Abstract: Tetrodotoxin (TTX) has been widely used in pharmacology, food poisoning analysis, therapeutic use, and neurobiology. In the last decades, the isolation and purification of TTX from natural sources ( e.g. , pufferfish) were mostly based on column chromatography. Recently, functional magnetic nanomaterials have been recognized as promising solid phases for the isolation and purification of bioactive compounds from aqueous matrices due to their effective adsorptive properties. Thus far, no studies have been reported on the utilization of magnetic nanomaterials for the purification of TTX from biological matrices. In this work, an effort has been made to synthesize Fe 3 O 4 @SiO 2 and Fe 3 O 4 @SiO 2 –NH 2 nanocomposites for the adsorption and recovery of TTX derivatives from a crude pufferfish viscera extract. The experimental data showed that Fe 3 O 4 @SiO 2 –NH 2 displayed a higher affinity toward TTX derivatives than Fe 3 O 4 @SiO 2 , achieving maximal adsorption yields for 4epi-TTX, TTX, and Anh-TTX of 97.9, 99.6, and 93.8%, respectively, under the optimal conditions of contact time of 50 min, pH of 2, adsorbent dosage of 4 g L −1 , initial adsorbate concentration of 1.92 mg L −1 4epi-TTX, 3.36 mg L −1 TTX and 1.44 mg L −1 Anh-TTX and temperature of 40 °C. Interestingly, desorption of 4epi-TTX, TTX, and Anh-TTX from Fe 3 O 4 @SiO 2 –NH 2 -TTX investigated at 50 °C was recorded to achieve the highest recovery yields of 96.5, 98.2, and 92.7% using 1% AA/ACN for 30 min reaction, respectively. Remarkably, Fe 3 O 4 @SiO 2 –NH 2 can be regenerated up to three cycles with adsorptive performance remaining at nearly 90%, demonstrating a promising adsorbent for purifying TTX derivatives from pufferfish viscera extract and a potential replacement for resins used in column chromatography-based techniques.

Abstract: A fatal neurotoxic poisoning case happened in Khanh Hoa province in 2020 after eating a certain number of marine gastropods, later identified as Nassarius glans, one of the common marine gastropods in Vietnam. As the remaining causative food in the incident, 62 specimens were collected to examine tetrodotoxin toxicity individual variation and frequency of toxic specimens by using HILIC/MS-MS analysis. 100% of studied specimens exhibited toxicity (556 ± 821 MU/g) beyond the regulatory level of consumption (10 MU/g) for puffer(fish) recommended in Japan and extensive variation (18–4,046 MU/g). The result pointed out that only 5 g of soft tissue from N. glans (equivalent to 2–3 specimens) containing maximum toxicity detected in the present study may cause human death if consumed. Fhe first time, this study identified TTXs in the gastropods as a causative toxin in the poisoning in Vietnam. Moreover, 65.5% of studied specimens with high toxicity higher than 100 MU/g, including 16.1%, showing extremely high toxicity ( 〉 1,000 MU/g). The results suggested that this gastropod is quite dangerous for human consumption and should be alerted to public awareness.

Abstract: Seagrasses are a paraphyletic group of marine angiosperms that evolved three to four times from land plants and returned to the sea. Halophila ovalis, Thalassia hemprichii and Enhalus acoroides (Hydrocharitaceae) are species that can occur in wide salinity ranges. Sulfated polysaccharides (SPs) comprise a complex group of macromolecules with many critical biological functions. We assume that SP may play a role in salt tolerance in seagrass. In this study, three seagrass species collected in both rainy and dry seasons from the fields were analyzed to determine the total SP contents and different functional groups of SP. Quantification of total SP was done by photometric assays. High-performance anion-exchange chromatography with Pulsed Electrochemical Detection (HPAEC) determined different functional groups of SPs. The results indicated higher total SP contents in seagrass are present in plants at higher salinities and environmental temperatures. The percent of functional groups of SPs are present in the following order: glucose 〉 galactose 〉 arabinose 〉 mannose 〉 rhamnose 〉 fucose. The order is not different between the two seasons.

Abstract: Tetrodotoxin analogues (TTXs) used in research and development were originated from biological sources for the last decades. Among TTXs‐bearing aquatic animals, horseshoe crab Carcinoscorpius rotundicauda from Vietnam's Coast was reported to contain a high level of TTXs, indicating a promising source for extraction and purification of TTXs. In this work, horseshoe crab C. rotundicauda collected from Can Gio Coast, Vietnam was used for TTX extraction using homogenization‐assisted method with acetic acid solvent. The extraction process was analysed and optimized by single‐factor investigation and response surface methodology (RSM). Single‐factor investigation pointed out that the optimal conditions for TTX recovery yield from C. rotundicauda 's soft tissues were homogenization time of 20 min, acetic acid concentration of 1.0%, solvent/matrix ratio of 4 mL/g, extraction temperature of 70°C, extraction time of 30 min and number of extraction of two times. The most significant factors including acetic acid concentration, solvent/matrix ratio and extraction temperature were utilized for constructing the RSM model to predict TTX recovery yield. The maximal TTX recovery yield predicted from RSM model was 97.3% under solvent/matrix ratio of 4 mL/g, extraction temperature of 70°C and acetic acid concentration of 1% (v/v). The RSM model displayed a high stability and precision for prediction of TTX recovery yield from experiments of 1–10 kg C. rotundicauda 's soft tissues/batch extractions producing low errors (−1.95% to 1.85%). TTXs in C. rotundicauda 's soft tissues were elucidated by HPLC‐MS/MS, revealing the presence of three analogues TTX, 4epi‐TTX and Anh‐TTX with their contents of 43.02, 22.54 and 45.07 μg/g, respectively.

Abstract: The study was carried out to investigate lipid and protein oxidation of herring (Sardinella gibbosa) that was treated with (2.5% w/v) sodium acetate at 4oC. After 3, 6, 9, and 12 days, lipid and protein oxidation were evaluated by indications such as pH value, peroxide (PV), thiobarbituric acid-reactive substances (TBARS), protease enzyme activity and sulfhydryl. The results showed that, pH was from 6.33 ± 0.005 to 7.42 ± 0.005 (p 〈 0.05); PV was 1.73 ± 0.31 to 7.13 ± 0.5 (mEq/kg) (p 〈 0.05), and TBA was from 53.3 ± 0.28 μmol MDA/kg to 207.4± 0.47 μmol MDA/kg (p 〈 0.05). Protease enzyme activity reduced from 0.323–0.175 U/mg protein (p 〈 0.05), and sulfhydryl was from 14.39–7.198 µmol/g protein (p 〈 0.05). Consequently, sodium acetate effectively prevents lipid and protein oxidation, extending herring meat’s life.
