    Notes: Sublingual and submandibular glands were prepared for light and electron microscopy, and for histochemical staining with periodic acid-Schiff (PAS), Alcian Blue (AB) or both (AB-PAS). Between 15 and 17 days post-conception, the sublingual gland undergoes active morphogenesis from a single, solid bud into a branched glandular tree. At 18 days the first overt signs of secretory differentiation appear in the formation of cells with three kinds of secretion granules; that is, electron-dense serous granules, empty-looking mucous granules with fine thread-like substructures, and granules which have the general appearance of mucous granules but also contain an internal, electron-dense core (“mixed” granules). During the period from 18 to 20 days, all three types of granulated cells increase in number, with mucous cells predominating, and they all border directly on the acinar lumina, in seemingly random combinations in different acini. This diversity is reflected in the histochemical staining, since most acini and cells are both PAS- and AB-positive, but a substantial minority stain only with PAS, indicating that they contain serous granules. By comparison, all secretory cells in the submandibular gland stain with PAS but not with AB after the initial appearance of secretory granules at 18 days. From 20 days to birth (at 22 days), the cells with mixed granules disappear, while the cells with serous granules become fewer in number and displaced to the peripheral outpocketings of the acini. As a result of these changes, the general organization in the newborn is similar to that in the adult, i.e., purely mucous acini with serous demilunes.

    Notes: Sublingual glands of rats were prepared for light and electron microscopy and for the histochemical demonstration of myofibrils and alkaline phosphatase (AkPase) activity. Through 17 days in utero, the epithelial cells of the glandular rudiment are relatively undifferentiated. At 18 days, the inner cells of the terminal buds begin to assemble around a lumen and accumulate secretory granules, while the outer cells flatten and form long processes. At 19 days, many of the outer cells have dilated cisternae of rough endoplasmic reticulum engorged with finely granular material. At 20 days, some of the outer cells have thin bands of microfilaments in their processes, suggesting that they are differentiating into myoepithelial cells (MEC). Though the secretory cells are almost mature at birth, only a few of the MEC have myofibrils detected with an actomyosin reaction, and AkPase activity is very weak. Progressive increases in AkPase activity and in myofibril size and number continue until the acini and intercalated ducts are fully invested with mature MEC at about 14 days after birth. Thus, the MEC and secretory cells begin to differentiate at the same time, but the MEC subsequently differentiate asynchronously with the secretory cells and with each other. Although the sublingual MEC are only partly differentiated in the newborn rat, their overall development occurs somewhat more rapidly than in the adjacent submandibular gland.

    Notes: The seromucous acinar cells of the adult rat submandibular gland secrete a characteristic mucin glycoprotein and a family of unusual glutamine/glutamic acid-rich proteins (GRP). Monoclonal antibodies to the mucin and GRP localized in a very few Type III cells in glands of newborn and 1 day-old rats, using light and electron microscopic immunocytochemistry. Both mucin and GRP reactivities were present in the polymorphic Type IIIP granules during the 1st postnatal week. By 9 days after birth, the granules contained both mucin and GRP and were mucous-like in appearance. At earlier stages, however, cells containing only GRP or mucin could be found, indicating that the initiation of GRP and mucin biosynthesis may not be coordinately regulated. No reactivity was seen in the neonatal Type I cells or in duct cells at any age. Northern and Western blot analysis showed GRP mRNA and protein levels to be barely detectable at birth, with marked increases during the first 2 postnatal weeks. In contrast, Western blots of B1-immunoreactive proteins (B1-IP) showed levels highest in the 1st week and markedly decreased in the adult. Immunocytochemical colocalization, using gold particles of different sizes, showed that the B1-IP, mucin, and GRP colocalized in the granules. These results strengthen the hypothesis that the adult acinar cells develop from the neonatal Type III cells. No evidence was obtained for the involvement of Type I cells in the pathway of acinar cell development.
