    Notes: Abstract Four days of fasting in the rat reduced brown-adipose-tissue (BAT) mass, mitochondrial protein, and tissue protein content. Specific binding of guanosine diposphate (GDP) to BAT mitochondria was depressed by 55% in 4d-fasted rats. Rats fasted for 3 d, and then refed a single carbohydrate meal (40 kJ), showed a significant increase in specific GDP-binding (27% above fasted) 24 h later, and a large increase in total binding. Specific activities of cytochrome oxidase and α-glycerophosphate dehydrogenase in BAT mitochondria were not significantly affected by fasting or refeeding. These results suggest that BAT may be partly responsible for the fall in metabolic rate associated with fasting and the delayed increase after carbo-hydrate refeeding. These effects may be due to changes in the mitochondrial proton-conductance pathway in brown fat.

    Notes: Abstract Fatty-acid synthesis has been measured in vivo with3H2O in cafeteria-fed rats exhibiting diet-induced thermogenesis. Synthesis was decreased in brown adipose tissue, the liver, white adipose tissue, and the carcass of the cafeteria-fed animals compared to rats fed the normal stock diet. Whole-body synthesis was also decreased in the cafeteria-fed group. Diet-induced thermogenesis, in contrast to cold-induced non-shivering thermogenesis does not lead to increased fatty-acid synthesis and this is presumably due to the inhibitory effects on lipogenesis of the high dietary fat intake characteristic of cafeteria diets. The results also indicate that the energy cost of body fat deposition in cafeteria-fed rats is lower than in animals fed a low-fat/high-carbohydrate stock diet.

    Notes: Abstract In rats fed a normal (22% protein) diet, injection of clenbuterol (1 mg/kg/d for 21 d) did not affect energy intake, energy expenditure or weight gain, but reduced energetic efficiency, and fat and energy gains and increased body protein content. Presenting a low-protein (8%) diet reduced energy intake, gain and efficiency, body protein content and the mass of the gastrocnemius muscle when compared to rats fed the control diet. Injection of the protein-deficient rats with clenbuterol (1 mg/kg/d for 21 d) caused hypophagia and reduced body weight and energy gains, energy expenditure and total body fat. However, the total body content of protein was not significantly reduced and the percentage of body protein in this protein deficient, clenbuterol-treated group was greater than that of untreated rats on both the high- and low-protein diets. The ratio of body protein to fat following clenbuterol treatment was increased by over 50% in both normal and protein-deficient rats. The results show that in protein deficient animals, clenbuterol treatment may help conserve body protein at the expense of fat, resulting in a smaller, but leaner body mass.

    Notes: Abstract Female dystrophic mice (mdx on C57 Black background) gained weight more rapidly than age-matched controls and had a higher body fat content (% body weight), a slightly lower protein content and a reduced mass of muscle. Chronic treatment (21 d) of the mice with theβ 2-agonist clenbuterol stimulated weight gain in both genotypes without affecting energy intake. Clenbuterol increased the mass of the gastrocnemius and soleus muscle by 13% and 29% in normal and dystrophic mice, respectively, and raised body protein but depressed body fat. Body water and energy content were unaffected by clenbuterol, but the ratio of protein to fat in the carcasses was enhanced by 17% in normal and 56% in dystrophic mice following clenbuterol treatment. Thus, theβ 2-agonist restored the body composition of dystrophic mice to normal and enhanced the protein to fat ratio in both these and normal mice.

    Notes: Summary Measurements of resting and exercising metabolic rate were made on eight subjects (five male, three female) before and after consumption of a standard liquid meal (1.67 MJ). This test was conducted after a day of complete fast and again after a day of overeating (average intake, 19.8 MJ). There were no significant changes in resting or exercising metabolic rate due to the previous day's energy intake. The resting thermic effect (post-prandial rise in metabolic rate) of the standard meal was similar on both test days but the exercising thermic effect was 50% greater after the day of overeating. It was concluded that the metabolic response to food in exercising subjects may be affected by the previous day's energy intake but the overall energetic efficiency of the body at rest and during exercise is unaffected.

    Description: Volatile elements play an important role in many aspects of the physicochemical architecture of sub-volcanic plumbing systems, from the liquid line of descent to the dynamics of magma storage and eruption. However, it remains difficult to constrain the behaviour of magmatic volatiles on short timescales before eruption using established petrological techniques (e.g. melt inclusions); specifically, in the final days to months of magma storage. This study presents a detailed model of pre-eruptive volatile behaviour in the Campi Flegrei system (Italy), through combined analyses of apatite crystals and glass. The deposits of eight eruptions were examined, covering the full spectrum of melt compositions, eruptive styles and periods of activity at Campi Flegrei in the past 15 kyr. Measured apatite compositions are compared with thermodynamic models that predict the evolution of the crystal compositions during different fractional crystallization scenarios, including (1) volatile-undersaturated conditions, (2) H2O-saturated conditions and (3) varying P–T conditions. The compositions of clinopyroxene-hosted and biotite-hosted apatite inclusions are consistent with crystallization under volatile-undersaturated conditions that persisted until late in magmatic evolution. Apatite microphenocrysts show significantly more compositional diversity, interpreted to reflect a mixed cargo of crystals derived from volatile-undersaturated melts at depth and melts that have undergone cooling and degassing in discrete shallow-crustal magma bodies. Apatite microphenocrysts from lavas show some re-equilibration during cooling at the surface. Clinopyroxene-hosted melt inclusions within the samples typically contain 2–4 wt % H2O, indicating that they have been reset during temporary magma storage at 1–3km depth, similar to the depth of sill emplacement during recent seismic crises at Campi Flegrei. Comparable apatite compositional trends are identified in each explosive eruption analysed, regardless of volume, composition or eruption timing. However, apatites from the different epochs of activity appear to indicate subtle changes in the H2O content of the parental melt feeding the Campi Flegrei system over time. This study demonstrates the potential utility of integrated apatite and glass analysis for investigating pre-eruptive volatile behaviour in apatite-bearing magmas

    Description: Magma reservoirs are thought to grow relatively slowly, assembling incrementally under volatile-saturated conditions. Eruptions may be triggered by injections of volatile-rich melt, or generation of over-pressure due to protracted crystallization. Here, we analyse fluorine, chlorine and water in apatite crystals trapped at di erent stages of magma evolution, and in melt inclusions from clinopyroxene and biotite crystals expelled during an explosive eruption of the Campi Flegrei caldera, Italy, about 4,000 years ago. We combine our geochemical analyses with thermodynamic modelling to reconstruct the evolution of magmatic volatile contents leading up to the explosive eruption. We find that the magma reservoir remained persistently water-undersaturated throughout most of its lifetime. Even crystals in contact with the melt shortly before eruption show that the magma was volatile-undersaturated. Our models suggest that the melt reached volatile saturation at low temperatures, just before eruption.We suggest that late-stage volatile saturation probably triggered the eruption, and conclude that ‘priming’ of the magma system for eruption may occur on timescales much shorter than the decadal to centennial timescales thought typical for magma reservoir assembly. Thus, surface deformation pulses that record magma assembly at depth beneath Campi Flegrei and other similar magmatic systems may not be immediately followed by an eruption; and explosive eruptions may begin with little warning.

    Description: The Campi Flegrei caldera (southern Italy) is one of the most hazardous volcanic systems on Earth, having produced 〉60 eruptions in the past 15 ka. The caldera remains active and its potential for future eruptions is high, posing a danger to the dense population living nearby. Despite this, our understanding of pre-eruptive processes and the architecture of the sub-volcanic system are poorly constrained. Here, we combine established petrological techniques, geothermobarometric evaluation, and high resolution trace element crystal mapping, to present a multifaceted, coherent reconstruction of the complex pre eruptive dynamics and eruption timescales of Astroni volcano located in the eastern sector of Campi Flegrei caldera. The Astroni volcano is an important case study for investigating plumbing system processes and dynamics at Campi Flegrei caldera because it produced the most recent (ca. 4 ka ago) Plinian eruption within the caldera (Astroni 6); current long-term forecasting studies postulate that a similar sized event in this location is a probable future scenario. Geothermobarometric results indicate interaction between an evolved, shallow magma chamber, and a less evolved, deeper pocket of magma, in agreement with previous studies focused on the Astroni 6 eruption products. In addition, a range of textural and trace element zoning patterns point to a complex evolution of both magmas prior to their subsequent interaction. High resolution trace element crystal maps reveal discrete zonations in compatible elements. These zonations, combined with knowledge of Kfeldspar growth rates, highlight a recharge event in the shallow plumbing system a few hours to days before the Astroni 6 eruption.

    Description: The East Scotia Ridge is an active back-arc spreading centre located to the west of the South Sandwich island arc in the Southern Ocean. Initial exploration of the ridge by deep-tow surveys provided the first evidence for hydrothermal activity in a back-arc setting outside of the western Pacific, and we returned in 2010 with a remotely operated vehicle to precisely locate and sample hydrothermal sites along ridge segments E2 and E9. Here we report the chemical and isotopic composition of high- and low-temperature vent fluids, and the mineralogy of associated high-temperature chimney material, for two sites at E2 (Dog’s Head and Sepia), and four sites at E9 (Black & White, Ivory Tower, Pagoda and Launch Pad). The chemistry of the fluids is highly variable between the ridge segments. Fluid temperatures were ∼350 °C at all vent sites except Black & White, which was significantly hotter (383 °C). End-member chloride concentrations in E2 fluids (532–536 mM) were close to background seawater (540 mM), whereas Cl in E9 fluids was much lower (98–220 mM) indicating that these fluids are affected by phase separation. Concentrations of the alkali elements (Na, Li, K and Cs) and the alkaline earth elements (Ca, Sr and Ba) co-vary with Cl, due to charge balance constraints. Similarly, concentrations of Mn and Zn are highest in the high Cl fluids but, by contrast, Fe/Cl ratios are higher in E9 fluids (3.8–8.1 × 10−3) than they are in E2 fluids (1.5–2.4 × 10−3) and fluids with lowest Cl have highest Cu. Although both ridge segments are magmatically inflated, there is no compelling evidence for input of magmatic gases to the vent fluids. Fluid δD values range from 0.2‰ to 1.5‰, pH values (3.02–3.42) are not especially low, and F concentrations (34.6–54.4 μM) are lower than bottom seawater (62.8 μM). The uppermost sections of conjugate chimney material from E2, and from Ivory Tower and Pagoda at E9, typically exhibit inner zones of massive chalcopyrite enclosed within an outer zone of disseminated sulphide, principally sphalerite and pyrite, in an anhydrite matrix. By contrast, the innermost part of the chimneys that currently vent fluids with lowest Cl (Black & White and Launch Pad), is dominated by anhydrite. By defining and assessing the controls on the chemical composition of these vent fluids, and associated mineralisation, this study provides new information for evaluating the significance of hydrothermal processes at back-arc basins for ocean chemistry and the formation of seafloor mineral deposits.
