    Description: We describe a new technique for transgenerational marking of embryonic otoliths that promises significant advancements in the study of larval dispersal and population connectivity in marine fishes. The approach is based on maternal transmission of 137Ba from spawning females to egg material that is ultimately incorporated into the otoliths of embryos produced by an individual after exposure to the isotope. We injected females of a benthic-spawning clownfish (Amphiprion melanopus) and a pelagic-spawning serranid (Centropristis striata) with enriched 137BaCl2 and then reared the resulting progeny through to settlement. Barium isotope ratios in the cores of larval otoliths were quantified using laser ablation inductively coupled plasma mass spectrometry. Larval otoliths from both species contained unequivocal Ba isotope signatures over a wide range of doses (0.8–23 μg 137Ba·g female–1). Female A. melanopus continued to produce marked larvae over multiple clutches and for at least 90 days after a single injection. The ability to administer different combinations of stable Ba isotopes provides a new means of mass-marking larvae of benthic- and pelagic-spawning fishes from multiple populations over extended spawning periods.

    Description: We assembled a comprehensive atlas of geochemical signatures in juvenile American shad (Alosa sapidissima) to discriminate natal river origins on a large spatial scale and at a high spatial resolution. Otoliths and (or) water samples were collected from 20 major spawning rivers from Florida to Quebec and were analyzed for elemental (Mg:Ca, Mn:Ca, Sr:Ca, and Ba:Ca) and isotope (87Sr:86Sr and δ18O) ratios. We examined correlations between water chemistry and otolith composition for five rivers where both were sampled. While Sr:Ca, Ba:Ca, 87Sr:86Sr, and δ18O values in otoliths reflected those ratios in ambient waters, Mg:Ca and Mn:Ca ratios in otoliths varied independently of water chemistry. Geochemical signatures were highly distinct among rivers, with an average classification accuracy of 93% using only those variables where otolith values were accurately predicted from water chemistry data. The study represents the largest assembled database of otolith signatures from the entire native range of a species, encompassing approximately 2700 km of coastline and 19 degrees of latitude and including all major extant spawning populations. This database will allow reliable estimates of natal origins of migrating ocean-phase American shad from the 2004 annual cohort in the future.

    Description: We used natural variation in the strontium concentration (Sr:Ca) and isotope composition (87Sr:86Sr) of stream waters and corresponding values recorded in otoliths of westslope cutthroat trout (Oncorhynchus clarkii lewisi) to examine movements during their life history in a large river network. We found significant spatial differences in Sr:Ca and 87Sr:86Sr values (strontium isoscapes) within and among numerous spawning and rearing streams that remained relatively constant seasonally. Both Sr:Ca and 87Sr:86Sr values in the otoliths of juveniles collected from nine natal streams were highly correlated with those values in the ambient water. Strontium isoscapes measured along the axis of otolith growth revealed that almost half of the juveniles had moved at least some distance from their natal streams. Finally, otolith Sr profiles from three spawning adults confirmed homing to natal streams and use of nonoverlapping habitats over their migratory lifetimes. Our study demonstrates that otolith geochemistry records movements of cutthroat trout through Sr isoscapes and therefore provides a method that complements and extends the utility of conventional tagging techniques in understanding life history strategies and conservation needs of freshwater fishes in river networks.
