    Abstract: The aims of this study were to identify endophytic and epiphytic plant growth promoting bacteria (PGPB) from healthy almond trees and to characterize plant growth promoting mechanisms. By using MALDI-TOF analyses, 19 Gram-negative isolates, belonging to Serratia, Pseudomonas, Ochrobactrum and Enterobacter genus and 26 Gram-positive isolates, belonging to Bacillus and Arthrobacter genus were identified. Among the tested bacterial isolates, 46 isolates were positive for siderophore production, 45 isolates for ammonium production, 35 isolates solubilized phosphorus varying ratios, and all isolates produced IAA between 5.48-127.81 µg/ml concentrations. Isolates of Pseudomonas spp and Enterobacter cloacae were found highly effective for siderophore production, S. marcescens for solubilisation of phosphorus, E. cloacae for IAA production, Bacillus spp for ammonia production. Bacterial isolates tested showed significant differences in siderophore, ammonia and IAA production and phosphorus solubilisation efficiency. It was found that the efficacies of endophytic bacterial isolates were lower compared to the same species of epiphytic bacterial isolates. Overall results suggest that use of the most efficient PGPB isolates have an excellent potential to be used as biofertilizer for cultivation of economically important crops.

    Abstract: Bu çalışmanın amacı, farklı bitki türlerinden elde edilen uçucu yağların kimyasal bileşimini ve gal (ur) oluşturan bitki patojenik bakteriyel hastalık etmeni Rhizobium radiobacter, Pseudomonas savastanoi pv. savastanoi ve P.savastanoi pv. nerii’ye karşı antibakteriyel etkinlerini belirlemektir. Thymbra spicata var. spicata, Thymus serpyllum, Thymus sipyleus, Origanum syriacum, Origanum majorana, Ocimum basilicum, Mentha spicata, Melissa officinalis, Lavandula stoechas var. stoechas, Rosmarinus officinalis, Salvia officinalis, Laurus nobilis ve Foeniculum vulgare uçucu yağlarının kimyasal bileşenleri GC/MS ile tanımlanmıştır. Carvacrol Thymbra spicata var. spicata (% 66.88) ve Origanum syriacum (% 79.8), thymol Thymus serpyllum (% 41.03), geranial Thymus sipyleus (% 13.72) ve Melissa officinalis (30.4), 4-terpineol Origanum majorana (31.67 %), linalool Ocimum basilicum (% 30.23), carvone Mentha spicata (% 55.58), 1,8 cineole Lavandula stoechas var. stoechas (% 35.5), Laurus nobilis (% 35.5) ve Rosmarinus officinalis (% 18.47), camphor Salvia officinalis (% 24.59) ve trans-anethole ise Foeniculum vulgare (% 82.8) uçucu yağlarının ana bileşenleri olarak belirlenmiştir. Engelleme zon çapı değerlerine dayanarak, uçucu yağlarıP.savastanoi pv. savastanoi (7.0-44.67 mm), ardından R.radiobacter (9.6-37.67 mm) ve P.savastanoi pv. nerii (6.33-18.33 mm) ye karşı oldukça güçlü antibakteriyel etkinlik göstermiştir. Lamiaceae familyasına ait bitkilerin uçucu yağlarının genellikle Lauraceae ve Apiaceae familyalarına göre daha etkili olduğu bulunmuştur. O.syriacum, T.serpyllum ve T.spicata var. spicata, test edilen tüm bakteri türlerine karşı en yüksek antibakteriyel aktiviteleri gösteren umut verici uçucu yağlar olarak belirlenmiştir. Bu çalışmanın bulguları uçucu yağların gal oluşturan bakteriyel hastalık etmenlerine karşı antibakteriyel ajan olarak kullanılma potansiyeline sahip olduğunu göstermiştir.

    Abstract: Amerika'nın yerli bir bitkisi olan Solanum elaeagnifolium (SOLEL), biyolojik çeşitlilik ve tarım üzerinde etkileri olan, dünya çapında oldukça yaygın istilacı bir yabancı türdür. Akdeniz havzasında ve Türkiye'nin bulunduğu Orta Doğu'da uzun süredir bulunmasına rağmen, 2006 yılındaki Türkiye'nin Doğu Akdeniz'inden göçmen kuşlar aracığıyla girişine atfedilen bir kayıt dışında bilgi yoktur. Çevre ülkelerde yaygın ve yoğun olarak bulunmasına rağmen Türkiye'de SOLEL'in tespit edilememesi şüpheyle karşılanmaktadır. Çalışmanın amacı Türkiye'deki SOLEL popülasyonlarını kontrol etmek ve Türkiye'deki düşük yoğunluğunun sebebini anlamaktır. Türkiye'de göçmen kuş yollarında bulunan ve ülke sınırlarına yakın iki alan seçilmiştir. Bu alanlarda 2020-2023 yılları arasında SOLEL popülasyonlarını saptamak için çok sayıda kontroller yapılmıştır. 2020-2023'te Türkiye'nin birbirinden 1000 km'den fazla uzak olan Doğu Akdeniz ve Ege Bölgelerinde yeni popülasyonlar tespit edilmiştir. SOLEL popülasyonları, Türkiye ve diğer ülkelerdeki kuş göç yolları, istilacı bir yabancı bitkinin girişinde ve yayılmasında kuşların rolü karşılaştırılmıştır. SOLEL'in giriş yolu; Türkiye'deki ilk kaydında ve literatürde diğer bitkiler için gösterilen kuş göçü yollarındadır.

    Abstract: Carrot [Daucus carota L.] is one of the most consumed healthy vegetables worldwide due to its vitamins, amino acids and high nutritional content. Fungal and bacterial disease agents, encountered in the fields where carrots are grown in the Amik Plain of Hatay province, were determined during 2018-2019 growing season. Disease surveys were carried out at three different phenological stages of the plant. Different fungal and bacterial disease agents were isolated and subsequently identified from diseased leaves, stems and fruits. In addition to the disease symptoms in the form of transverse brown dry depressions on carrot fruits, from the brown-black spots on the leaves and stems, Alternaria dauci, A. alternata, Rhizoctonia solani, R. carotae, Botrytis cinerea, Sclerotinia sclerotiorum, Geotrichum candidum, Pythium spp., Fusarium bra. equiseti, F. proliferatum and F. solani species were obtained and identified as a result of molecular and MALDI-TOF protein profile analyzes as well as their morphological features. Powdery mildew caused by Erysiphe heraclei was also determined in the surveyed area. By the bacterial isolates obtained from carrot showing soft rot symptoms, major soft rot disease agents, such as Pectobacterium carotovorum subsp. carotovorum and Pseudomonas corrugata and opportunistic soft rot disease agents, such as P. marginalis, P. fluorescens, P. mediterranea, P. veronii, Erwinia rhapontici, E. persicina, Enterobacter cloacae, Lelliottia amnigena, Rahnella aquatilis, Pantoea agglomerans, were identified by using MALDI-TOF analyses. To the best of our knowledge, this is the first report in which fungal disease agent Fusarium brachygibbosum and all soft rotting bacterial disease agents were determined at species level on carrot roots growing in Turkey.

    Abstract: Fungal disease agents that cause internal fruit rot in agricultural products before and after harvest cause serious quality and yield losses. At the same time, the mycotoxins they produce in foods pose a health risk for consumers. In this study, it was aimed to isolate fungal disease agents that cause internal rot symptoms in dried pepper fruits grown in Hatay province and sold in local bazaars, to identify by morphological and MALDI-TOF methods, and to determine the antifungal effects of different plant essential oils against the most common disease agents. Alternaria alternata and Aspergillus niger were determined as the most common fungal disease agents following the result of isolations, morphological and MALDI-TOF diagnostic studies from fruit samples showing signs of disease. Antifungal effects of different vapor doses of essential oils of different thyme (Thymus vulgaris L., Tymbra spicata L. and Origanum syriacum L.), fennel (Foeniculum vulgare Mill.), laurel (Laurus nobilis L.) and eucalyptus (Eucalyptus camaldulensis Dehnh) were investigated against fungal disease agents in vitro conditions. The highest antifungal activities (100% inhibition) against fungal isolates were displayed by essential oils of Thymbra spicata (2.0-4.0 µl Petri-1), Origanum syriacum (4.0 µl Petri-1) and Thymus vulgaris (4.0-6.0 µl Petri-1). Eucalyptus camaldulensis (16.0-40.0 Petri-1) exhibited the lowest antifungal activity against tested fungal isolates. In addition, the antifungal properties and EC50 values of essential oils were also determined. Plant essential oils showed antifungal effects against the tested fungal isolates in a dose-dependent manner. The results showed that plant essential oils might be applied as biofumigants in dried agricultural products.

    Abstract: Genellikle lahanagiller, Cruciferae veya hardal ailesi olarak bilinen Brassicaceae familyası, dünya çapında ekonomik açıdan tarımsal öneme sahip yağlı tohum, sebze ve çeşni amaçlı kullanılan bitki türleri içerir. Hatay ili, Türkiye’nin Brassicaceae familyasına dahil minör sebze türlerinin yetiştiriciliğinin yapıldığı önemli iller arasında yer alır. Bu çalışmada 2020-2021 yetiştirme sezonlarında Hatay ilinin sebze yetiştiriciliğinin yapıldığı ilçelerde Brassica cinsine dahil minör sebzelerden lahana (kırmızı ve beyaz baş), brokoli, karnabahar, tere, roka ve turp bitkilerinde sorun olan fungal, oomycet ve bakteriyel hastalık etmenlerinin tanılanması ve yaygınlık durumlarının belirlenmesini amaçlanmıştır. Sürvey yapılan alanlarda hastalık belirtisi gösteren şüpheli lahana, brokoli, karnabahar ve turp bitkilerin kök, gövde ve yapraklarından yapılan izolasyonlarda Rhizoctonia solani, Sclerotinia sclerotiorum, Fusarium solani, Fusarium oxysporum, Fusarium equiseti, Alternaria alternata, Botrytis cinerea ve Stemphylum sp., ait fungal hastalık etmenlerin yanısıra yanısıra Albugo lepidii, Albugo candida ve Perofascia lepidii gibi obligat parazit oomycet patojenler tere, roka ve turp bitkilerinin yaprak ve gövdelerinde değişen yaygınlık ve rastlama sıklıklarında tespit edilmiştir. Fungal etmenlerin yanısıra yapılan morfolojik, biyokimyasal, patojenisite testler ve MALDI-TOF analizleri sonucunda Xanthomonas campestris pv. campestris, Pectobacterium carotovorum subsp. carotovorum, Pectobacterium parmentieri, Pseudomonas corrugata ve Pseudomonas cichorii primer bakteriyel hastalık etmenleri olarak belirlenmiştir. Pantoea agglomerans, Pseudomonas marginalis, Pseudomonas fluorescens, Enterobacter cloacae ve Bacillus pumilus ise fırsatçı sekonder bakteriyel yumuşak çürüklük etmenler olarak tanılanmışlardır. Bilindiği kadarıyla F. solani, F. oxysporum, F. equiseti, A. alternata, B. cinerea, Stemphylum sp., P. parmentieri, P. corrugata ve P. cichorii primer, P. agglomerans, P. marginalis, P. fluorescens, E. cloacae ve B. pumilus ise sekonder fırsatçı patojenler olarak Türkiye’de yetiştirilen farklı Brassica spp ait lahana (kırmızı ve beyaz baş), brokoli, karnabahar, tere, roka ve turp gibi minör sebzelerde fungal ve bakteriyel hastalıklara neden olduğu ilk kez bu çalışma ile tespit edilmiştir.

    Abstract: During the 2018-2019 vegetation period, survey was conducted to identify soft rot disease agent in the white head cabbage production areas of Yozgat. Crystal Violet Pectate (CVP) medium was used for the isolation of bacterial agent from tissues showing watery-wet lesions and rotting signs on leaves, stems and roots. A total of 16 pectolytic isolates that formed cavity in the CVP medium were obtained from 24 suspicious plant samples collected from different fields. The colonies of bacterial isolates detected as transparent, convex and crater with edges were gram negative, facultative anaerobe, oxidase negative, catalase positive, and did not show fluorescence on the King B-medium under UV-light. It has been determined that isolates caused soft rot symptoms in potato slices and cabbage plants could grow at 37 ºC, had turbidity in liquid medium containing 5% NaCl and cause hypersensitivity reaction in tobacco leaves. In PCR analyses with obtained 16 isolates revealed that all of the isolates generated 434 bp and 550 bp PCR product using Y1/Y2 and EXPCCF/EXCPCCR primers which were recommended for the identification of Pectobacterium spp, and P. carotovorum subsp. carotovorum, respectively. Based on biochemical, physiological, pathogenicity and molecular analysis results, 16 isolates were identified as P. carotovorum subsp. carotovorum. It has been also observed that all pathogenic isolates caused signs of soft rot in the form of watery-wet lesions in artificially inoculated cabbage plants. It was determined for the first time with this study that P. carotovorum subsp. carotovorum is the source of soft rot disease infection in the white head cabbage production areas of Yozgat province.

    Abstract: In this study, in vitro antagonistic potentials of bacterial microbiomes, obtained from earthworm fertiliser, vermicompost, were investigated on inhibitions of mycelial growth of major foliar and soilborne fungal disease agents Sclerotinia sclerotiorum, Macrophomina phaseolina, Botrytis cinerea, Verticilium dahliae. Total of 69 putative bacterial biocontrol agent (BCA) isolates were obtained from commercial vermicompost lots. Among them, 28 bacterial isolates (49.12% of total isolates) were inhibited mycelial growth of S. sclerotiorum by 1.72-75.43%, M. phaseolina by 1.67-65.83%, B. cinerea by 3.44-57.18%, V dahliae by 2.28-58.74%, respectively. Majorities of bacterial isolates were identified as Bacillus spp. Certain isolates of Bacillus spp. have caused noticeable morphological changes on mycelia of S. sclerotiorum. Antagonistic potentials of bacterial isolates were found to increase by pre-incubation time prior the fungal inoculation. Due to high antagonistic properties, efficient isolates of Bacillus spp.  may be used as biocontrol agent against soilborne diseases as an alternative to pesticides to promote organic and sustainable agriculture.

    Abstract: Potato is the one of the most consumed vegetable in Turkey. During field surveys in June of 2019, new disease symptoms on potato tubers, displaying a water-soaked soft rubbery rot with a sour, fermented smell, and white mycelial growth, were observed in commercial fields in Çumra district of Konya, Turkey. Diseased tubers were collected and small tissue pieces from surface disinfected symptomatic tuber were placed on potato dextrose agar (PDA) and incubated at 25°C. Fast growing, flat, white to cream, dry and finely suede-like with no reverse pigmented fungal colonies were subsequently developed from infected tissues. Hyphae were hyaline, dichotomously branched and septate, producing aerial chains of hyaline, smooth, one-celled, cylindrical arthroconidia. Cultural and morphological characters were consistent with Geotrichum candidum. All artificially inoculated tubers displayed the same symptoms as seen for naturally infected tubers and the fungus having same morphological characters was re-isolated from inoculated tubers. Morphological identification of disease agent was confirmed molecularly based on protein profiling by using MALDI-TOF MS and also by sequencing amplified Internal Transcribed Spacer (ITS) region ((ITS1-5.8S rDNA-ITS4). The sequence was 99% identical and a representative sequence of isolate was submitted to GenBank. The presence of potato rubbery rot disease caused by G. candidum has not been previously reported in Turkey. This is the first report of morphological and molecular characterization of G. candidum causal disease agent of rubber rot on potato tubers grown in Turkey.

    Abstract: Fungal disease agent Pestalotiopsis funerea causes conifer seedlings shoot or tip blight disease. In this study, chemical compositions and antifungal activities of different blended essential oils obtained from fennel (Foeniculum vulgare Mill.) and laurel (Laurus nobilis L.) were investigated against P. funerea in vitro conditions. Chemical compositions of essential oils were determined using GC-MS analysis. Antifungal volatile phase effects of different concentrations of essential oils mixtures were determined on inhibition of mycelial growth and conidial germination in vitro conditions. The effect of most effective concentrations of essential oil mixtures on the morphology of fungal hypha and conidia were also investigated using light microscope. GC-MS analysis of essential oils revealed that eucalyptol (52.88%) and trans-anethole (81.55%) were the major components of laurel and fennel essential oils. Individual use of fennel and laurel essential oils were found to completely inhibit mycelial growth at 20.0 and 25.0 µl petri-1 concentrations, respectively. Dual blending fennel/laurel essential oil mixtures showed synergistic antifungal activity on inhibition of mycelial growth and conidial germination. By blending fennel and laurel essential oils at proportion of 3:1, antifungal activities were significantly increased that complete mycelial growth and conidial inhibition were recorded at relatively lower concentration (10 µl petri-1). Microscopic observations on hyphae, exposed to volatile phase of the most efficient concentrations of essential oil mixtures, revealed considerable structural deformations such as cytoplasmic coagulation, vacuolization and protoplast leakage from fungal hyphae and conidia. The results showed that blending of fennel and laurel essential oils, which have synergistic antifungal effects, might be used as an effective bio fungicide in management of fungal plant disease agents.
