Synthesis of a Structural Gene for the Peptide Hormone Angiotensin II, Part 21). - Synthesis of Protected Mono-, Di-, and Trideoxyribonucleotides with 5'-Phosphate Group by Direct Phosphorylation.The synthesis of the following mononucleotides pTd(Ac)2), pan4Cd(Ac), pbz6Ad(Ac), pbz2Gd(Ac), dinucleotides pbz6Ad-Td(Ac), pan4Cd-ib2Gd(iB), pbz6Ad-bz6Ad(Ac), pTd-Td(Ac), pTd-ib2Gd(Ac), pTd-an4Cd(Ac), pib2Gd-ib2Gd(iB), and trinucleotides pTd-Td-bz6Ad(Ac), pib2Gd-ib2Gd-ib2Gd(iB), pbz6Ad-bz6Ad-bz6Ad(Ac), and pTd-Td-Td(Ac) by the method of direct phosphorylation with phosphorus oxychloride is described in detail. This new and economical method makes phosphate-protecting groups unnecessary, pan4cd-an4Cd(Ac) and pan4Cd-an4Cd-an4Cd(Ac) have been synthesized using 3-hydroxypropionitrile; pan4Cd-ib2Gd(Ac) and pib2Gd-ib2Gd-ib2Gd(Ac) also by phosphorylation with 2-cyanoethyl phosphate; and pbz6Ad-Td(Ac) also with 2,6-dimethyl-4-tritylaniline (11) as lipophilic protecting group.

Die Synthese der Mononucleotide pTd(Ac)2), pan4Cd(Ac), pbz6Ad(Ac), pbz2Gd(Ac), der Dinucleotide pbz6Ad-Td(Ac), pan4Cd-ib2Gd(Ac), pTd-Td(Ac), pTd-ib2Gd(Ac), pTd-an4Cd(Ac), pib2Gd-ib2Gd(iB), pbz6Ad-bz6Ad(Ac) und der Trinucleotide pTd-Td-bz6Ad(Ac), pib2Gd-ib2Gd-ib2Gd(iB), pTd-Td-Td(Ac) und pbz6Ad-bz6Ad-bz6Ad(Ac) nach der Methode der direkten Phosphorylierung mit Phosphorylchlorid wird beschrieben. Diese neue, rationelle Methode macht Phosphatschutz-gruppen unnötig. pan4Cd-an4Cd(Ac) und pan4Cd-an4Cd-an4Cd(Ac) wurden unter Verwendung von 3-Hydroxypropionitril, pan4Cd-ib2Gd(Ac), pib2Gd-ib2Gd-ib2Gd(Ac) auch durch Phosphorylierung mit 2-Cyanethylphosphat, pbz6Ad-Td(Ac) wurde auch mit 2,6-Dimethyl-4-tritylanilin (11) als lipophiler Schutzgruppe dargestellt.
