Zusammenfassung Es werden drei Methoden zur Zukkerbausteinanalytik von Polysacchariden des Weines auf erreichte Ausbeuten, Arbeitsaufwand, Trennleistung, Einsatz für spezielle Problemstellungen und Reproduzierbarkeit miteinander verglichen, und zwar: 1. Hydrolyse mit Trifluoressigsäure und instrumentelle DC, 2. Saeman-Hydrolyse mit Schwefelsäure und HRGC der Alditolacetate, 3. Methanolyse und HRGC der trimethylsilylierten Methylzucker. Für die quantitative Bestimmung der Uronsäuren wird die Trimethylsilylierung benötigt, für eine schnelle Auswahl eignet sich die Dünnschichtchromatographie, für den zur Strukturaufklärung von Polysacchariden notwendigen Nachweis von Zukkerbausteinen in Spurenanteilen ist die Alditolacetatmethode am besten geeignet. Die Methodenwahl ist abhängig von der Zielsetzung.

Summary Three methods for the analysis of the monomer composition of wine polysaccharides were compared with respect to the yields of released sugars, resolution, applicability for special problems and reproducibility. These methods were: 1. Hydrolysis with trifluoroacetic acid and use of thin-layer chromatography (TLC); 2. Saeman-Hydrolysis and high-resolution gas chromatography (HRGC) of the corresponding alditol acetates; and 3. Methanolysis and HRGC of the trimethylsilyl derivatives. Trimethylsilylation is necessary for the quantitative determination of uronic acids. TLC is appropriate for fast screening, whereas for the trace analysis of sugars in wine polysaccharides the alditol acetate procedure is advantageous.
