    Kurzfassung: Many studies reported that long noncoding RNAs (lncRNAs) play a critical role in gastric cancer (GC) metastasis and tumorigenesis. However, the underlying mechanisms of lncRNAs in GC remain unexplored to a great extent. LINC01537 expression level was detected using quantitative reverse transcription-polymerase chain reaction (qRT-PCR) and immunohistochemistry (IHC). Its biological roles in GC were then investigated using functional experiments. In order to investigate the underlying mechanism of LINC01537 in GC, RNA pull-down, RNA immunoprecipitation, and ubiquitination assays were performed. LINC01537 was significantly overexpressed in GC tissues and associated with a poor prognosis. Functional experimental results revealed that LINC01537 promoted the proliferation, invasion, and migration of GC cells. The animal experiments revealed that LINC01537 promoted tumorigenesis and metastasis in vivo. Mechanistically, LINC01537 stabilizes RIPK4 by reducing the binding of RIPK4 to TRIM25 and reducing its ubiquitination degradation, thereby promoting the expression of the NF-κB signaling pathway. According to our findings, the LINC01537-RIPK4-NF-κB axis promoted GC metastasis and tumorigenesis.

    Kurzfassung: Microbial genome sequencing has uncovered a myriad of natural products (NPs) that have yet to be explored. Bacteria in the genus Pseudomonas serve as pathogens, plant growth promoters, and therapeutically, industrially, and environmentally important microorganisms. Though most species of Pseudomonas have a large number of NP biosynthetic gene clusters (BGCs) in their genomes, it is difficult to link many of these BGCs with products under current laboratory conditions. In order to gain new insights into the diversity, distribution, and evolution of these BGCs in Pseudomonas for the discovery of unexplored NPs, we applied several bioinformatic programming approaches to characterize BGCs from Pseudomonas reference genome sequences available in public databases along with phylogenetic and genomic comparison. Our research revealed that most BGCs in the genomes of Pseudomonas species have a high diversity for NPs at the species and subspecies levels and built the correlation of species with BGC taxonomic ranges. These data will pave the way for the algorithmic detection of species- and subspecies-specific pathways for NP development.

    Kurzfassung: Artifacts are irreplaceable treasures of human culture, and transportation monitoring is critical for safeguarding valuable artifacts against damage during culture exchanges. However, current collision-monitoring technologies have limitations in regard to real-time monitoring, cushioning protection, and power supply requirements. Here, we present a method for constructing a smart artifact-monitoring system (SAMS) based on a porous carbon black (CB)/Ecoflex triboelectric nanogenerator (PCE-TENG) that can monitor collisions in real time and absorb vibrations during artifact transportation. The PCE-TENG is assembled using a flexible printed circuit board (FPCB) and a porous Ecoflex layer with CB powder. It exhibits cushioning protection, stretchability, pressure sensitivity, and durability. To enhance its electrical output, modifications were made to optimize the CB content and surface structure. The SAMS comprises six PCE-TENGs attached to the inner wall of the artifact transport package and enables collision monitoring and protection in different directions. Moreover, the SAMS has the capability to instantly transmit warning information to monitoring terminals in the event of improper operations, empowering carriers to promptly and efficiently safeguard artifacts by taking necessary measures. This paper presents a practical strategy for artifact transportation monitoring and package engineering that could have significant implications for the field.

    Kurzfassung: Background: In order to study the applicability of Low Vibration Track (LVT) in heavy-haul railway tunnels, this paper carried out research on the dynamic effects of LVT heavy-haul railway wheels and rails and provided a technical reference for the structural design of heavy-haul railway track structures. Methods: Based on system dynamics response sensitivity and vehicle-track coupling dynamics, the stability of the upper heavy-haul train, the track deformation tendency, and the dynamic response sensitivity of the vehicle-track system under the influence of random track irregularity and different track structure parameters were calculated, compared and analyzed. Results: Larger under-rail lateral and vertical structural stiffness can reduce the dynamic response of the rail system. The vertical and lateral stiffness under the block should be set within a reasonable range to achieve the purpose of reducing the dynamic response of the system, and beyond a certain range, the dynamic response of the rail system will increase significantly, which will affect the safety and stability of train operation. Conclusions: Considering the changes of track vehicle body stability coefficients, the change of deformation control coefficients, and the sensitivity indexes of dynamic performance coefficients to track structure stiffness change, the recommended values of the vertical stiffness under rail, the lateral stiffness under rail, the vertical stiffness under block, and the lateral stiffness under block are, respectively 160 kN/mm, 200 kN/mm, 100 kN/mm, and 200 kN/mm.

    Kurzfassung: Natural products play vital roles against infectious diseases since ancient times and most drugs in use today are derived from natural sources. Worldwide, multi-drug resistance becomes a massive threat to the society with increasing mortality. Hence, it is very crucial to identify alternate strategies to control these ‘super bugs’. Pseudomonas aeruginosa is an opportunistic pathogen reported to be resistant to a large number of critically important antibiotics. Quorum sensing (QS) is a cell–cell communication mechanism, regulates the biofilm formation and virulence factors that endow pathogenesis in various bacteria including P. aeruginosa. In this study, we identified and evaluated quorum sensing inhibitors (QSIs) from plant-based natural products against P. aeruginosa. In silico studies revealed that catechin-7-xyloside (C7X), sappanol and butein were capable of interacting with LasR, a LuxR-type quorum sensing regulator of P. aeruginosa. In vitro assays suggested that these QSIs significantly reduced the biofilm formation, pyocyanin, elastase, and rhamnolipid without influencing the growth. Especially, butein reduced the biofilm formation up to 72.45% at 100 µM concentration while C7X and sappanol inhibited the biofilm up to 66% and 54.26% respectively. Microscale thermophoresis analysis revealed that C7X had potential interaction with LasR (KD = 933±369 nM) and thermal shift assay further confirmed the biomolecular interactions. These results suggested that QSIs are able to substantially obstruct the P. aeruginosa QS. Since LuxR-type transcriptional regulator homologues are present in numerous bacterial species, these QSIs may be developed as broad spectrum anti-infectives in the future.

    Kurzfassung: The erosion of the valve core causes valve failure problems. Thus, a novel method to extend the erosion resistance of the valve was innovatively proposed, namely, nanosecond laser ablation micro-pits on the substrate surface and high velocity oxygen fuel (HVOF) spraying WC coating to extend the erosion resistance of the valve. The characterization and evaluation of the erosion resistance of the WC-sprayed coating after the pretreatment of the 3Cr13 substrate surface polishing/grit blasting/nanosecond laser ablation circular micro-dimple were conducted using the unit coupon erosion test of liquid–solid two-phase flow, followed by the test evaluation and analysis of the erosion resistance test of the WC coating after different pretreatments of the full-size valve core. Results showed that the micro-dimple pretreatment on the surface of the 3Cr13 substrate increased the contact area rate and bonding strength of the substrate and the WC coating. By taking erosion volume loss as the evaluation index, the erosion resistance of the micro-dimple pretreatment on the surface of the 3Cr13 substrate was increased by about 31.98% compared with that of the polishing pretreatment. Therefore, the new method of surface nanosecond laser texture pretreatment and HVOF-spraying WC coating can effectively improve the erosion resistance of the valve.

    Kurzfassung: The pressure-controlled injection tool (PCIT) is the key equipment in the process of high-pressure water jet fragmentation in the solid fluidization exploitation of deep-sea natural gas hydrate (NGH). The internal flow field erosion wear numerical simulation model of PCIT is established through computational fluid dynamics software to study the influence law and main factors of the drilling fluid erosion wear of PCIT. The influence laws of different drilling fluid physical parameters and different structural parameters on PCIT erosion wear were analyzed based on the Euler–Lagrangian algorithm bidirectional coupled discrete phase model (DPM) and the solid–liquid two-phase flow model. The results show that the easily eroded areas are the cone of the sliding core, the plug transition section, the plug surface, and the axial flow passage. The sliding core inlet angle and solid particle size are the main factors affecting the PCIT erosion rate. When the inlet angle of the sliding core is 30°, the diameter of solid-phase particles in drilling fluid is less than 0.3 mm, and the erosion degree of the PCIT could be effectively reduced. The research results can provide guidance for the design and application of the PCIT and advance the early realization of the commercial exploitation of hydrate.

    Kurzfassung: Dual-layer pipe dual-gradient drilling technology is an emerging technology for solving the problem of the narrow safety density window in deepwater drilling. The unstable spacer fluid interface in this technology directly affects the dual-gradient pressure system in the annulus, causing changes in the drilling mud performance and affecting the control of bottom hole pressure and rock removal with drilling mud. Therefore, the key to the stable operation of dual-layer pipe dual-gradient drilling technology is to maintain the stability of the spacer fluid interface. Based on this, a seawater-spacer fluid-drilling mud annular flow model was established in this study, with a bottom hole pressure control step of 0.2 MPa, and the spacer fluid height after a single control was used as the evaluation index to study the influence of annular flow velocity, the spacer fluid properties, and the drill string rotation speed on the stability of the spacer fluid interface. The results show that in the determined conditions of the seawater and drilling mud system, the annular fluid flow rate and the physical parameters of the spacer fluid are the main factors affecting the stability of the spacer fluid interface. When the annular fluid flow rate increased within the range of 0.04~0.2 m/s, the liquidity index of the spacer fluid increased between 0.5 and 0.9, the consistency coefficient increased in the range of 0.6 to 1.4 Pa⋅sn, and the stability of the spacer fluid interface decreased. However, the stability of the spacer fluid interface increased with the increase in its density in the range of 1100~1500 kg/m3. The results obtained in this study can provide a reference for selecting the operating parameters to ensure the stable operation of dual-gradient pressure systems.
