    Notes: Abstract: A μ-opioid receptor protein (μ-ORP) purified to homogeneity from bovine striatal membranes has been functionally reconstituted in liposomes with highly purified heterotrimeric guanine nucleotide regulatory proteins (G proteins). A mixture of bovine brain G proteins, predominantly GoA, was used for most of the experiments, but some experiments were performed with individual pure G proteins, GoA, GoB, Gi1, and Gi2. Low Km GTPase was stimulated up to 150% by μ-opioid receptor agonists when both μ-ORP and a G protein (either the brain G protein mixture or a single heterotrimeric G protein) were present in the liposomes. Stimulation by a selective μ-agonist was concentration dependent and was reversed by the antagonist (−)-naloxone, but not by its inactive enantiomer, (+)-naloxone. The μ selectivity of μ-ORP was demonstrated by the inability of δ and κ agonists to stimulate GTPase in this system. High-affinity μ-agonist binding was also restored by reconstitution with the brain G protein mixture and with each of the four pure Gi and Go proteins studied. The binding of μ agonists is sensitive to inhibition by GTPγS and by sodium.

    Notes: Summary A 2-year prospective study was carried out in which 71 patients with primary cutaneous vasculitis were classified using the American College of Rheumatology (ACR) classification and the Chapel Hill Consensus Conference (CHCC) recommendations for Henoch Schonlein purpura (HSP). The sensitivity of the ACR criteria was 64.8% and that of the CHCC definition 31%. When the ACR criteria were combined with results of direct immunofluorescence (DIF) the sensitivity was 78.9%. The concordance between the two systems was low as only 12 patients fulfilled criteria for both classifications. Although the ACR criteria were found to be more useful in the classification of HSP our data suggest that they need to be modified to include adults with disease. The age at onset of disease was higher than that in the west. Seventy per cent of patients identified by either classification were 〉 20 years of age. The prevalence of gut involvement, microhaematuria and proteinuria was 〈 25% in both groups. The sensitivity of histopathology on the other hand was 80.4% and was not influenced by the duration of the lesion. The DIF test was a useful adjunct to histopathology if it was done within 48 h as the yield of a positive test was significantly higher in this group as compared to the patients who had the test done later.

    Notes: We have studied 56 unfractured chalk samples of the Upper Cretaceous Tor Formation of the Dan, South Arne and Gorm Fields, Danish North Sea. The samples have porosities of between 14% and 45% and calcite content of over 95%. The ultrasonic compressional- and shear-wave velocities (VP and VS) for dry and water-saturated samples were measured at up to 75 bar confining hydrostatic pressure corresponding to effective stress in the reservoir. The porosity is the main control of the ultrasonic velocities and therefore of the elastic moduli. The elastic moduli are slightly higher for samples from the South Arne Field than from the Dan Field for identical porosities. This difference may be due to textural differences between the chalk at the two locations because we observe that large grains (i.e. filled microfossils and fossil fragments) that occur more frequently in samples from the Dan Field have a porosity-reducing effect and that samples rich in large grains have a relatively low porosity for a given P-wave modulus. The clay content in the samples is low and is mainly represented by either kaolinite or smectite; samples with smectite have a lower P-wave modulus than samples with kaolinite at equal porosity. We find that ultrasonic VP and VS of dry chalk samples can be satisfactorily estimated with Gassmann's relationships from data for water-saturated samples. A pronounced difference between the VP/VS ratios for dry and water-saturated chalk samples indicates promising results for seismic amplitude-versus-offset analyses.

    Notes: A part of the mechanism of the gastrointestinal toxicity exhibited by non-steroidal anti-inflammatory drugs is believed to involve the uncoupling of mitochondrial oxidative phosphorylation. Most previous uncoupling studies have used rat liver mitochondria. There is little information on the effects of the drugs on mitochondria from other species.〈section xml:id="abs1-2"〉〈title type="main"〉Aim:To study the effect of indometacin on isolated liver mitochondria from rats, mice and humans.〈section xml:id="abs1-3"〉〈title type="main"〉Methods:We studied the effects of indometacin on respiration and adenosine triphosphate synthesis by isolated liver mitochondria from rats, mice and humans. Its effects were compared with those of dinitrophenol, a classical uncoupler.〈section xml:id="abs1-4"〉〈title type="main"〉Results:Indometacin uncoupled oxidative phosphorylation at low concentrations (P 〈 0.05) and inhibited respiration at high concentrations (P 〈 0.01) in all three species. Adenosine triphosphate synthesis was, however, more sensitive to dinitrophenol or indometacin at lower concentrations in mouse and human compared to rat liver mitochondria (P 〈 0.05).〈section xml:id="abs1-5"〉〈title type="main"〉Conclusions:The current study shows that indometacin acts as an inhibitory uncoupler in human mitochondria. It also demonstrates that the responses of rat, mouse and human mitochondria to indometacin are broadly similar.
