A Novel Replication-Competent Vaccinia Vector MVTT Is Superior to MVA for Inducing High Levels of Neutralizing Antibody via Mucosal Vaccination

Groups of 3 mice were injected with MVTT-S (solid symbol) or MVA-S (empty symbol) twice at a dose of 104 (square), 105 (cycle), 106 (triangle) PFU at a three-week interval. Sera were collected two weeks after each vaccination. A pseudotype-based neutralization assay was performed to determine the level of Nab response. The Y axis stands for the percentage of inhibition whereas the X axis represents the serum dilution factor. The dash line indicates the level of 50% of viral inhibition. The neutralization experiment was repeated twice with similar results obtained. Moreover, two independent immunization experiments were conducted with consistent results obtained.
