Abstract:
Mikrotubuli werden durch so genannte Mikrotubuli‐assoziierte Proteine reguliert. Wenig ist jedoch über die Struktur dieser Proteine im Komplex mit Mikrotubuli bekannt. Hier zeigen wir, dass das Mikrotubuli‐assoziierte Protein Tau, das in Lösung intrinsisch ungeordnet ist, bei Bindung an Mikrotubuli lokal in eine stabile Struktur faltet. Während Tau flexibel in Lösung ist und eine β‐Faltblattstruktur in Amyloidfibrillen annimmt, falten die konservierten Hexapeptide in den Wiederholungssequenzen 2 und 3 in eine Haarnadelschleife im Komplex mit Mikrotubuli. Dies zeigt, dass die Bindung an Mikrotubuli eine spezifische Konformation im Tau‐Protein stabilisiert.
