    Notizen: Suspension cells of Daucus carota L. cv. Lunga di Amsterdam are extremely sensitive to both nalidixic acid and novobiocine. The synthesis of DNA, RNA and protein are inhibited within minutes of exposure even at concentrations less than 0.1 mM of both drugs. Moreover, uptake of deoxythymidine by cells is impaired more severely than by sodium azide. Reduction of oxygen consumption caused by the two drugs is slower than in the case of NaN3 and occurs a few minutes after the inhibition of macromolecular biosynthesis. This excludes the possibility that the block in oxidation is the primary cause of the observed inhibition of cellular functions and suggests that the inhibition of ATP synthesis, which is immediately affected in both cases, may be responsible for these effects. A possible interaction with other cellular targets may be secondary to the uncoupling effect. Cell growth is completely inhibited by 0.3 mM concentration of both drugs, but this inhibition is irreversible only in the case of novobiocine. The irreversibility of cell growth inhibition by novobiocine can be explained by the total disappearance of ATP from the cell.

    Notizen: Abstract Growth of the cyanobacterium Spirulina platensis, like that of many prokaryotic and eukaryotic organisms, is inhibited by low concentrations of valine, one of the three end-products of the branched-chain amino acid biosynthetic pathway. We assayed and partially characterized the activity of acetyhydroxy acid synthase (AHAS), the first common enzyme of the branched pathway in cell-free extracts from axenic S. platensis cultures. Assays performed at various pH values showed two peaks of activity, both inhibited by valine. FAD was not required for enzyme activity but protected it during dialysis. We also investigated whether the three amino acids were able to cause repression of AHAS synthesis and a significant drop in the enzyme-specific activity could be seen only when cultures were grown in the presence of valine. Chromatography on hydroxylapatite showed one single peak of activity.

    Notizen: Abstract Rats were trained to press a lever under a variable-interval (VI) schedule of water reinforcement. After stable responding had developed, a 4.5-KHz tone (CS) was conditioned classically to a 2.5-mA electric shock (US) in groups of animals which had been given various psychoactive drugs or saline. Twenty-four hours later, a stimulus generalization test was conducted in the absence of drug; during this session, tones that varied in frequency around 4.5 KHz were presented while the animals were responding under the VI schedule. In animals conditioned under saline, all tones (non-differentially) suppressed responding which, however, recovered gradually over time. This suppressive effect was eliminated by lysergic acid diethylamide (LSD; 0.2 and 0.32 mg/kg), cocaine (20 mg/kg), diazepam (2.5 mg/kg), lisuride (0.08 mg/kg), mescaline (20 mg/kg) and 5-methoxy-N,N-dimethyltryptamine (4 mg/kg), and was attenuated by amphetamine (4 mg/kg), pentobarbital (15 mg/kg) and morphine (4 mg/kg). Atropine (10 mg/kg), scopolamine (1 mg/kg), clonazepam (0.5 mg/kg), and chlorpromazine (4 mg/kg) did not alter the suppressive effect of the tone. The serotonin antagonist BC-105 (6 mg/kg) reversed the effect of 0.2 mg/kg of LSD. These results suggest (1) that drug-induced stimuli may “overshadow” other (e.g., external) stimuli during classical conditioning and, (2) that drugs might affect behavior by altering processes (stimulus control or others) that do not simultaneously involve response or motor control.

    Notizen: Abstract Dihydrofolate reductase (DHFR) and thymidylate synthase (TS) activities from cell suspension cultures of Daucus carota were shown to copurify on (NH4)2SO4 fractionation, DEAE Sephadex and methotrexate-Sepharose affinity chromatography and to share approximately the same Mr(183 kDa and 185 kDa respectively) as judged by gel filtration on Sephacryl S-200. The copurified protein migrated as a single band on polyacrylamide gel electrophoresis under denaturing conditions. Both activities could be eluted from the same position of the native gel. Moreover, methotrexate-resistant cell lines which overproduce DHFR revealed to have a parallel higher level of TS. It is therefore proposed and discussed that in carrot, similarly to protozoa, TS and DHFR are present on a single bifunctional polypeptide of 58 kDa.
