    Notizen: Spatially resolved spectroscopic measurements revealed the existence of poloidal vortex motion in spheromaks produced in a conical theta pinch. The poloidal velocity on the axis was estimated to be 5 cm/μsec. Large Doppler broadening, which previously was inferred as high ion temperature, is attributed to large internal fluid velocity. A possible effect of the vortex motion in a spheromak merging experiment is studied and found to enhance the fusion reaction rate as a finite temperature beam collider.

    Notizen: Summary We studied five cases of central nervous system neuronal tumor, one gangliocytoma and four gangliogliomas, both ultrastructurally and immuno-histochemically, using antibodies to neuroendocrine markers including tyrosine hydroxylase (TH), serotonin (5HT), somatostatin (SOM), met-enkephalin (MEK), leu-enkephalin (LEK), substance P (SP), gastrin, vasopressin, oxytocin, vasoactive intestinal polypeptide, adrenocorticotropic hormone and calcitonin. In all cases, the presence of dense-core vesicles (60–250 nm) in the neuronal elements was the characteristic ultrastructural finding. Synapses were observed in two cases. Immunohistochemically, variable numbers of neuronal cells showed positive staining for SOM in five cases, TH, MEK and LEK in three cases, and 5HT and SP in one case each. The others were negative. Positive immunoreactivity for multiple markers was shown in all cases. SOM, TH, 5HT and SP were present in the small- to medium-sized cells, while MEK and LEK were almost exclusively confined to the large cells. Our study clearly indicated that these tumors contained neuronal cells which were not homogeneous with regard to neuroendocrine markers.

    Notizen: Summary The interaction between BW755C (3-amino-1-[m-(trifluoromethyl)phenyl]-2-pyrazoline), a potent inhibitor of both lipoxygenase and cyclo-oxygenase, and respiratory chain in mitochondria and electron transport particles (ETP) from rat livers was examined. BW755C accelerated the oxygen uptake by mitochondria without the addition of substrate for the respiratory chain. Spectrophotometric study revealed that BW755C was quickly oxidized by cytochrome oxidase in mitochondria to a compound possessing an absorption maximum at 524 nm. p-Phenylenediamine (p-diaminobenzene, PPDA), which, like BW755C, serves as an electron donor to cytoschrome oxidase, was shown to inhibit the generation of active oxygen in macrophages; the inhibition was stronger than that of BW755C. These results strongly suggest that the oxidative conversion of BW755C by mitochondrial cytochrome oxidase is associated with its potentially inhibitory action on the active oxygen-generating system in phagocytes.

    Notizen: Summary Ca2+-ATPase activity on the astrocyte plasma membrane was investigated ultracytochemically, using the lead salt technique. Normal astrocytes showed a weak cytochemical reaction for Ca2+-ATPase activity, deposits of the reaction product being small. At 7 and 15 days after cold lesioning, reactive astrocytes apparently in the process of repair of the edematous lesion were observed; these demonstrated an intense cytochemical reaction for Ca2+-ATPase activity in their plasma membranes facing the extracellular fluid, with reaction product accumulation. At 2 months, the lesion had progressed to glial scars containing sporadic microcysts. The reactive astrocytes surrounding the microcysts demonstrated a moderate cytochemical reaction for Ca2+-ATPase activity in their free plasma membranes, whereas those arranged in a cell-to-cell pattern showed little reaction product in their plasma membranes. In conclusion, a more intense cytochemical reaction was always observed in the free plasma membrane of reactive astrocytes.

    Notizen: Summary To investigate the functional relationship between astrocytes and Ca2+-ATPase of cerebral capillary endothelial cells (capillary Ca2+-ATPase), cold lesions were produced and the cytochemical reaction (CR) for Ca2+-ATPase activity and morphological changes of astrocytes were chronologically studied. Under normal conditions, CR for capillary Ca2+-ATPase activity was mild. However, at 20 min after the operation, astrocytic end-feet embracing the capillaries were swollen, and CR was moderate. Deposits of slightly coarsened reaction product (RP) appeared and accumulated on the abluminal surface. CR became stronger as edema fluid accumulated. At 4, 7 and 15 days, detachment of the astrocytic processes from the capillary wall was observed and in the uncovered capillaries, CR was intense, especially on the abluminal surface. It could be thus possible that the enzyme was related to the blood-brain barrier (BBB). At 2 months, reactive astrocytes has recovered lesionresistant capillaries. CR was mild and its associated deposits were coarser, the number decreasing on both surfaces. The nature and localization of the deposits of RP in the scar were different from those under normal conditions, possibly due to the functional differences between normal and reactive astrocytes in the BBB. CR was mild in association with astrocytes embracing the capillary wall and was intense without astrocytes. Therefore, it might be possible that astrocytes exerted certain effects on capillary Ca2+-ATPase activity in relation to BBB function.

    Notizen: Summary The localization and distribution of peroxidase (PPO) activity were studied ultracytochemically in thrombocytes from lampreys, carps, frogs, snakes, tortoises, rabbits, sheep, dogs, and monkeys. PPO activity was not deteetable in the thrombocytes of lampreys, carps, frogs, and snakes. However, this enzyme activity was demonstrated in the nuclear envelope and endoplasmic reticulum of tortoise thrombocytes. Dog and monkey thrombocytes (blood platelets) exhibited PPO activity in the dense tubular system, but this enzyme activity was not detectable in rabbit and sheep thrombocytes. Our observations are interpreted to suggest that thrombocytes from animals lower than amphibia are peroxidase negative. Furthermore, it can be said that thrombocytes from animals higher than reptiles are generally positive, although there are exceptions. PPO activity was localized in the endoplasmic-reticulum system, but not in the cytoplasmic granules of thrombocytes common to submammals and mammals. In this study, we also compared the distribution of peroxidase activity in thrombocytes, neutrophils, and eosinophils and conclude that these are significant differences in the distribution of PPO and myeloperoxidase.

    Notizen: Summary To investigate the pathology of osteonecrosis of the femoral head (ON) in patients with rheumatoid arthritis (RA), we examined 26 hips clinically and histologically. In this study, we diagnosed ON by both the radiological evidence of femoral head collapse, with or without narrowing of the joint space, and by histological evidence of extensive areas of bone necrosis with surrounding reparative new bone. Thus 14 hips were diagnosed as ON, and 12 hips were not. All of the patients with ON had a history of steroid medication. The frequency of lipid-containing osteocytes observed in the subchondral area of the femoral head significantly correlated with the occurrence of ON (P〈0.05). In electron micrographs, these osteocytes showed degenerative features, with their nuclei pressed towards one side of the cell by plump fatty droplets (fatty degeneration). In patients with RA, there was a significant correlation between the appearance of lipid-containing osteocytes and steroid medication (P〈0.05). No relationship existed between the severity of RA synovitis and the occurrence of ON. These data suggest that ON in rheumatoid hips may relate to the administration of a steroid and the fatty degeneration of osteocytes.
