6-Formylindolo(3,2-b)Carbazole (FICZ) Modulates the Signalsome Responsible for RA-Induced Differentiation of HL-60 Myeloblastic Leukemia Cells

FICZ induces a c-Cbl-AhR association (mean fluorescence).

HL-60 cells were initiated in culture at 0.1 x 106 cells/ml with 1 μM RA, 100 nM FICZ, 1 μM α-NF and 1 μM β-NF as indicated. Cells were harvested, fixed for 10 min with 2% paraformaldehyde, and permeabilized with ice cold methanol. Cells were labeled with the primary antibodies (or isotype controls) and then stained with Alexa 488- and 594-conjugated goat anti-mouse and goat anti-rabbit secondary antibodies, respectively (Invitrogen). The immunocomplexes were analyzed using flow cytometry (BD FACS ARIA III SORP, BD Biosciences). Mean fluorescence intensity is shown.
