    Notes: The proliferation of human monocytic Mono Mac 6 cells was significantly retarded by treatment with lovastatin (LOV, 10 μM) for 72 h. Treatment of Mono Mac 6 cells with LOV increased surface protein expression of monocyte-associated CD14 and the integrin-chain CD11b towards levels found in isolated human blood monocytes. These effects were dose-dependent and completely reversed by the isoprenoid precursor mevalonate (MVA). LOV failed to induce growth retardation and upregulation of CD11b or CD14 in the less mature premonocytic U937 cell line. While CD11b expression was comparable in Mono Mac 6 cells treated with LOV (10 μM), TNF (100 U ml-1) or LPS (10 ng ml-1), upregulation of CD14 by LOV was less pronounced. Basal CD23 expression was unaffected by LOV but markedly reduced by treatment with TNF or LPS. Moreover, LOV enhanced Mono Mac 6 adhesiveness to human umbilical vein endothelial cells to levels found in isolated human blood monocytes, probably due to the increased CD11b and CD14 expression. In conclusion, LOV can induce differentiation of monocytic cells which is reflected by the retardation of growth, expression of CD14 and CD11b, and enhanced adhesiveness.

    Notes: A lysine sensor for process control of lysine fermentation was developed based on a Clark-type electrode in combination with L-lysine-α-oxidase. The enzyme, isolated from Trichoderma viride, was immobilized between a cellulose and a polypropylene foil using a polyurethane resin. Lysine determinations were carried out in a flow-through system as anodic measurements when H2O2 was measured and as chathodic measurements when the consumption of O2 was followed. The sensitivity of the sensor toward other amino acids was determined.

    Notes: CD36, a multifunctional adhesion receptor e.g. for thrombospondin and collagen, as well as a scavenger receptor for oxidized low density lipoprotein, is expressed e.g. on platelets and monocytes. By this dual role it might be involved in early steps of atherosclerosis like the recruitment of monocytes and formation of foam cells. We therefore studied the effects of n-3 fatty acids on CD36 expression in human monocytic cells. Incorporation of eicosapentaenoic acid (EPA, C20:5n-3) and docosahexaenoic acid (DHA, C22:6n-3) into cellular phospholipids resulted in a significant reduction of CD36 expression at the mRNA and protein level, whereas arachidonic acid (AA, C20: 4n-6) and linoleic acid (LA, C18:2n-6) tended to increase CD36 expression compared to the control. This specific down-regulation of CD36 by n-3 fatty acids in cells involved in the initiation and progression of atherogenesis and inflammation, represents a further mechanism that may contribute to the beneficial effects of n-3 polyunsaturated fatty acids (PUFA) in these disorders.

    Notes: This study describes experiments on sedimentestuarine water slurries originating from a Spartina alterniflora salt marsh. We investigated the fate of tin(IV) or monomethyltin cation (MeSn3+) chlorides after their additon to slurries under anaerobic and aerobic conditions. We did not observe methylation of tin in anaerobic or aerobic slurries with and without added tin(IV). MeSn3+-amended samples occasionally formed small amounts of Me2Sn2+ or Me3Sn+ after extended periods of time, particularly when MeSn3+ remained in solution. The stability of MeSn3+ in slurries demonstrates that the absence of net methylation of tin(IV) is not due to rapid demethylation of MeSn3+ or its further methylation. Inorganic tin concentrations in the aqueous phase of anaerobic slurries spiked with MeSn3+ and unspiked slurries decreased by about 85% in 21 days and remained relatively constant until the end of the 59-day experiments. In similar anaerobic experiments about 25% of the MeSn3+ spike was adsorbed to sediment within 1 h and about 75% was adsorbed within 10 days. The lack of methylation and demethylation reactions in our aerobic and anaerobic slurries, which contrasts with two previous reports, undoubtedly reflects the absence of added nutrients and low concentrations of added tin(IV) in our experiments. We believe that our model experiments more accurately reflect conditions in salt marshes than do previous studies. We conclude that future model studies on methylation of inorganic tin should include. S. alterniflora because it is so prominent in observations of methyltin compounds in the estuary.

    Notes: The conformation of the staphylococcal nuclease-bound metal-dTdA complex, previously determined by NMR methods [Weber, D.J., Mullen, G.P., Mildvan, A.S. (1991) Biochemistry 30:7425-7437] was docked into the X-ray structure of the enzyme-Ca2+-3′,5′-pdTp complex [Loll, P.J., Lattman, E.E. (1989) Proteins: Struct., Funct., Genet. 5:183-201] by superimposing the metal ions, taking into account intermolecular nuclear Overhauser effects from assigned aromatic proton resonances of Tyr-85, Tyr-113, and Tyr-115 to proton resonances of the leaving dA moiety of dTdA, and energy minimization to relieve small overlaps. The proton resonances of the Phe, Tyr, and Trp residues of the enzyme in the ternary enzyme-La3+-dTdA complex were sequence specifically assigned by 2D phase-sensitive NOESY, with and without deuteration of the aromatic protons of the Tyr residues, and by 2D heteronu-clear multiple quantum correlation (HMQC) spectroscopy and 3D NOESY-HMQC spectros-copy with 15N labeling. While resonances of most Phe, Tyr and Trp residues were unshifted by the substrate dTdA from those found in the enzyme-La3+-3′,5′-pdTp complex and the enzyme-Ca2+-3′,5′-pdTp complex, proton resonances of Tyr-85, Tyr-113, Tyr-115, and Phe-34 were shifted by 0.08 to 0.33 ppm and the 15N resonance of Tyr-113 was shifted by 2.1 ppm by the presence of substrate. The optimized position of enzyme-bound dTdA shows the 5′-dA leaving group to partially overlap the inhibitor, 3′,5′-pdTp (in the X-ray structure). Tne 3′-TMP moiety of dTdA points toward the solvent in a channel defined by Ile-18, Asp-19, Thr-22, Lys-45, and His-46. The phosphate of dTdA is coordinated by the metal, and an adjacent inner sphere water ligand is positioned to donate a hydrogen bond to the general base Glu-43 and to attack the phosphorus with inversion. Arg-35 and Arg-87 donate monodentate hydrogen bonds to different phosphate oxygens of dTdA, with Arg-87 positioned to protonate the leaving 5′-oxygen of dA, thus clarifying the mechanism of hydrolysis. Model building of an additional 5′-dGMP onto the 3′-oxygen of dA placed this third nucleotide onto a surface cleft near residues Glu-80, Asp-83, Lys-84, and Tyr-115 with its 3′-OH group accessible to the solvent, thus defining the size of the substrate binding site as accommodating a trinucleotide. © 1992 Wiley-Liss, Inc.

Based on 8 Co2+ -nucleus distances measured by paramagnetic effects on T1, and 9 measured and 45 lower limit interproton distances determined by 1D and 2D NOE studies of the ternary Ca2+ complex, the conformation of enzyme-bound 3′,5′-pdTp is high-anti (χ = 58 = 10°) with a C2′ endo/O1′ endo sugar pucker (δ = 143 ± 2°), (-) synclinal about the C3′-O3′ bond (ε = 273 ± 4°), trans, gauche about the C4′-C5′ bond (γ = 301 ± 29°) and either (-) or (+) clinal about the C5′-O5′ bond (β = 92 ± 8° or 274 ± 3°). The structure of 3′,5′-pdTp in the crystalline complex differs due to rotations about the C4′-C5′ bond (γ = 186 ± 12°, gauche, trans) and the C5′-O5′ bond [β = 136 + 10°, (+) anticlinal]. The undistorted conformation of enzyme-bound metal-3′,5′-pdTp determined by NMR was docked into the X-ray structure of the enzyme, using 19 intermolecular NOEs from ring proton resonances of Tyr-85, Tyr-113, and Tyr-115 to proton resonances of the inhibitor. van der Waals overlaps were then removed by energy minimization. Subsequent molecular dynamics and energy minimization produced no significant changes, indicating the structure to be in a global rather than in a local minimum. While the metal-coordinated 5′-phosphate of the NMR-docked structure of 3′,5′-pdTp overlaps with that in the X-ray structure, and similarly receives bifunctional hydrogen bonds from both Arg-35 and Arg-87, the thymine, deoxyribose, and 3′-phosphate are significantly displaced from their positions in the X-ray structure, with the 3′-phosphate receiving hydrogen bonds from Lys-49 rather than from Lys-84 and Tyr-85. The repositioned thymine ring permits hydrogen bonding to the phenolic hydroxyl of Tyr-115. These new interactions, found in the NMR docked structure, are supported by reduced affinities for 3′,5′-pdTp by appropriate mutants of staphylococcal nuclease (Chuang, W.-J., Weber, D.J., Gittis, A.G., and Mildvan, A.S. (1993) accompanying paper, this issue). An inner sphere, rather than a second sphere water ligand of the metal, is optimally positioned to donate a hydrogen bond to Glu-43 and to attack the coordinated 5′-phosphate with inversion. It is concluded that the NMR docking procedure can be used to correct structural artifacts created by lattice contacts in crystals, when they occur at or near ligand binding sites, such as the active sites of enzymes. © 1993 Wiley-Liss, Inc.

    Notes: Ferritin is a 24 subunit protein that controls biomineralization of iron in animals, bacteria, and plants. Rates of mineralization vary among members of the ferritin family, particularly between L and H type subunits of animal ferritins which are differentially expressed in various cell types. To examine ferritin from a highly differentiated cell type and to clarify the relationship between ferritin structure and function, bullfrog red cell L ferritin has been cloned, overexpressed in E. coli, and crystallized under two conditions. Crystals were obtained at high ionic strength in the presence of MnCl2 at a concentration comparable to that of the protein and in the presence of MgCl2 at a concentration much higher than that of the protein. Under both crystallization conditions, the crystals are tetragonal bipyramids in the space group F432 with unit cell dimensions a=b=c= 182 ± 0.5 Å. Crystals obtained in the presence of manganese and ammonium sulfate diffract to 1.9 Å, while those obtained in the presence of magnesium and sodium tartrate diffract to 1.6 Å. Isomorphous crystals have been obtained under similar conditions for a site-directed mutant with a reduced mineralization rate in which Glu-57, -58, -59, and -61 are all replaced by Ala. The structure of wild type L-subunit with magnesium has been solved by molecular replacement using the calcium salt of human liver H subunit (Lawson et al., Nature (London) 349:541-544, 1991) as the model. The crystallographic R factor for the 6-2.2 Å shell is 0.21. The overall fold of human H and bullfrog L ferritins is similar with an rms difference in backbone atomic positions of 0.97 Å. The largest structural differences occur in the D helix and the loop connecting the D and E helices of the four helix bundle. Because red cell L ferritin and liver H ferritin show differences in both rates of mineralization and three-dimensional structure, more detailed comparisons of these structures are likely to shed new light on the relationship between conformation and function. © 1994 John Wiley & Sons, Inc.

    Notes: Dimethyl phosphonomethylheptamethylcyclotetrasiloxane (II) and 1, 3-bis(dimethyl phosphonomethyl)tetramethyldisiloxane (III) have been prepared by Arbuzov reaction of trimethyl phosphite with bromomethylheptamethylcyclotetrasiloxane (I) and 1, 3-bis(bromomethyl)tetramethyldisiloxane, respectively. Dimethyl phosphonomethylmethylsiloxane dimethylsiloxane copolymers have been prepared by acid-catalyzed ring-opening polymerization of II with hexamethyldisiloxane (MM) as an end-capping reagent and by reaction of II with III as an end-capping reagent. Dimethylsiloxane polymers with dimethyl phosphonomethyldimethylsiloxy end groups have been prepared by acid-catalyzed polymerization of octamethylcyclotetrasiloxane (D4) and III. Under these conditions hydrolysis of the dimethyl phosphonate ester groups was a problem. On the other hand Arbuzov reaction of trimethyl phosphite with bromomethylmethylsiloxane dimethylsiloxane copolymer gave a dimethyl phosphonomethylmethylsiloxane dimethylsiloxane copolymer with uniform properties. These polymers have been characterized by 1H-, 13C-, 29Si-, and 31P-NMR spectroscopy. Their molecular weight distributions have been determined by gel permeation chromatography (GPC) and their thermal stability measured by TGA.

    Notes: In the X-ray structure of the staphylococcal nuclease-Ca2+ -3′,5′-pdTp complex, the conformation of the inhibitor 3′,5′-pdTp is distroteed Lys-70* and Lys-71* from an adjacent molecule of staphylococcal nuclease (Loll, P.J., Lattman, E.E. Proteins 5 : 183-201, 1989). In order to correct this crystal packing problem, the solution conformation of enzyme-bound 3′,5′-pdTp in the staphylococcal nuclease-metal-pdTp Complex determined by NMR methods was docked into the X-ray structure of the enzyme [Weber, D. J., Serpersu, E. H., Gittis, A. G., Lattman, E. E., Mildvan, A. S. (preceding paper)]. In the NMR-docked structure, the 5′-phophate of 3′,5′-pdTp overlaps with that in the X-ray Structure. However the 3′-phosphate accepts a hydrogen bond from Lys-49 (2.89Å) rather than from Lys-84 (8.63 Å), and N3 of thymine donates a hydrogen bond to the OH of Tyr-115 (3.16 Å) which does not occur in the X-ray structure (5.28 Å). These interactions have been tested by binding studies of 3′,5′-pdTp, Ca2+, and Mn2+ to the K49A, K84A, and Y115A mutants of staphylococcal nuclease using water proton relaxation rate and EPR methods. Each mutant was fully active and structurally intact, as found by CD and two-dimensional NMR spectroscopy, but bound Ca2+ 9.1- to 9.9-fold more weakly than the wild-type enzyme. While thye K84A mutation did not significantly weaken 3′,5′-pdTp binding to the enzyme (1.5 ± 0.7 fold), the K49A mutation weakened 3′,5′-pdTp binding to the enzyme by the factor of 4.4 ± 1.8-fold. Similarly, the Y115A mutation weakened 3′,5′-pdTp binding to the enzyme 3.6 ± 1.6-fold. Comparable weakening effects of these mutations were found on the binding of Ca2+-3′,5′-pdTp. These results are more readily explained by the NMR-docked structure of staphylococcal nuclease-metal-3′,5′-pdTp than by the X-ray structure. © 1993 Wiley-Liss, Inc.

    Notes: The protein structures of six comparative modeling targets were predicted in a procedure that relied on improved energy minimization, without empirical rules, to position all new atoms. The structures of human nucleoside diphosphate kinase NM23-H2, HPr from Mycoplasma capricolum, 2Fe-2S ferredoxin from Haloarcula marismortui, eosinophil-derived neurotoxin (EDN), mouse cellular retinoic acid protein I (CRABP1), and P450eryf were predicted with root mean square deviations on Cα atoms of 0.69, 0.73, 1.11, 1.48, 1.69, and 1.73 Å, respectively, compared to the target crystal structures. These differences increased as the sequence similarity between the target and parent proteins decreased from about 60 to 20% identity. More residues were predicted than form the common region shared by the two crystal structures. In most cases insertions or deletions between the target and the related protein of known structure were not correctly positioned. One two residue insertion in CRABP1 was predicted in the correct conformation, while a nine residue insertion in EDN was predicted in the correct spatial region, although not in the correct conformation. The positions of common cofactors and their binding sites were predicted correctly, even when overall sequence similarity was low. © 1995 Wiley-Liss, Inc.
