    Notizen: In contrast to the significant importance of tropical rainforest ecosystems as one of the major sources within the global atmospheric N2O budget (2.2–3.7 Tg N yr−1), regional estimates of their N2O source strength are still limited and highly uncertain. To contribute toward more reliable estimates of the N2O source strength of tropical rainforest ecosystems on a regional scale, we modified a process-oriented biogeochemical model, PnET-N-DNDC, and parameterized it to simulate C and N turnover and associated N2O emissions in and from tropical rainforest ecosystems. Model modifications included: (1) new parameterizations associated with plant physiology and soil hydrology and the addition of algorithms relating daily leaf litterfall to water stress as well as to daily rainfall to account for the effects of heavy rainfall damage; (2) the development of a denitrifier activity index that depends on soil moisture conditions and influences N turnover by denitrification; and (3) the addition of a biological N fixation algorithm. Daily simulated N2O emissions based on site data were in good agreement (model efficiencies up to 0.83) with field observations in the Wet Tropics of Australia and Costa Rica. The model was even able to reproduce the highly dynamic pattern of N2O emissions with short-term increases during the wet season. Sensitivity analyses demonstrated that the PnET-N-DNDC model was sensitive to changes in soil properties such as pH, clay content, soil organic carbon and climatic factors such as rainfall and temperature. By linking the PnET-N-DNDC model to a geographic information systems database, tropical rainforests in a 9000 km2 area of the Wet Tropics of Australia are estimated to emit 962 t N2O-N yr−1 (2.4 kg N2O-N ha−1 yr−1) between July 1997 and June 1998.

    Notizen: Abstract The present communication defines the conditions under which thioredoxin activates glutamine synthetase from Anabaena cylindrica. Effects are obtained at pH values around neutrality, and the activation is affected by Mg2+ in the assays. The thioredoxin systems from A. cylindrica and spinach are functionally interchangeable in the activation of glutamine synthetase. The enzyme is efficiently activated by thioredoxinm and also by thioredoxinf, but at much higher concentrations. Thioredoxinm has previously been shown to activate NADPH-dependent malate dehydrogenase and isocitrate dehydrogenase from cyanobacteria. It is speculated that thioredoxinm plays a role in the differentiation of vegetative cells to heterocysts.

    Notizen: Abstract Methane emissions from rice paddies were quantified by using an automatic field system stationed in Zhejiang Province, one of the centres for rice cultivation in China. The data set showed pronouned interannual variations over 5 consecutive vegetation periods; by computing average values of all experimental plots the annual emissions were 177 g CH4 m−2 yr−1 in 1987, 50 g CH4 m−2 yr−1 in 1988, and 187 g CH4 m−2 yr−1 in 1989. The field preparations encompassed 4 different treatments: (1) no fertilizers, (2) mineral fertilizer (KCl, K2SO4), (3) organic manure (rape seeed cake, animal manure), (4) mineral fertilizer plus organic manure. The methane emission rates of the different fertilizer treatments did not show significant differences. The mean emission rates, calculated over the entire observation period of 5 seasons, were 30.4 mg CH4 m−2 h−1 (non-fertilized plot) and 28.3 mg CH4 m−2 h−1 (mineral fertilizers). These values indicate a high level of methane production even without additional input of organic material into the rice-soils. In the other plots, the organic fertilizers were added once per vegetation period at app. 1 t fresh weight per ha, a relatively low application rate by agronomical standards. The mean emission rates were 35.1 mg CH4 m−2 h−1 when manure was applied as sole fertilizer and 27.5 mg CH4 m−2 h−1 when applied jointly with potassium fertilizers. Based on the results of this study we estimate a range of 18–28 Tg CH4 yr−1 as the total methane emission from Chinese rice fields. However, more field data from representative sites in China are needed to reduce the uncertainties in this estimate.

    Notizen: Abstract Long lasting batch cultures of Azospirillum brasilense SP 7 ATCC 29145 grown in liquid malate medium for 8–14 days without any fixed nitrogen source exhibited a biphasic nitrogenase activity, when incubated under gas atmospheres of 99.0% N2 and 1.0% O2 or 99.5% N2 and 0.5% O2 respectively. Maximum specific nitrogenase activity was 1100 nmol C2H4·mg protein-1·h-1. Poly-3-hydroxybutanoic acid (PHBA) synthesis and growth of the cells also showed two phases. Maxima and minima of glutamine synthetase activity developed synchronously with nitrogenase activity, whereas those of glutamate dehydrogenase and alanine aminotransferase were reverse. During a 192 h period of growth protein increased 3–4-fold and PHBA 25 fold. At maximum accumulation of the polymer the PHBA-nitrogen ratio was 6:1 or 8:1. Azospirillum brasilense was also able to fix nitrogen on agar surfaces exposed to air, but nitrogen fixation was monophasic under these conditions during a 14 d period. Specific nitrogenase activity was dependent on the type and concentration of the source of fixed nitrogen (leucine, ammonia) in solidified media. With 1 mM leucine maximum specific nitrogenase activity was 110 nmol C2H4·mg protein-1·h-1.

    Notizen: Abstract The present communication describes the properties of isocitrate dehydrogenase in crude extracts from the unicellular Anacystis nidulans and from heterocysts and vegetative cells of Nostoc muscorum and Anabaena cylindrica. The activity levels of this enzyme are much higher in heterocysts than in vegetative cells of N. muscorum and A. cylindrica. Isocitrate dehydrogenase is virtually inactive in vegetative cells of A. cylindrica. The enzyme is negatively regulated by the reduction charge and scarcely affected by oxoglutarate in the three cyanobacteria. The inhibition by ATP and ADP is competitive with respect to isocitrate and NADP+ in A. cylindrica and N. muscorum and noncompetitive in A. nidulans. Isocitrate dehydrogenase from the three cyanobacteria seems to be a hysteretic enzyme. All the experimental data suggest that the major physiological role of isocitrate and the isocitrate dehydrogenase in heterocysts is not to generate reducing equivalents for N2-fixation. Oxoglutarate formed by the enzyme reaction is likely required for the biosynthesis of glutamate inside the heterocysts. Thioredoxin preparations from spinach chloroplasts or from A. cylindrica activate isocitrate dehydrogenase from either heterocysts or vegetative cells of A. cylindrica. Activation is completed within seconds and requires dithiothreitol besides thioredoxin. The thioredoxin preparation which activates isocitrate dehydrogenase also activates NADP+-dependent malate dehydrogenase from spinach chloroplasts or heterocysts of A. cylindrica. Isocitrate dehydrogenase from A. cylindrica is deactivated by oxidized glutathione. It is speculated that isocitrate dehydrogenase and thioredoxin play a role in the differentiation of vegetative cells to heterocysts.

    Notizen: Abstract Four from 18 strains of Erwinia herbicola tested had nitrogenase activity and grew with N2 as sole source of nitrogen under strict anaerobic conditions with a doubling time of 20–24 h. Nitrogenase activity started only 96–120 h after transfer to a special medium maintained under anaerobic conditions. A ten fold increase in protein per culture found after the maximum nitrogenase activity of 80–130 nmol C2H4. mg protein-1·min-1 was accompanied by a fall in pH of the medium (20 mM phosphate buffer and in 125 mM Tris-buffer) from pH 7.2 to 5.4 or less, but only to 6.8 in 100 mM phosphate buffer. In all cases we found a sharp curtailing of nitrogenase activity 48 h after the maximum. The bacteria utilized only 35–50% of the nitrogen fixed for growth. Erwinia herbicola strains differed from two strains of Enterobacter agglomerans in being unable to fix nitrogen on agar surfaces exposed to air. Specific nitrogenase activity in Erwinia herbicola is compared with data reported for other Enterobacteriaceae and is found to be higher than that reported for Klebsiella pneumoniae, Enterobacter cloacae or Citrobacter freundii.

    Notizen: Abstract Oscillating nitrogenase activity in long lasting batch cultures ofAzospirillum brasilense ATCC 29145 is independent of the carbon source malate. With fumarate, succinate or pyruvate as sole carbon source nitrogenase activity is also oscillating. Cultivation in a medium with 20-fold the buffer concentration also results in oscillating nitrogenase activity. Nitrogen-fixing cultures ofAzospirillum brasilense ATCC 29145 excrete ammonia into the culture medium varying between 0.02 and 0.04 mM concentrations. This is not sufficient to cause a drop of nitrogenase activity inAzospirillum brasilense after the first maximum. During growth under nitrogen-fixing conditions with malate as carbon source, the cells excrete significant quantities of succinate into the culture medium. Cultures with only 0.05% malate reutilized the excreted succinate as soon as malate disappeared from the medium. Azospirillum brasilense ATCC 29145 is shown to have the capability of encystation. Encysted cells are different from vegetative cells in their resistance to desiccation, by the spherical shape and by immotility. The results indicate that oscillating nitrogenase activity in long lasting cultures reflects the development from vegetative cells to cysts and again to vegetative cells under microaerobic conditions.

    Notizen: Abstract. The heterotrophic nitrifier Pseudomonas putida aerobically oxidized ammonia to hydroxylamine, nitrite, and nitrate. Product formation was accompanied by a small but significant release of NO, whereas N2O evolution could not be detected under the assay conditions employed. The isolate reduced nitrate to nitrite and partially further to NO under anaerobic conditions. Aerobically grown cells utilized γ-aminobutyrate as a carbon source and as a N-source by ammonification. The physiological experiments, in particular the inhibition pattern by C2H2, indicated that P. putida expressed an ammonia monooxigenase. DNA-hybridization with an amoA gene probe coding for the smaller subunit of the ammonia monooxigenase of Nitrosomonas europaea allowed us to identify, to clone, and to sequence a region with an open reading frame showing distinct sequence similarities to the amoA gene of autotrophic ammonia oxidizers.
