    Abstract: Background. Pathological changes of the adrenal gland and the possible underlying molecular mechanisms are currently unclear in the case of atherosclerosis (AS) combined with chronic stress (CS). Methods. New Zealand white rabbits were used to construct a CS and AS animal model. Proteomics and bioinformatics were employed to identify hub proteins in the adrenal gland related to CS and AS. Hub proteins were detected using immunohistochemistry, immunofluorescence assays, and Western blotting. Real-time quantitative polymerase chain reaction (RT-qPCR) was used to analyze the expression of genes. In addition, a neural network model was constructed. The quantitative relationships were inferred by cubic spline interpolation. Enzymatic activity of mitochondrial citrate synthase and OGDH was detected by the enzymatic assay kit. Function of citrate synthase and OGDH with knockdown experiments in the adrenal cell lines was performed. Furthermore, target genes-TF-miRNA regulatory network was constructed. Coimmunoprecipitation (IP) assay and molecular docking study were used to detect the interaction between citrate synthase and OGDH. Results. Two most significant hub proteins (citrate synthase and OGDH) that were related to CS and AS were identified in the adrenal gland using numerous bioinformatic methods. The hub proteins were mainly enriched in mitochondrial proton transport ATP synthase complex, ATPase activation, and the AMPK signaling pathway. Compared with the control group, the adrenal glands were larger and more disordered, irregular, and necrotic in the AS+CS group. The expression of citrate synthase and OGDH was higher in the AS+CS group than in the control group, both at the protein and mRNA levels ( P 〈 0.05 ). There were strong correlations among the cross-sectional areas of adrenal glands, citrate synthase, and OGDH ( P 〈 0.05 ) via Spearman’s rho analysis, receiver operating characteristic curves, a neural network model, and cubic spline interpolation. Enzymatic activity of citrate synthase and OGDH increased under the situation of atherosclerosis and chronic stress. Through the CCK8 assay, the adrenal cell viability was downregulated significantly after the knockdown experiment of citrate synthase and OGDH. Target genes-TF-miRNA regulatory network presented the close interrelations among the predicted microRNA, citrate synthase and OGDH. After Coimmunoprecipitation (IP) assay, the result manifested that the citrate synthase and OGDH were coexpressed in the adrenal gland. The molecular docking study showed that the docking score of optimal complex conformation between citrate synthase and OGDH was -6.15 kcal/mol. Conclusion. AS combined with CS plays a significant role on the hypothalamic–pituitary–adrenal (HPA) axis, promotes adrenomegaly, increases the release of glucocorticoid (GC), and might enhance ATP synthesis and energy metabolism in the body through citrate synthase and OGDH gene targets, providing a potential research direction for future related explorations into this mechanism.

    Abstract: Unsmooth pulse is one of the most important pulses in TCM diagnostics. We constructed the wave intensity (WI) images of unsmooth pulse based on the pressure wave ( P ), flow velocity wave ( U ), and WI [ ( d P / d t ) ( d U / d t ) ] by ALOKA Prosound α 10 Color Doppler. The characteristic of Cunkou normal pulse could be summarized as follows: compared to Renying pulse, its W 1 amplitude is smaller and NA wave is more obvious, while the W 2 wave is indistinct or even invisible, and the R -1st is longer than that of Renying pulse. The principal U wave of Renying pulse looks like “Λ” shape, while it looks like an arched blunt “ ∩ ” shape in Cunkou pulse, and the amplitude of U wave in Cunkou pulse is smaller. The direction of the principal U wave in Cunkou unsmooth pulse is up, which shows hoof boots “h” shape with high amplitude and a significant notch on declined branch; the amplitude of predicrotic wave in unsmooth pulse P wave is significantly higher, which could be even higher than that of h1, resulting in early appearance of h3 or integrating with h1, which forms a wide and blunt peak. Unsmooth pulse shows poorer vascular elasticity and greater vascular stiffness.

    Abstract: An electron-muon collider with an asymmetric collision profile targeting multi-ab-1 integrated luminosity is proposed. This novel collider, operating at collision energies of, e.g., 20–200 GeV, 50–1000 GeV, and 100–3000 GeV, would be able to probe charged lepton flavor violation and measure Higgs boson properties precisely. The collision of an electron and muon beam leads to less physics background compared with either an electron-electron or a muon-muon collider, since electron-muon interactions proceed mostly through higher-order vector boson fusion and vector boson scattering processes. The asymmetric collision profile results in collision products that are boosted towards the electron beam side, which can be exploited to reduce beam-induced background from the muon beam to a large extent. With this in mind, one can imagine a lepton collider complex, starting from colliding order 10 GeV electron and muon beams for the first time in history and to probe charged lepton flavor violation, then to be upgraded to a collider with 50-100 GeV electron and 1-3 TeV muon beams to measure Higgs properties and search for new physics and finally to be transformed to a TeV-scale muon-muon collider. The cost should vary from order 100 million to a few billion dollars, corresponding to different stages, which make the funding situation more practical.

    Abstract: Bach1 disrupts Wnt/ β -catenin signaling, reduces the proliferation, migration, and tube formation activity of endothelial cells (ECs), and suppresses angiogenesis in mice with surgically induced hind-limb ischemia (HLI). However, the function of Bach1 during developmental angiogenesis in zebrafish remains unclear. Here, we found that zebrafish Bach1 was expressed ubiquitously during early embryonic development in zebrafish. Bach1b mRNA injection of Tg(fli1:gfp) fish disrupted intersegmental vessels (ISV) and dorsal longitudinal anastomotic vessels (DLAV) and suppressed endogenous Wnt/ β -catenin signaling and Wnt8a stimulated vascular endothelial growth factor (VEGF) and interleukin-8 (IL-8) gene expression at early embryonic stages of zebrafish. Furthermore, chromatin immunoprecipitation experiments confirmed that Bach1 occupied the TCF/LEF-binding site of the VEGF promoter in human umbilical vein endothelial cells (HUVECs). Bach1 inhibited VEGF transcription by recruiting histone deacetylase 1 (HDAC1) to the VEGF promoter in HUVECs. Exogenous administration of VEGF or IL-8 partially rescued Bach1-driven antiangiogenic functions in HUVECs. Taken together, these observations indicate that Bach1 suppresses the developmental angiogenesis of zebrafish and that this function is associated with declines in Wnt/ β -catenin signaling and VEGF and IL-8 expression.

    Abstract: Acupuncture strongly alleviates gastrointestinal symptoms and especially promotes gastrointestinal motility. However, the mechanism underlying these processes is poorly understood. This study was designed to examine the effect of electroacupuncture (EA) at gastric back-shu (BL21) and front-mu (RN12) acupoints on gastric motility in functional dyspepsia (FD) rats and to investigate the mechanisms of its effects on the glutamatergic system in the hippocampus. We found that EA at RN12 or BL21 enhanced gastric motility in FD rats, whereas EA at the combination of RN12 and BL21 showed an additional effect. Microdialysis combined with HPLC showed that EA reduced the glutamate content in the hippocampus, and the NMDAR-NO-cGMP signalling pathway was downregulated, as determined by Western blot assays, in FD rats. In addition, we found that decreased gastric motility was significantly restored by the hippocampal infusion of an NMDAR, nNOS, or sGC antagonist. Interestingly, EA had no further effects on gastric motility in the presence of these antagonists in FD rats. Taken together, these results suggest that the hippocampal glutamatergic system is involved in the regulation of gastric motility by EA at RN12 and BL21.

    Abstract: Many new therapies are currently being used to treat cancer. Among these new methods, chemotherapy based on peptides has been of great interest due to the unique advantages of peptides, such as a low molecular weight, the ability to specifically target tumor cells, and low toxicity in normal tissues. In treating cancer, peptide-based chemotherapy can be mainly divided into three types, peptide-alone therapy, peptide vaccines, and peptide-conjugated nanomaterials. Peptide-alone therapy may specifically enhance the immune system’s response to kill tumor cells. Peptide-based vaccines have been used in advanced cancers to improve patients’ overall survival. Additionally, the combination of peptides with nanomaterials expands the therapeutic ability of peptides to treat cancer by enhancing drug delivery and sensitivity. In this review, we mainly focus on the new advances in the application of peptides in treating cancer in recent years, including diagnosis, treatment, and prognosis.

    Abstract: The paper develops a multiclass, multimodal dynamic traffic equilibrium model with consideration of the departure time choice problem. Travelers choose the departure time and the route simultaneously with a Logit-based structure. The route travel cost is a summation of travel time and schedule delay which is associated with arrival time at destination. In addition, the travelers are classified into three groups according to their value of time. A variational inequality (VI) formulation is proposed based on the equilibrium conditions. Two examples are given to testify the effectiveness of the model and the solution algorithm. The model can give the optimal travel route as well as the best departure time, which would contribute to traffic control and dynamic route guidance.

    Abstract: As the most popular renewable energy, solar energy could be converted into electricity by photovoltaic (PV) systems directly. To maximize the effectiveness of the conversion, it is critical to find the precise and accurate parameters of the PV model. In this paper, we propose a level-based learning swarm optimizer with stochastic fractal search (LLSOF) to tackle the parameter estimation of several kinds of solar PV models. The population is separated into multiple levels according to their fitness at first. The individuals at the lower levels evolve through learning from the individuals at the higher levels. Benefiting from the interactive learning among levels, the population could approach the multiple optimal regions rapidly. To enhance the local search ability, stochastic fractal search is introduced to locate the optima accurately. Combination of both, the proposed LLSOF could achieve a good balance on both exploration and exploitation. To evaluate the performance of LLSOF, it is used to obtain the parameters of three PV models and compared with nine well-established algorithms. Comparative results validate the excellent performance of LLSOF. Moreover, the application manufactory’s data sheets report the superior efficiency and effectiveness of LLSOF for the parameter estimation of PV systems.
