    Notes: A liquid handling system for setting up crystallization experiments by the hanging-drop technique has been developed. The system, organized about a pipetting station and multiport rotary valve under computer control, increases the speed at which experiments can be set up and facilitates systematic studies of protein crystallization. The system is described and results of crystallization trials on 14 proteins are reported.

    Notes: When two similar protein structures are compared, it is often difficult to analyse, or even identify, small differences, especially if the proteins are large. A method is described in which the refined coordinates of two nearly identical protein structures are compared by vector averaging of the differences in atomic coordinates over several consecutive residues. This technique detects regions of the protein where all the atoms move in the same direction. The magnitude and direction of the resultant average vector give the local conformational difference between the two structures. Regions with such correlated movements have been identified in several different protein structures.

    Notes: The program package UNISOFT has been developed for lattice-dynamical calculations. Phonon dispersion curves can be calculated for any crystal structure with up to 20 atoms per primitive cell. The interaction between each individual pair of atoms can be modelled by one or more interaction types such as longitudinal and transverse springs (Born–von Karman model), Born–Mayer potential, Lennard-Jones potential, van der Waals potential, Coulomb potential, shell model. In addition to the model calculation part, UNISOFT contains several auxiliary programs dealing with group-theoretical analysis, neighbourhood analysis, plot of the phonon dispersion, calculation of partial derivatives, check of rotational invariance, calculation of local electrical fields and gradients, calculation of phonon intensities. In combination with a spectrometer for inelastic neutron scattering, UNISOFT provides a tool for the optimization of experiments as well as for a first interpretation of the results.

    Notes: Two data sets containing altogether 131 Bragg intensities have been collected from a CaF2 crystal of 200 μm3 volume, which corresponds to a cube of about 6 μm edge length. The scattering power S of this crystal amounts to 1.3 × 1014. It is the smallest crystal ever used for an X-ray diffraction experiment. Crystal orientation and data collection were carried out at the storage ring DORIS II, HASYLAB, DESY, Hamburg, with the wavelength λ = 0.91 Å. For the first data set all reflections up to sin θ/λ = 0.58 Å-1 were collected, for the second only those reflections considered as observed by a prescan were measured up to sin θ/λ = 0.78 Å-1. The refinements, carried out as a function of the degree of polarization K, converged to Rw(F) = 0.005 for K = 0.93 and Rw(F) = 0.031 for K = 0.94 for the first and second data set, respectively. For comparison, a 90 μm sphere was investigated with synchrotron radiation [sin θ/λ ≤ 0.91 Å-1, Rw(F) = 0.020, K = 0.50] and with a normal X-ray tube [Mo Kα Rw(F) = 0.013]. The intensities are strongly affected by secondary extinction, in contrast to the 6 μm crystal. The following isotropic temperature factors were obtained for the 6 μm, 90 μm (synchrotron) and 90 μm (X-ray tube) data: B(Ca) = 0.38 (5), 0.64 (3), 0.610 (6); B(F) = 0.83 (4), 0.92 (3), 0.812 (8) Å2. The TDS correction increased only the temperature factors from the data set measured with the X-ray tube owing to the difference in FWHM (0.03° for synchrotron radiation, 0.210° for the X-ray tube).

    Notes: Based on the concept of bond polarizabilities the optical properties of a crystal are traced back to the orientation of its bonds and to the individual bond parameters \bar \betaand Δβ. A test with various sulfate compounds shows that the average bond parameters \bar \beta describe the average refractive indices with a higher accuracy than the traditional model of ionic polarizabilities. The anisotropic part of a bond polarizability, Δβ, is considered to be the origin of optical birefringence. Values for Δβ are evaluated in MCO3 and MSO4 (M = Mg, Ca, Sr, Ba). In different crystals the same values for Δβ have been discovered for the C-O and the S-O bonds. Δβ of the M-O bonds depends in a characteristic way on the bond length. A simple method is used to calculate local field effects in accordance with the model of bond polarizabilities. Possible applications of bond polarizabilities for the determination of structural quantities are discussed.

    Notes: The analysis of the full-width at half-maximum (FWHM) of X-ray diffraction profiles measured with monochromatic synchrotron radiation enables the evaluation of the mosaic spread of the sample. This method presupposes the high collimation of synchrotron radiation (FWHM ̃20” of arc). As an example the results from a CaF2 crystal sphere (90 μm diameter) are presented. The reflections strongly affected by extinction show a significantly greater FWHM, thereby indicating at once the need for an extinction correction. A mosaic spread of 75(3)” of arc was determined from the FWHM of all reflections recorded in the step-scan mode, which was used to correct the secondary extinction and to estimate the radius R of the mosaic blocks [R = 4 (1) μm].
