang cell line, which has undetectable FABP1 mRNA levels, with human FABP1 cDNA, the Chang cells were shown to express FABP1. Using the transfected and control (normal) Chang cells and subjecting them to oxidative stress, transfected cells were reported to be associated with enhanced cell viability. This study extends those observations by investigating the effect of FABP1 on acetaminophen (AAP)-induced hepatotoxicity. We hypothesized that presence of FABP1 would enhance cell viability compared to control cells (vector transfected cells). Methods: Following AAP treatment of Chang FABP1 transfected and control cells, cell viability, oxidative stress, and apoptosis were evaluated using lactate dehydrogenase (LDH) release, the fluorescent probe DCF, and Bax expression, respectively. Results: FABP1 cDNA transfected cells showed greater resistance against AAP toxicity than vector transfected cells. Significantly lower LDH levels (p 〈 0.05) were observed as were lower DCF fluorescence intensity (p 〈 0.05) in FABP1 cDNA transfected cells compared to vector transfected cells. FABP1 expression also attenuated the expression of Bax following AAP induced toxicity. Conclusion: FABP1 attenuated AAP-induced toxicity and may be considered a cytoprotective agent in this in vitro model of drug induced oxidative stress.

    Description: Leucine-rich repeat-containing G protein–coupled receptor 5 (LGR5) is a bona fide marker of adult stem cells in several epithelial tissues, most notably in the intestinal crypts, and is highly up-regulated in many colorectal, hepatocellular, and ovarian cancers. LGR5 activation by R-spondin (RSPO) ligands potentiates Wnt/β-catenin signaling in vitro; however, deletion of LGR5 in stem cells has little or no effect on Wnt/β-catenin signaling or cell proliferation in vivo. Remarkably, modulation of LGR5 expression has a major impact on the actin cytoskeletal structure and cell adhesion in the absence of RSPO stimulation, but the molecular mechanism is unclear. Here, we show that LGR5 interacts with IQ motif-containing GTPase-activating protein 1 (IQGAP1), an effector of Rac1/CDC42 GTPases, in the regulation of actin cytoskeleton dynamics and cell–cell adhesion. Specifically, LGR5 decreased levels of IQGAP1 phosphorylation at Ser-1441/1443, leading to increased binding of Rac1 to IQGAP1 and thus higher levels of cortical F-actin and enhanced cell–cell adhesion. LGR5 ablation in colon cancer cells and crypt stem cells resulted in loss of cortical F-actin, reduced cell–cell adhesion, and disrupted localization of adhesion-associated proteins. No evidence of LGR5 coupling to any of the four major subtypes of heterotrimeric G proteins was found. These findings suggest that LGR5 primarily functions via the IQGAP1–Rac1 pathway to strengthen cell–cell adhesion in normal adult crypt stem cells and colon cancer cells.

    Description: Using the daily electric power load and meteorological data over the period from January 2006 to September 2010 in Beijing, the general features of the electric power load and the main factors affecting this load have been analysed. The results indicate that winter and summer are the two peak power consumption seasons in each year. This finding could be mainly attributed to the consumption of energy for urban residential cooling or heating. In the summer, the daily minimum temperature was most closely correlated with the maximum power load (correlation co-efficient of 0.65, significant at the 0.1% level); the primary effect of temperature on the maximum power load varied largely from 376 to 858 MW °C −1 , and it increased sharply when the daily minimum temperature was higher than 24 °C or the daily maximum temperature was higher than 32 °C. In the winter, the daily maximum temperature was most closely correlated with the power load (−0.45, 0.1% significance), and the primary effect varied from 109 to 391 MW °C −1 . However, the relationship between the power load and the temperature was more complicated in the winter: this relationship varied during the central heating period and its pre- and post-periods. Moreover, the temperature humidity index (THI) could explain more of the variance in the daily maximum power load than any single temperature factor in the summer, while the wind-chill temperature index (WCT) did not contribute to this variance in the winter. Finally, due to the rapid urbanization of Beijing, the power load and the priamry effect increased each year. Copyright © 2012 Royal Meteorological Society

    Description: [1]  In this article, the relationship between the Antarctic Oscillation (AAO) and the tropical cyclone (TC) activity in the Southern Indian Ocean (SIO) was examined. It was found that, on the interannual time scale, the AAO is well linked with the TC activity in the SIO during TC season (December to March). The rank correlation coefficient between the AAO index and the TC frequency in the SIO is 0.37, which is significant at the 95% confidence level. When the AAO is in a positive phase, TC passage in the northwestern coast of Australia (100E°-120°E and 10°S-30°S) increases by approximately 50-100% from the climatology. The increase in the TC passage is primarily the result of more frequent TCs originating in this region due to an enhanced water vapor convergence and ascending motions, which are caused by a cyclonic height anomaly over the western coast of Australia associated with the positive AAO phases. In addition, the AAO-height co-variations, which are essential to the formation of the AAO-TC links in the SIO, were investigated through a historical climate simulation using the Community Climate System Model 4 from the Coupled Model Intercomparison Project Phase 5. The AAO-height links were well reproduced in the simulation. The similarity in the AAO-height links between the observation and the simulation supports the physical robustness of the AAO-TC links in the SIO.

    Description: T cell functional exhaustion during chronic hepatitis B virus (HBV) infection may contribute to the failed viral clearance; however, the underlying molecular mechanisms remain largely unknown. Here we demonstrate that jumonji domain-containing protein 6 (JMJD6) is a potential regulator of T cell proliferation during chronic HBV infection. The expression of JMJD6 was reduced in T lymphocytes in chronic hepatitis B (CHB) patients, and this reduction in JMJD6 expression was associated with impaired T cell proliferation. Moreover, silencing JMJD6 expression in primary human T cells impaired T cell proliferation. We found that JMJD6 promotes T cell proliferation by suppressing the mRNA expression of CDKN3. Furthermore, we have identified platelet derived growth factor-BB (PDGF-BB) as a regulator of JMJD6 expression. PDGF-BB downregulates JMJD6 expression and inhibits the proliferation of human primary T cells. Importantly, the expression levels of JMJD6 and PDGF-BB in lymphocytes from CHB patients were correlated with the degree of liver damage and the outcome of chronic HBV infection treatment. Our results demonstrate that PDGF-BB and JMJD6 regulate T cell function during chronic HBV infection and may provide insights for the treatment strategies for CHB patients.
