    Notes: Moving intra-axonal organelles demonstrate frequent variations in speed when viewed over several seconds. To evaluate these and other motion variations, a long-term analysis of organelle motion in isolated axoplasm of Myxicola infundibulum was carried out using differential interference contrast optics and analog and digital image enhancement techniques. Motion characteristics of individual organelles were analyzed for periods of up to 58 minutes. Three principle observations on organelle motion were made: (1) Classes of organelles of the same size demonstrated a 5- to 25-fold variation of speed, with the slowest speeds occurring most frequently; (2) organelle speeds over individual translocations (motion without stopping) are inversely proportional to their size, but the speeds calculated for the long-term analysis of organelle motion (total distance travelled/total observation time, including pauses) did not reflect this observation; and (3) organelles displayed variable trip lengths, durations, mean speeds, and pause durations, and the relationships between these variations showed no repetitive patterns. In contrast to reported observations of uniform velocities of organelles moving on isolated microtubule preparations, these observations suggest that a variety of factors must play a role in organelle translocation in Myxicola axoplasm.

    Notes: A scanning electron microscopic cast technique was used to determine the nature of the canalicular system in human cellular cementum. Prior light microscopic observations suggested the presence of two distinct types of lacunae: bonelike and a large irregular type generally confined to the interradicular region. Only the bone-like lacunae were visualized in the SEM cast preparations. The canalicular system associated with the bone-like lacunae was usually continuous from the surface of the dentin to the surface of the cementum in newly-erupted teeth. Casts having a sponge-like configuration were observed near the cementodentinal junction in some of the specimens from the interradicular region. The presence of these casts could not be predicted from prior light microscopic observations and it was concluded that they may represent infiltration of hypomineralized matrix rather than lacunae.

    Notes: Human third molars were partially demineralized in an acid-alcohol solution and embedded in Epon 812. Six-micron sections were cut from regions of the cervical enamel exhibiting prominent Retzius lines. The plastic was removed from the specimens by microincineration and were examined with the scanning electron microscope. The most prominent structural feature seen along the Retzius lines was the cervical translocation of some of the prisms. The scanning electron microscopic images also suggested that prisms were translocating in the transverse plane of the tooth. A series of pores, which appeared to be empty, were observed in association with the translocations occurring along the Retzius lines.

    Notes: A scanning electron microscope cast technique was used to examine the interface between primary and secondary dentine in young and old human teeth. In addition, more traditional methods were used to examine this interface with light microscopy, identical regions being viewed before and after demineralization. No continuity was seen between the tubules in primary dentine and those in irregular secondary dentine in the scanning electron microscope preparations. These preparations did show the tubules to be continuous between primary dentine and regular secondary dentine in young and old teeth. Both the scanning electron microscopic and light microscopic observations suggested that regular secondary dentine becomes highly sclerosed in old teeth.

    Notes: A scanning electron microscopic (SEM) cast method that has been utilized to examine the internal structure of dentine was modified to examine canalicular communications in the cortices of a human femur and ulna. Although some preparations in which all of the matrix was removed were examined, etched preparations were found to be the most informative. Casts of lacunae and canaliculi along with the underlying matrix could be visualized in these preparations. In the femur, whose cortex exhibited a typical lamellar pattern, canalicular communication was seen between first and second generation osteons and occasionally between osteons and interstitial regions. The interstitial regions in the ulna appeared to be primarily woven rather than lamellar bone. Extensive communication between the outermost lacunae of osteons and interstitial regions was observed in the ulna.

    Notes: Catalase activity was determined in human semen by measuring the oxygen burst with a Clark electrode, after H2O2 addition. Significant catalase activities (mean ± SD) were found in migrated, motile spermatozoa (44 ± 17 nmoles O2/min/108 cells) and in seminal plasma of normozoospermic men (129 ± 59 nmoles O2/min/ml). It has been demonstrated that seminal catalase originated from prostate; however, its activity was not correlated with the usual prostatic markers (such as citric acid and zinc). Our data suggest a multiglandular function secreted by this organ. The catalase activities measured in seminal samples from asthenozo-ospermic, infertile men were found lower than those from normozoospermic subjects. The understanding of the relative contribution of the different enzyme systems against O2 toxicity (superoxide dismutase, catalase, glutathione peroxidase) seem to be a priority area of research to understand disturbances of sperm function.

    Notes: To better understand, to optimize, and to validate the technique of intratesticular (i.t.) injection, several parameters related to i.t. injection were examined. Volumes exceeding 50 μl could be injected i.t.; however, testes frequently became excessively turgid and backflow of injected fluids occurred. Thus, a volume of 50 μl or less was deemed optimal for injection. To determine the rate of distribution of substances throughout the testis, trypan blue was injected i.t. near the caudal pole of the testis, and the movement of dye was monitored. Within 2 min, the dye had spread approximately 1 cm from the site of injection, and in 5 min it had spread twice that distance. In 2 h, the dye had become distributed throughout the testis except at its extreme cranial pole. Seminiferous tubules did not take up dye, indicating that the spread of dye was via peritubular lymphatics. Seminiferous tubule histology appeared virtually unaffected by i.t. injection, even at regions adjacent to the site of injection, when a sterile 26-gauge or smaller bore needle was utilized. To determine disappearance from the testis, radiolabeled inulin was injected i.t. Half time for absorption was achieved at 1.75 h. Potential vehicles were expolored in which compounds with a variety of physical properties could be injected. Gum tragacanth, normal saline, ethylene glycol, dimethyl sulfoxide (DMSO) mixed 1:1 with normal saline, sesame oil, and propylene glycol were found to be suitable injection vehicles, whereas ethanol, dissolved in normal saline in concentrations as low as 0.5% was found unsuitable. To assess vehicle efficiency, various vehicles were utilized with a known testicular toxin (taxol) and injected into one testis, and the histology was compared with the contralateral testis injected with vehicle alone. All vehicles, found suitable above, allowed dispersion of taxol to influence areas distant from the site of injection. Intratesticular injection assesses the potential of agents to directly affect the testis, and systemic metabolism is avoided. Their rapid spread throughout the lymphatics of the testes allows seminiferous tubules to be exposed to agents in innocuous vehicles more rapidly and in higher concentration than is often possible when using systemic injections.
