    Abstract: Fusarium oxysporum f. sp. conglutinans (FOC), the causal agent of cabbage fusarium wilt, is a serious threat to cabbage production in northern China, and most Chinese FOC isolates were identified as FOC race 1 (FOC1). To better understand the genetic diversity of FOC1 in northern China, we collected FOC isolates from five provinces in northern China and identified them as FOC1 through pathogenicity and race test. To evaluate the genome-level diversity of FOC1, we performed a genome assembly for a FOC1 isolate (FoYQ-1) collected from Yanqing, Beijing, where cabbage fusarium wilt was first reported in China. Using resequencing data of FOC1 isolates, we conducted a genome-wide SNP (single nucleotide polymorphism) analysis to investigate the genetic diversity and population structure of FOC1 isolates in northern China. Our study indicated that Chinese FOC1 can be grouped into four populations and revealed that the genetic diversity of FOC1 were closely associated with geographical locations. Our study further suggests that genetic differentiation occurred when FOC1 spread to the northwest provinces from Beijing Province in China. The FOC1 genetic diversity based on whole-genome SNPs could deepen our understanding of FOC1 variation and provide clues for the control of cabbage fusarium wilt in China.

    Abstract: Trichoderma hamatum is a filamentous fungus that serves as a biological control agent for multiple phytopathogens and as an important resource promising for fungicides. However, the lack of adequate knockout technologies has hindered gene function and biocontrol mechanism research of this species. This study obtained a genome assembly of T. hamatum T21, with a 41.4 Mb genome sequence comprising 8170 genes. Based on genomic information, we established a CRISPR/Cas9 system with dual sgRNAs targets and dual screening markers. CRISPR/Cas9 plasmid and donor DNA recombinant plasmid were constructed for disruption of the Thpyr4 and Thpks1 genes. The result indicates the consistency between phenotypic characterization and molecular identification of the knockout strains. The knockout efficiencies of Thpyr4 and Thpks1 were 100% and 89.1%, respectively. Moreover, sequencing revealed fragment deletions between dual sgRNA target sites or GFP gene insertions presented in knockout strains. The situations were caused by different DNA repair mechanisms, nonhomologous end joining (NHEJ), and homologous recombination (HR). Overall, we have successfully constructed an efficient and convenient CRISPR/Cas9 system in T. hamatum for the first time, which has important scientific significance and application value for studies on functional genomics of Trichoderma and other filamentous fungi.

    Abstract: Cytoplasmic male sterility (CMS) is the main mechanism employed to utilize the heterosis of Brassica napus. CMS three-line rapeseed hybrids have dramatically enhanced yield and brought about the global revolution of hybrid varieties, replacing conventional crop varieties. Over the last half century, China has led the development of hybrid Brassica napus varieties. Two sterile lines, polima (pol) and shaan 2A, were of particular importance for the establishment of three-line hybrid systems in rapeseed, which has opened up a new era of heterosis utilization. However, in current breeding practices, it takes up to three years to identify the restorer or maintainer relationship and the cytoplasmic type of any inbred material. This greatly affects the breeding speed of new varieties and inhibits the rapid development of the rapeseed industry. To address this problem, we developed a set of molecular markers for the identification of fertile cytoplasmic gene N and sterile cytoplasmic gene S, as well as for the fertile nucleus gene R and sterile nucleus gene r, based on differences in the gene sequences between the CMS line, maintainer line and restorer line of Brassica napus. Combining these markers can accurately identify the CMS line, maintainer and restorer of both the pol and shaan systems, as well as their hybrids. These markers can not only be used to identify of the maintainer and restorer relationship of inbred materials; they can also be used as general molecular markers to identify the CMS-type hybrid purity of pol and shaan systems.

    Abstract: Cucumber green mottle mosaic virus (CGMMV) belongs to the Tobamovirus genus and is an important quarantine virus of cucurbit crops. Seedborne transmission is one of the principal modes for CGMMV spread, and effective early detection is helpful to prevent the occurrence of the disease. Quantitative real-time reverse-transcription PCR (RT-qPCR) is a sensitive and rapid method for detecting CGMMV nucleic acids, but it cannot distinguish between infectious and noninfectious viruses. In the present work, a propidium monoazide (PMA) assisted RT-qPCR method (PMA-RT-qPCR) was developed to rapidly distinguish infectious and inactive CGMMV. PMA is a photoactive dye that can selectively react with viral RNA released or inside inactive CGMMV virions but not viral RNA inside active virions. The formation of PMA-RNA conjugates prevents PCR amplification, leaving only infectious virions to be amplified. The primer pair cp3-1F/cp3-1R was designed based on the coat protein (cp) gene for specific amplification of CGMMV RNA by RT-qPCR. The detection limit of the RT-qPCR assay was 1.57 × 102 copies·μL−1. PMA at 120 μmol·L−1 was suitable for the selective quantification of infectious CGMMV virions. Under optimal conditions, RT-qPCR detection of heat-inactivated CGMMV resulted in Ct value differences larger than 16 between PMA-treated and non-PMA-treated groups, while Ct differences less than 0.23 were observed in the detection of infectious CGMMV. For naturally contaminated watermelon leaf, fruit and seedlot samples, infectious CGMMV were quantified in 13 out of the 22 samples, with infestation levels of 102~105 copies·g−1. Application of this assay enabled the selective detection of infectious CGMMV and facilitated the monitoring of the viral pathogen in watermelon seeds and tissues, which could be useful for avoiding the potential risks of primary inoculum sources.

    Abstract: The honeycomb sandwich structure is widely used in energy-absorbing facilities because it is lightweight, has a high specific stiffness and high specific strength, and is easy to process. It also has dynamic mechanical characteristics such as a high impact resistance and high energy absorption. To explore the influence of the Poisson’s ratio on the local impact resistance, this paper compares and analyzes the local impact resistance of a series of honeycomb cores with different Poisson’s ratios under the impact of a spherical projectile at different speeds. Three typical honeycombs with negative/zero/positive Poisson ratios (re-entrant hexagon, semi-re-entrant hexagon, and hexagon) are selected to change the geometric parameters in order to have the same relative density and different Poisson ratios (−2.76–3.63). The relative magnitude of the rear face sheet displacement is in the order of negative Poisson’s ratio 〉 zero Poisson’s ratio 〉 positive Poisson’s ratio, which reveals that the honeycomb structure with the positive Poisson’s ratio has better protection ability than the others. Finally, a dual-wall hexagonal honeycomb is proposed. The rear face sheet displacement of the dual-wall hexagonal honeycomb sandwich structure is reduced by 34.4% at 25 m/s compared with the hexagonal honeycomb, which has a better local impact resistance.

    Abstract: The reform and opening up of the Chinese economy over the last 40 years has led to rapid economic development. However, with the rapid expansion of the economy, increasingly serious air pollution is apparent. In order to control urban air pollution effectively, Chinese governments at all levels have invested large sums every year. However, it has become a difficult issue which influences public government decisions with respect to how and according to what standard to distribute financial funds so as to improve air quality while saving money at the same time. Taking Beijing as an example, this paper investigates the ten-year change in the annual daily mean of inhalable particulate matter (PM10), sulfur dioxide (SO2), and nitrogen dioxide (NO2) from the year of 2006 to 2015, researches the invested funds in environmental protection in Beijing, and establishes a relationship between the atmospheric indexes of the above three parameters and government-invested funds in environmental protection. According to model analysis, government financial input has an obvious influence on the improvement of air quality. However, during the long period of financial input, the degree of air quality improvement will reduce gradually as time goes by. There exists a direct link between the effectiveness of government financial input to promote air quality and the air quality index, which means when the pollutant standards index is poor (i.e., the corresponding pollutant concentration is higher), the effectiveness will be more apparent. On the contrary, when the index is at a good level, the effectiveness of government financial input is very small. To achieve the best air quality conditions, the government should set the detailed financial input at or over the first-grade standard according to urban air quality standards.

    Abstract: Injury to corpus cavernosal endothelial cells (CCECs) is an important pathological basis of diabetes mellitus-induced erectile dysfunction (DMED), while low-intensity pulsed ultrasound (LIPUS) has been shown to improve erectile function in DMED. To further understand its therapeutic mechanism of action, in this study, we first demonstrated increased apoptosis and shedding in the CCECs of DMED patients, accompanied by significant mitochondrial injury by immunohistochemistry and electron microscopy of corpus cavernosum tissue. Next, we used advanced glycation end products (AGEs) to simulate the diabetic environment in vitro and found that AGES damaged mitochondria and inhibited angiogenesis in CCECs in a dose-dependent manner, while LIPUS treatment significantly reversed its effects. Mechanistic studies based on transcriptome sequencing showed that LIPUS significantly up-regulated LC3 and PARKIN protein levels in mitochondria, promoted mitophagy, and affected mitochondrial dynamics and reactive oxygen species (ROS) production. In addition, the protective effects of LIPUS were abrogated when mitophagy was inhibited by 3-methyladenine. In summary, LIPUS exerted potent inhibitory effects on AGES-induced CCEC failure via mitophagy, providing a theoretical basis for DMED treatment that encompasses the protection of endothelial structure and function.

    Abstract: Tobacco (Nicotiana tabacum Linn.) is a famous traditional herb used in folk medicine. The essential oils of tobacco have been demonstrated in modern studies to possess antioxidant, anti-inflammatory, and neuroprotective properties, while its anxiolytic effect has not been reported. The purpose of this study was to evaluate the anxiolytic effect of Yunnan tobacco essential oil (YTO) and Zimbabwe tobacco essential oil (ZTO) on mice. The constituents of YTO and ZTO were analyzed by GC/MS. The anxiolytic effect of YTO and ZTO (0.1%, 1%, and 10%, v/v) on male ICR mice was evaluated in the light–dark box test (LDB) and the elevated plus maze test (EPM) test via inhalation and transdermal administration. After the behavioral tests, salivary corticosterone levels in mice were measured. The behavioral analysis showed that the administration of both YTO and ZTO elevated the time that the mice spent in the light chamber in the LDB test compared to the untreated control. In the EPM test, YTO and ZTO increased the time spent in open arms and the number of entries into the open arms. In addition, both YTO and ZTO significantly decreased salivary corticosterone levels in mice (p ≤ 0.001). In summary, our results demonstrated that inhalation and transdermal administration of both YTO and ZTO showed anxiolytic effect on male ICR mice.

    Abstract: The Cu/Zn-zeolitic imidazolate framework (Cu/Zn-ZIF) was synthesized using the traditional hydrothermal method, and its surface morphology was controlled by adding polyvinylpyrrolidone (PVP) during its synthesis. It was then calcined at 800 °C to form the nitrogen-containing carbon material CuZn@NC, which improved the electron transfer rate. Scanning electron microscopy (SEM), X-ray crystal diffraction (XRD), and X-ray photoelectron spectroscopy (XPS) were used to investigate the surface morphology and structure. Finally, the electrochemical sensing platform for luteolin was effectively constructed by changing the metal–ion ratio during synthesis to achieve the most suitable electrode material. The sensor platform detects luteolin well, with an operating curve equation of Ip (A) = 0.0571C (nM) − 1.2913 and a minimum detection limit of 15 nM, and the platform has been successfully employed for luteolin detection in real samples.
