    Description: Diurnal variation of net community production (NEP) and net community calcification (NEC) were measured in coral reef and seagrass biomes during October 2012 in the lower Florida Keys using a mesocosm enclosure and the oxygen gradient flux technique. Seagrass and coral reef sites showed diurnal variations of NEP and NEC, with positive values at near-seafloor light levels 〉100-300 µEinstein/m**2/s. During daylight hours, we detected an average NEP of 12.3 and 8.6 mmol O2/m**2/h at the seagrass and coral reef site, respectively. At night, NEP at the seagrass site was relatively constant, while on the coral reef, net respiration was highest immediately after dusk and decreased during the rest of the night. At the seagrass site, NEC values ranged from 0.20 g CaCO3 /m**2/h during daylight to -0.15 g CaCO3/m**2/h at night, and from 0.17 to -0.10 g CaCO3/m**2/h at the coral reef site. There were no significant differences in pH and aragonite saturation states (Omega ar) between the seagrass and coral reef sites. Decrease in light levels during thunderstorms significantly decreased NEP, transforming the system from net autotrophic to net heterotrophic.

    Description: Sea surface salinity (SSS) is the least constrained major variable of the past (paleo) ocean but is fundamental in controlling the density of seawater and thus large-scale ocean circulation. The hydrogen isotopic composition (δD) of non-exchangeable hydrogen of algal lipids, specifically alkenones, has been proposed as a promising new proxy for paleo SSS. The δD of surface seawater is correlated with SSS, and laboratory culture studies have shown the δD of algal growth water to be reflected in the δD of alkenones. However, a large-scale field study testing the validity of this proxy is still lacking. Here we present the δD of open-ocean Atlantic and Pacific surface waters and coincident δD of alkenones sampled by underway filtration. Two transects of approximately 100° latitude in the Atlantic Ocean and more than 50° latitude in the Western Pacific sample much of the range of open ocean salinities and seawater δD, and thus allow probing the relationship between δD of seawater and alkenones. Overall, the open ocean δD alkenone data correlate significantly with SSS, and also agree remarkably well with δD water vs δD alkenone regressions developed from culture studies. Subtle deviations from these regressions are discussed in the context of physiological factors as recorded in the carbon isotopic composition of alkenones. In a best-case scenario, the data presented here suggest that SSS variations as low as 1.2 can be reconstructed from alkenone δD.

    Description: A simple ecosystem model is coupled to a 3-dimensional general circulation model for the North Atlantic. The physical model is based on the Los Alamos Parallel Ocean Program (POP) and forced by climatological monthly mean data. Four biological components(phytoplankton, zooplankton, nutrients and detritus) are incorporated into POP as additional tracers with biological sources and sinks. The model solutions, obtained with different physical and biological parameterizations are compared against monthly mean SeaWiFS colour data averaged over the period 1997-2003 and Levitus's climatological nitrate data. A reference model solution, with constant biological model parameters over the whole basin,underestimates both the average chlorophyll level and its regionalvariability at mid- to high-latitudes. Experiments with a different parameterization of heat and freshwater fluxes, which affects upper ocean mixing, indicate a strong impact of such parameterizations on nutrient supply to the surface layer at high latitudes, but with little impact on simulated chlorophyll. Other experiments where advection of the biological tracers is turned off show basically the same result: strong impact on regional nutrient patterns but a negligible impact on phytoplankton patterns. Only model runs with spatially variable biological parameters, obtained from a previouszero-dimensional ecosystem model calibration on CZCS ocean colour data, could reproduce regional scale patterns in the SeaWiFS imagery. We hypothesize that some of these patterns can be linked to coccolithophore blooms in areas influenced by the N. Atlantic Drift during summer and to effects of temperature on plankton loss ratesduring spring. Future work should focus on identifying the main factors responsible for these spatial patterns and developing the ecosystem models that can capture them.

    Description: Background.   Staphylococcus aureus is a leading cause of superficial and invasive human disease that is often refractory to antimicrobial therapy. Vaccines have the potential to reduce the morbidity, mortality, and economic impact associated with staphylococcal infections. However, single-component vaccines targeting S. aureus have failed to show efficacy in clinical trials. Methods.  A novel glycoengineering technology for creation of a multicomponent staphylococcal vaccine is described. Genes encoding S. aureus capsular polysaccharide (CP) biosynthesis, PglB (a Campylobacter oligosaccharyl transferase), and a protein carrier (detoxified Pseudomonas aeruginosa exoprotein A or S. aureus α toxin [Hla]) were coexpressed in Escherichia coli . Recombinant proteins N-glycosylated with S. aureus serotype 5 or 8 CPs were purified from E. coli. Results.  Rabbits and mice immunized with the glycoprotein vaccines produced antibodies that were active in vitro in functional assays . Active and passive immunization strategies targeting the CPs protected mice against bacteremia, and vaccines targeting Hla protected against lethal pneumonia. The CP-Hla bioconjugate vaccine protected against both bacteremia and lethal pneumonia, providing broad-spectrum efficacy against staphylococcal invasive disease. Conclusions.  Glycoengineering technology, whereby polysaccharide and protein antigens are enzymatically linked in a simple E. coli production system, has broad applicability for use in vaccine development against encapsulated microbial pathogens.
