PD-L1 upregulation by IFN-α/γ-mediated Stat1 suppresses anti-HBV T cell response

IFN-α/γ upregulate PD-L1 expression in a Stat1 dependent manner.

(A and C) L02 cells were transfected with PD-L1 promoter luciferase reporter plasmid with a wild type or mutated Stat1 binding site and incubated with 80 U/ml IFN-γ (A) or 50 U/ml IFN-α (C) for 48 h. (B and D) L02 cells transfected with PD-L1 promoter luciferase reporter plasmid were co-treated with IFN-γ (B) or IFN-α (D) and Stat1 siRNA or fludarabine (5 μg/ml) for 48 h. The relative luciferase activity was determined using dual luciferase assay. Data are presented as mean ± SD for three independent experiments. ** p<0.01, and *** p<0.001 compared to the control.
