    Description: Catechol 2,3-dioxygenase (C23O) from a new phenolic compound degrader Thauera sp. K11 was purified and characterized. The native form of the enzyme was determined as a homotetramer with a molecular weight of 140 kDa, and its isoelectric point was close to 6.4. One iron per enzyme subunit was detected using atom absorption spectroscopy, and the effective size of C23O in its dilute solution (0.2 g L −1 , pH 8.0) was 14.5 nm. The optimal pH and temperature were 8.4 and 45 °C, respectively. The addition of Mg 2+ , Cu 2+ , Fe 2+ , and Mn 2+ could improve the enzyme activity, while Ag + was found to be a strong inhibitor. C23O was stable in alkali conditions (pH 7.6–11.0) and thermostable below 50 °C. The final purified C23O had a sheet content of 53%, consistent with the theoretical value. This showed that the purified catechol 2,3-dioxygenase folded with a reasonable secondary structure.

    Description: Tricholoma matsutake (S. Ito et Imai) is an ectomycorrhizal basidiomycete associated with Pinaceae and Fagaceae trees in the Northern Hemisphere. It is still unknown whether the symbiotic relationship with this ectomycorrhiza could affect the host plant's endophytic microbial community. In this study, we used high throughput sequencing to analyze the endophytic microbial communities of different Quercus aquifolioides tissues with or without T. matsutake partner. About 35,000 clean reads were obtained per sample, representing 34 bacterial phyla and 7 fungal phyla. We observed 3980 operational taxonomic units (OTUs) of bacteria and 457 OTUs of fungi at a 97% similarity level. Three bacterial phyla, Proteobacteria, Cyanobacteria, and Bacteroidetes, and the fungal phylum Ascomycota were dominant in all tissues. The relative abundance of these taxa differed significantly between Q. aquifolioides tissues with and without T. matsutake partner ( p  〈 0.05). The bacterial genus Pseudomonas and the fungal genus Cryptosporiopsis were more abundant in mycorrhized roots than in control roots. This study showed that the community structure and dominant species of endophytic microbial communities in Q. aquifolioides tissues might be altered by colonization with T. matsutake . This work provides a new insight into the interactions between ectomycorrhizal fungus and host plant.

    Description: Mixed bacterial cultures are increasingly being used in the production of polyhydroxyalkanoates (PHAs), as they have the potential to be more cost effective than axenic pure cultures. The purpose of this study was to use pure cultures in combination to identify their potential of PHA production. In this work we used volatile fatty acids (VFAs) and glucose as carbon source to check the ability of selected strains ST2 ( Pseudomonas sp.) and CS8 ( Bacillus sp.) as co-culture. The production of PHA in pure co-cultures of bacteria was therefore investigated in order to understand the effect of combining cultures on PHA production parameters and material properties. Bacteria could use the feed in better way when mixed as compared to individual strain. In undertaking this analysis, model volatile fatty acids (i.e., acetic and propionic acids) were used alone and in combination with glucose as feedstock. The production by Pseudomonas was 34% while 24% by Bacillus . However when combined and mixed feed (glucose + propionic acid) was used, 35% PHA produced. Overall, it was found that the ability of the pure cultures to produce PHA was low but when selected cultures were mixed, their ability to produce PHA was enhanced. Copolymers were obtained instead of homopolymers with improved properties. This suggests that industrial wastewater rich in volatile fatty acids and carbohydrates can be a good carbon source for PHA production with variable properties.

    Description: Overlap-extension PCR is a method for splice of gene segments to produce focused fragments for constructing recombinant plasmid, but its complexity limits its application. To simplify the protocol and to improve the effectiveness, we employed gradient temperatures to replace the single annealing temperature in the thermo-cycling program, and optimize the templates ratio. The concentration of each fragment was adjusted to 10 ng µl −1 . Fragment concentration ratio was the inverse of the fragment size ratio. The products of fused segments were 2000–5000 bp in length using the revised one-step method. This method splices effective two or more fragments to fused gene and produce recombinant plasmid.

    Description: Melanization of carbon stressed Aspergillus nidulans cultures were studied. Melanin production showed strong positive correlation with the activity of the secreted chitinase and ß-1,3-glucanase. Deletion of either chiB encoding an autolytic endochitinase or engA encoding an autolytic ß-1,3-endoglucanase, or both, almost completely prevented melanization of carbon stressed cultures. In contrast, addition of Trichoderma lyticase to cultures induced melanin production. Synthetic melanin could efficiently inhibit the purified ChiB chitinase activity. It could also efficiently decrease the intensity of hyphal fragmentation and pellet disorganization in Trichoderma lyticase treated cultures. Glyphosate, an inhibitor of L-3,4-dihydroxyphenylalanine-type melanin synthesis, could prevent melanization of carbon-starved cultures and enhanced pellet disorganization, while pyroquilon, a 1,8-dihydroxynaphthalene-type melanin synthesis inhibitor, enhanced melanization, and prevented pellet disorganization. We concluded that cell wall stress induced by autolytic cell wall hydrolases was responsible for melanization of carbon-starved cultures. The produced melanin can shield the living cells but may not inhibit the degradation and reutilization of cell wall materials of dead hyphae. Controlling the activity of autolytic hydrolase production can be an efficient approach to prevent unwanted melanization in the fermentation industry, while applying melanin synthesis inhibitors can decrease the resistance of pathogenic fungi against the chitinases produced by the host organism.

    Description: A new minireplicon ( rep26 minireplicon) from pBMB26, the 188 kb indigenous plasmid related to spore-crystal association (SCA) phenotype in Bacillus thuringiensis strain YBT-020, was characterized. A 12 kb Eco RI fragment, which encoded 10 putative open reading frames (ORFs), was capable of supporting replication when cloned in a replication probe vector. Deletion and frame shift mutation analysis showed that a 4.1 kb region encompassing two putative ORFs ( orf21 and orf22 ) was essential for the plasmid replication in B. thuringiensis . Gene orf21 encoding a 49.8 kDa protein (named Rep26) with a helix-turn-helix motif showed no homology with known replication proteins and gene orf22 encoding a protein of 82.6 kDa showed homology to bacterial PcrA helicase. The replication origin of rep26 minireplicon was proved to be located in the coding region of orf21 . Plasmid stability experiments indicated that the recombinant plasmid containing rep26 minireplicon has excellent segregational stability. BLASTP analysis revealed that amino acid sequences of ORF21 and ORF22 were well conserved among Bacillus cereus group strains. The rep26 minireplicon was widely distributed and could be defined as a new typical replicon in the megaplasmids of B. cereus group.

    Description: The growth of an organism is highly dependent on the acquisition of carbon and metals, and availability of these nutrients in the environment affects its survival. Organisms can obtain both nutrients simultaneously through proteins of the CitMHS superfamily. Bioinformatic studies suggested a CitMHS gene (Accession number ABS03965.1) in Kineococcus radiotolerans . Radio flux assays following 14-C radiolabelled citrate, either free or complexed to a variety of metal ions, in K. radiotolerans demonstrated internalization of the citrate when bound to select metal ions only, primarily in the form of calcium-citrate. A pH response was also observed, consistent with a permease (ATP independent) mechanism as noted for other CitMHS family members, with greater uptake at pH 7 compared to pH 10. These results confirm the ability of K. radiotolerans to transport complexed citrate.

    Description: Beauveria bassiana is a broad spectrum microbial bioagent used for the control of agriculturally important insect pests. However, in our experiments, two virulent isolates of B. bassiana (B2 and B10) showed specific preference toward Maruca vitrata and Helicoverpa armigera of pigeonpea. To better understand this feature, we developed a qPCR assay to quantify the chitinase (virulence factor) of B. bassiana during the infection process. Isolates of B. bassiana were grown on insect cuticle amended medium and minimal medium (without insect cuticle) to assess the induction of chitinase. Our results revealed a positive correlation between expression of chitinase by B. bassiana and the substrates (with or without cuticles of M. vitrata and H. armigera ) used. This study showcases the methodology to quantify the chitinase and analysis of variation in virulence of B. bassiana (B2 and B10) against M. vitrata and H. armigera .
