    Kurzfassung: Mast cells (MCs) are an important treatment target for high-affinity IgE Fc receptor (FcεRI)-mediated allergic diseases. The plant-derived molecule 4-methylumbelliferone (4-MU) has beneficial effects in animal models of inflammation and autoimmunity diseases. The aim of this study was to examine 4-MU effects on MC activation and probe the underlying molecular mechanism(s). We sensitized rat basophilic leukemia cells (RBLs) and mouse bone marrow-derived mast cells (BMMCs) with anti-dinitrophenol (DNP) immunoglobulin (Ig)E antibodies, stimulated them with exposure to DNP-human serum albumin (HSA), and then treated stimulated cells with 4-MU. Signaling-protein expression was determined by immunoblotting. In vivo allergic responses were examined in IgE-mediated passive cutaneous anaphylaxis (PCA) and ovalbumin (OVA)-induced active systemic anaphylaxis (ASA) mouse models. 4-MU inhibited β-hexosaminidase activity and histamine release dose-dependently in FcεRI-activated RBLs and BMMCs. Additionally, 4-MU reduced cytomorphological elongation and F-actin reorganization while down-regulating IgE/Ag-induced phosphorylation of SYK, NF-κB p65, ERK1/2, p38, and JNK. Moreover, 4-MU attenuated the PCA allergic reaction (i.e., less ear thickening and dye extravasation). Similarly, we found that 4-MU decreased body temperature, serum histamine, and IL4 secretion in OVA-challenged ASA model mice. In conclusion, 4-MU had a suppressing effect on MC activation both in vitro and in vivo and thus may represent a new strategy for treating IgE-mediated allergic conditions.

    Kurzfassung: After 2000, China’s vegetation underwent great changes associated with climate change and urbanization. Although many studies have been conducted to quantify the contributions of climate and human activities to vegetation, few studies have quantitatively examined the comprehensive contributions of climate, urbanization, and CO2 to vegetation in China’s 32 major cities. In this study, using Global Land Surface Satellite (GLASS) fractional vegetation cover (FVC) between 2001 and 2018, we investigated the trend of FVC in China’s 32 major cities and quantified the effects of CO2, urbanization, and climate by using generalized linear models (GLMs). We found the following: (1) From 2001 to 2018, the FVC in China generally illustrated an increasing trend, although it decreased in 23 and 21 cities in the core area and expansion area, respectively. (2) Night light data showed that the urban expansion increased to varying degrees, with an average increasing ratio of approximately 168%. The artificial surface area increased significantly, mainly from cropland, forest, grassland, and tundra. (3) Climate factors and CO2 were the major factors that affected FVC change. The average contributions of climate factors, CO2, and urbanization were 40.6%, 39.2%, and 10.6%, respectively. This study enriched the understanding of vegetation cover change and its influencing factors, helped to explain the complex biophysical mechanism between vegetation and environment, and guided sustainable urban development.

    Kurzfassung: Grasslands are crucial ecosystem biomes for breeding livestock and combatting climate change. By 2018, the national nature reserves (NNRs) in the Inner Mongolia Autonomous Region (IMAR) had constituted 8.55% of the land area. However, there is still a knowledge gap about their effectiveness in grasslands. Based on a multiyear time series of the growing season composite from 2000 to 2020, we proposed an effectiveness score to assess the effectiveness of the NNRs, using the 250 m MOD13Q1 NDVI data with Theil–Sen and Mann–Kendall trend analysis methods. We found the following: 22 of 30 NNRs were deemed effective in protecting the Inner Mongolian grasslands. The NNRs increased pixels with a sustainable trend 19.26% and 20.55% higher than the unprotected areas and the IMAR, respectively. The pixels with a CVNDVI 〈 0.1 (i.e., NDVI coefficient of variation) in the NNRs increased 〉 35.22% more than those in the unprotected areas and the IMAR. The NDVI changes within the NNRs showed that 63.64% of NNRs had a more significant trend of greening than before the change point, which suggests a general greening in NNRs. We also found that the NNRs achieved heterogeneous effectiveness scores across protection types. Forest ecology protection and wildlife animal protection types are the most efficient, whereas wildlife vegetation protection is the least effective type. This study enriches the understanding of grassland conservation and sheds light on the future direction of the sustainable management of NNRs.

    Kurzfassung: In this work, we propose novel layer-structured polymer composites (PCs) for manipulating the electromagnetic (EM) wave transport, which holds unique electromagnetic interference (EMI) shielding features. The as-prepared PCs with a multilayered structure exhibits significant improvement in overall EMI shielding effectiveness (EMI SE) by adjusting the contents and distribution of electrical and magnetic loss fillers. The layer-structured PCs with low nanofiller content (5 wt % graphene nanosheets (GNSs) and 15 wt % Fe3O4) and a thickness of only 2 mm exhibited ultrahigh electrical conductivity and excellent EMI SE, reaching up to 2000 S/m and 45.7 dB in the X-band, respectively. The increased EMI SE of the layer-structured PCs was mainly based on the improved absorption rather than the reflection of electromagnetic waves, which was attributed to the “absorb-reflect-reabsorb” process for the incident electromagnetic waves. This work may provide a simple and effective approach to achieve new EMI shielding materials, especially for absorption-dominated EMI shielding.

    Kurzfassung: Spermatogenesis, sperm motility, and apoptosis are dependent on the regulation of glandular hormones and mitochondria. Natural astaxanthin (ASTA) has antioxidant, anti-inflammatory, and anti-apoptotic properties. The present study evaluates the effects of ASTA on testosterone synthesis and mitochondrial function in aging roosters. Jinghong No. 1 layer breeder roosters (n = 96, 53-week old) were fed a corn–soybean meal basal diet containing 0, 25, 50, or 100 mg/kg ASTA for 6 weeks. The levels of plasma reproductive hormones and the mRNA and protein levels of molecules related to testosterone synthesis were significantly improved (p 〈 0.05) in the testes of the ASTA group roosters. In addition, antioxidant activities and free radical scavenging abilities in roosters of the ASTA groups were higher than those of the control group (p 〈 0.05). Mitochondrial electron transport chain complexes activities and mitochondrial membrane potential in sperm increased linearly with dietary ASTA supplementation (p 〈 0.05). The levels of reactive oxygen species and apoptosis factors decreased in roosters of the ASTA groups (p 〈 0.05). Collectively, these results suggest that dietary ASTA may improve testosterone levels and reduce sperm apoptosis, which may be related to the upregulation of the testosterone synthesis pathway and the enhancement of mitochondrial function in aging roosters.

    Kurzfassung: The circadian rhythm of melatonin secretion in the pineal gland is highly conserved in vertebrates. Melatonin levels are always elevated at night. Acetylserotonin O-methyltransferase (ASMT) is the last enzyme in the regulation of melatonin biosynthesis (N-acetyl-5-hydroxytryptamine-melatonin). S-adenosylmethionine (SAM) is an important methyl donor in mammals and can be used as a substrate for the synthesis of melatonin. Methionine adenosyltransferase (MAT) catalyzes the synthesis of SAM from methionine and ATP and has a circadian rhythm. CircRNA is an emerging type of endogenous noncoding RNA with a closed loop. Whether circRNAs in the pineal gland can participate in the regulation of melatonin synthesis by binding miRNAs to target mat2a as part of the circadian rhythm is still unclear. In this study, we predicted the targeting relationship of differentially expressed circRNAs, miRNAs and mRNAs based on the results of rat pineal RNA sequencing. Mat2a siRNA transfection confirmed that mat2a is involved in the synthesis of melatonin. Circ-ERC2 and miR-125a-5p were screened out by software prediction, dual-luciferase reporter experiments, cell transfection, etc. Finally, we constructed a rat superior cervical ganglionectomy model (SCGx), and the results showed that circ-ERC2 could participate in the synthesis of melatonin through the miR-125a-5p/MAT2A axis. The results of the study revealed that circ-ERC2 can act as a molecular sponge of miR-125a-5p to regulate the synthesis of melatonin in the pineal gland by targeting mat2a. This experiment provides a basis for research on the circadian rhythm of noncoding RNA on pineal melatonin secretion.

    Kurzfassung: Prolactin (PRL) is an important hormone that is secreted by the pituitary gland and plays an important role in the growth, development and reproduction of organisms. Thyrotropin-releasing hormone (TRH) is a common prolactin-releasing factor that regulates the synthesis and secretion of prolactin. In recent studies, microRNAs (miRNAs) have been found to play a key role in the regulation of pituitary hormones. However, there is a lack of systematic studies on the regulatory role that TRH plays on the pituitary transcriptome, and the role of miRNAs in the regulation of PRL synthesis and secretion by TRH lacks experimental evidence. In this study, we first investigated the changes in PRL synthesis and secretion in the rat pituitary gland after TRH administration. The results of transcriptomic analysis after TRH treatment showed that 102 genes, including those that encode Nppc, Fgf1, PRL, Cd63, Npw, and Il23a, were upregulated, and 488 genes, including those that encode Lats1, Cacna2d1, Top2a, and Tfap2a, were downregulated. These genes are all involved in the regulation of prolactin expression. The gene expression of miR-126a-5p, which regulates the level of PRL in the pituitary gland, was screened by analysis prediction software and by a dual luciferase reporter system. The data presented in this study demonstrate that TRH can regulate prolactin synthesis and secretion through miR-126a-5p, thereby improving our understanding of the molecular mechanism of TRH-mediated PRL secretion and providing a theoretical basis for the role of miRNAs in regulating the secretion of pituitary hormones.

    Kurzfassung: Polydimethylsiloxane (PDMS) foam materials with lightweight, excellent oil resistance and mechanical flexibility are highly needed for various practical applications in aerospace, transportation, and oil/water separation. However, traditional PDMS foam materials usually present poor chemical resistance and easily swell in various solvents, which greatly limits their potential application. Herein, novel fluorosilicone rubber foam (FSiRF) materials with different contents of trifluoropropyl lateral groups were designed and fabricated by a green (no solvents used) and rapid ( 〈 10 min foaming process) foaming/crosslinking approach at ambient temperature. Typically, vinyl-terminated poly(dimethyl-co-methyltrifluoropropyl) siloxanes with different fluorine contents of 0–50 mol% were obtained through ring-opening polymerization to effectively adjust the chemical resistance of the FSiRFs. Notably, the optimized FSiRF samples exhibit lightweight (~0.25 g/cm−3), excellent hydrophobicity/oleophilicity (WCA 〉 120°), reliable mechanical flexibility (complete recovery ability after stretching of 130% strain or compressing of 〉 60%), and improved chemical resistance and structural stability in various solvents, making them promising candidates for efficient and continuous oil–water separation. This work provides an innovative concept to design and prepare advanced fluorosilicone rubber foam materials with excellent chemical resistance for potential oil–water separation application.
