    Notizen: Abstract  The Younger Dryas (YD, dated between 12.7–11.6 ky BP in the GRIP ice core, Central Greenland) is a distinct cold period in the North Atlantic region during the last deglaciation. A popular, but controversial hypothesis to explain the cooling is a reduction of the Atlantic thermohaline circulation (THC) and associated northward heat flux as triggered by glacial meltwater. Recently, a CH4-based synchronization of GRIP δ18O and Byrd CO2 records (West Antarctica) indicated that the concentration of atmospheric CO2 (COatm 2) rose steadily during the YD, suggesting a minor influence of the THC on COatm 2 at that time. Here we show that the COatm 2 change in a zonally averaged, circulation-biogeochemistry ocean model when THC is collapsed by freshwater flux anomaly is consistent with the Byrd record. Cooling in the North Atlantic has a small effect on COatm 2 in this model, because it is spatially limited and compensated by far-field changes such as a warming in the Southern Ocean. The modelled Southern Ocean warming is in agreement with the anti-phase evolution of isotopic temperature records from GRIP (Northern Hemisphere) and from Byrd and Vostok (East Antarctica) during the YD. δ13C depletion and PO4 enrichment are predicted at depth in the North Atlantic, but not in the Southern Ocean. This could explain a part of the controversy about the intensity of the THC during the YD. Potential weaknesses in our interpretation of the Byrd CO2 record in terms of THC changes are discussed.

    Notizen: Abstract Economic analyses of the greenhouse effect are typically carried out within the framework of computable general equilibrium models which represent the climate system by simple two box proxies based upon the pioneering work of Nordhaus. Since errors in predicting the carbon budget can imply high costs, there is some need to include more sophisticated climate models into the economics of global climate change. This paper presents a non-linear pulse representation of the process-based and data-validated Bern carbon model. Compared to the Nordhaus approach this leads to different results with respect to optimal climate policy and atmospheric carbon dioxide concentration. In particular, our results suggest that economic studies which use a Nordhaus representation of the climate system are biased towards high carbon emission and low abatement levels.

    Notizen: The evolution of a film with insoluble surfactant on a wavy horizontal wall differs from flow without surfactant (the way it usually is studied) in that the film passes through different stages. The first stage is as if the surfactant were absent. Once the surface tension gradient - induced by the nonuniform surfactant concentration adsorbed at the free surface - starts resisting the flow effectively, the evolution enters a transitional stage. A final stage is reached once the free surface becomes rigid due to the surface-tension gradient (high elasticity limit) or becomes virtually leveled before the surface-tension gradient is released (low elasticity limit). The velocity profile through the film changes with time, sol fluid is depleted or accumulated at different strata in the film as the flow evolves. The velocity profile and resulting deformations throughout the film can be influenced significantly by the viscosity distribution or stratification, which occurs, for example, when multiple layers of different viscosity are coated simultaneously. A model and applications for the leveling of such a film are presented. The evolution is described in general terms for a film of uniform viscosity and for a film of two discrete layers of different viscosity. Then the three limiting cases are established. For two of these limits, the effect on the exponential decay rate of the flow and the deformation of the different strata or layers is examined when the viscosity is changed in an infinitesimally thin layer or stratum, and in a layer of finite thickness in films of two and three discrete layers.

    Beschreibung: The ATLAS Phase-I upgrade (2019) requires a Trigger and Data Acquisition (TDAQ) system able to trigger and record data from up to three times the nominal LHC instantaneous luminosity. The Front-End LInk eXchange (FELIX) system provides an infrastructure to achieve this in a scalable, detector agnostic and easily upgradeable way. It is a PC-based gateway, interfacing custom radiation tolerant optical links from front-end electronics, via PCIe Gen3 cards, to a commodity switched Ethernet or InfiniBand network. FELIX enables reducing custom electronics in favour of software running on commercial servers. The FELIX system, the design of the PCIe prototype card and the integration test results are presented in this paper.

    Beschreibung: The Gigabit Link Interface Board (GLIB) project is an FPGA-based platform for users of high-speed optical links in high energy physics (HEP) experiments. The project delivers hardware, firmware/software and documentation as well as provides user support. These resources facilitate the development of evaluation platforms of optical links in the laboratory as well as triggering and/or data acquisition systems in beam or irradiation tests of detector modules. This article focuses on the demonstration of a triggering and data acquisition setup for HEP experiments using hardware and firmware/software resources provided by the GLIB project.

    Beschreibung: Natural transformation has a large impact on lateral gene flow and has contributed significantly to the ecological diversification and adaptation of bacterial species. Thermus thermophilus HB27 has emerged as the leading model organism for studies of DNA transporters in thermophilic bacteria. Recently, we identified a zinc-binding polymerization nucleoside triphosphatase (NTPase), PilF, which is essential for the transport of DNA through the outer membrane. Here, we present genetic evidence that PilF is also essential for the biogenesis of pili. One of the most challenging questions was whether T. thermophilus has any depolymerization NTPase acting as a counterplayer of PilF. We identified two depolymerization NTPases, PilT1 (TTC1621) and PilT2 (TTC1415), both of which are required for type IV pilus (T4P)-mediated twitching motility and adhesion but dispensable for natural transformation. This suggests that T4P dynamics are not required for natural transformation. The latter finding is consistent with our suggestion that in T. thermophilus , T4P and natural transformation are linked but distinct systems.

    Beschreibung: The traffic AAA-ATPase PilF is essential for pilus biogenesis and natural transformation of Thermus thermophilus HB27. Recently, we showed that PilF forms hexameric complexes containing six zinc atoms coordinated by conserved tetracysteine motifs. Here we report that zinc binding is essential for complex stability. However, zinc binding is neither required for pilus biogenesis nor natural transformation. A number of the mutants did not exhibit any pili during growth at 64 °C but still were transformable. This leads to the conclusion that type 4 pili and the DNA translocator are distinct systems. At lower growth temperatures (55 °C) the zinc-depleted multiple cysteine mutants were hyperpiliated but defective in pilus-mediated twitching motility. This provides evidence that zinc binding is essential for the role of PilF in pilus dynamics. Moreover, we found that zinc binding is essential for complex stability but dispensable for ATPase activity. In contrast to many polymerization ATPases from mesophilic bacteria, ATP binding is not required for PilF complex formation; however, it significantly increases complex stability. These data suggest that zinc and ATP binding increase complex stability that is important for functionality of PilF under extreme environmental conditions.
