    Notes: Hybrid cells have been recovered from selective culture medium after fusion of concanavalin-A-activated human lymphocytes with an AKR mouse thymoma (BW 5147). After 6 months of culture twenty-seven out of forty-nine clones still contained human chromosomes. Human chromosome 6 was present in 89% of these clones, and human X in 70%. Clones from one hybrid line contained several human chromosomes. In twelve of the clones carrying human chromosomes, the rosetting with sheep Erythrocytes (SRBC) was 3 times as high as In the BW 5147 cell line. All these clones Carried the human chromosome 6, and eight clones contained the human X chromosome as well. In some of these clones (25%) chromosome 6 was the only human one present. In the two clones In which human chromosome 6 was completely missing, the resetting with SRBC was at the level of the BW line. We therefore suggest that genes on human chromosome 6 are responsible for rosetting with SRBC.

    Notes: BALB/c mice were injected with an affinity-chromatography-purified placental transferrin receptor preparation. Spleen cells were fused with NS-1 myeloma cells. Sixteen hybrids producing monoclonal antibodies specific for the transferrin receptor and two hybrids specific for transferrin were identified by radioimmunoassay (RIA). Five hybrids were selected for cloning on the basis of antibody specificity and affinity. None of the antibodies inhibited the binding of transferrin to K562 cells. The binding of antibody ID9 to K562 cells was partially inhibited by transferrin or a polyclonal goat anti-transferrin receptor antiserum. Of the five antibodies, two (IIB6 and IIB2) reacted only with the purified receptor and solubilized cells and not with whole cells. The other three antibodies, when tested with normal human cells and leukaemia and tumour cell lines, showed identical reaction patterns. The antibodies precipitated a glycoprotein from K562 cells with an apparent molecular weight of 94,000, estimated from sodium dodecyl sulphate-polyacrylamide gel electrophoretograms run under reducing conditions, and a molecular weight of 188,000 when run under unreduced conditions. All antibodies have a high affinity with Ka values ranging from 1.44 ± 109 to 3.56 ± 1010 (1/mol). The antigen precipitated by all five antibodies showed identical peptide maps after partial proteolytic digestion.

    Notes: Chemotactic activities of circulating polymorphonuclear leukocytes (PMN) were determined in twenty patients with psoriasis and twenty healthy control persons. After serial dilution of the complement split product C5a and the formylated tripeptide f-met-Ieu-phe (FMLP), chemotaxis profiles showed that PMN migration toward both chemotaxins was significantly increased in psoriasis. In addition, PMN from psoriatic patients responded to chemotaxins at much lower concentrations compared with controls.The liberation of (lysosomal) β-glucuronidase was also determined in cytochalasin B-treated cells confronted with increased concentrations of the chemotaxins. Secretion of this marker enzyme started at lower concentrations in PMN derived from psoriatic patients.Our observations demonstrate migratory and secretory hyper-responsiveness of PMN from psoriatic patients. This may play a role in perpetuating the psoriatic tissue reaction.

    Notes: The chemotactic activity of polymorphonuclear leukocytes (PMNs) directed against serum was determined in a modified Boyden chamber assay. In a total of 176 experiments PMNs and sera from healthy controls were compared with PMNs and sera from patients with psoriasis. When PMNs from patients with psoriasis were confronted with psoriatic serum greatly enhanced chemotaxis was demonstrated. Non-psoriatic PMNs in the presence of psoriatic serum showed no enhancement. However, psoriatic PMNs in the presence of normal serum were chemotactically more active compared to non-psoriatic PMNs. Heat inactivation (56°C, 30 min) reduced the chemotactic activity of all sera by nearly 50%. However in psoriatic sera the enhancement of chemotaxis was still present after heat treatment. The results indicate the presence of a functional abnormality of chemotaxis in psoriasis. This is likely to be caused by the in situ generation of chemotactically active fragments of complement. Experiments showing increased chemotactic activity of sera exposed to migrating PMNs support this concept.

    Notes: Six patients suffering from widespread psoriasis were treated with 20 mg methotrexate (MTX) intramuscularly and the chemotactic activity of peripheral polymorphonuclear leucocytes (PMN) was measured before and after treatment. The modified Boyden chamber method was used and C5a, FMLP, casein and autologous fresh serum served as chemotaxins. Following MTX the chemotactic migration of PMN was inhibited by more than 50% for 2 days and slowly recovered within the following 5 days. The inhibition was present with all of the four chemotaxins used.The results demonstrate that treatment with low dose MTX in patients with psoriasis causes a profound inhibition of PMN function. These findings support the idea that PMN play an important role in the pathogenesis of psoriasis.

Sie ist durch den Beginn an der distalen Beinmuskulatur im frühen Erwachsenenalter und durch beachtliche Erhöhung der Creatin-Kinase (CK)-Aktivität im Serum (20–30fach bei unseren Patienten) charakterisiert. Bei den Eltern, die blutsverwandt sind, besteht keine Myopathie und keine Erhöhung der CK-Aktivität im Serum. Wir nehmen in Anbetracht dieser Sippenanamnese an, daß die hier beschriebene distale Myopathie autosomal recessiv vererbt wurde.
    Notes: Summary Two brothers are described with a distal myopathy different from the known hereditary distal myopathies. Early adult onset, beginning in the distal leg muscles with marked elevation of creatine kinase (CK) activity (20 to 30-fold) were the characteristic features. The parents of the patients had no symptoms or signs of myopathy. Their serum CK-activity was in the normal range. There was parental consanguinity, so the distal myopathy in these brothers is probably an autosomal recessive inheritance.

    Notes: Abstract The chromosome segregation of hybrid myelomas from a fusion of rat immunized spleen cells and the mouse myeloma P3-X63-Ag8 has been analysed. Chromosome loss appears to be nonrandom. Most mouse chromosomes are retained. The rat chromosomes are preferentially lost but a few — particularly 1–5 and 13-are rarely lost. The specific retention of some rat chromosomes explains the stability of rat Ig expression in mouse-rat hybrids. Correlation of chromosome loss and retention and loss of rat heavy and light chains leads us to propose chromosome 14 as coding for the heavy chain.
