    Notes: In this study, we evaluated the combination of polymerase chain reaction–restriction fragment length polymorphism (PCR–RFLP) and amplified fragment length polymorphism (AFLP) molecular typing techniques for the analysis of thermophilic campylobacter species isolated from clinical and poultry samples. 23S PCR–RFLP analysis performed to fingerprint 69 strains exhibited an excellent level of typability. Eleven different types were defined at 100% linkage level following numerical analysis of band patterns. Differentiation of Campylobacter jejuni and Campylobacter coli at species level was achieved although no significant relationship could be observed between the profiles and the origin of the strains. Simplified AFLP analysis of the isolates disclosed the presence of 66 different banding patterns. The resulting dendrogram showed a high diversity among the strains studied. All the isolates were grouped within eight main types with a 69% homology degree among them. Differentiation at subspecies level was possible but no significant relationship could be observed between the AFLP profiles and the origin of the strains. When used in combination, 23S PCR–RFLP and single-enzyme AFLP methods can be applied to determine taxonomic and epidemiological relationships among thermophilic campylobacters.

    Notes: Arbitrary primed PCR (AP-PCR) analysis was compared with serotyping as a means of high-resolution typing of Pseudomonas aeruginosa. Seventy-four isolates from 3 different hospitals and 18 reference strains were studied. Serotyping provided good index of discrimination, although eleven isolates could not be serotyped. Genomic DNA was amplified with a single 10 nucleotide primer (sequence 5′-AGG GGT CTT G-3′). The strains were genetically diverse and 61 different AP-PCR profiles of 2–7 bands between 0.3 and 2.4 kb were obtained. AP-PCR profiles were not consistently associated with serotypes, but they clearly subtyped strains of the same serotype. Numerical analysis of AP-PCR patterns defined 7 groups at the 55% similarity level, and identified predominant strains in each hospital. The results show that AP-PCR analysis provides a simple and practical approach to typing P. aeruginosa that is more discriminatory than traditional serotyping scheme. We suggest that maximum discrimination can be achieved by a combination of both methods.

    Notes: Summary Investigations on 24 patients hospitalized for treatment of duodenal ulcer recurrences at the Massachusetts General Hospital revealed that none considered that he had adhered to the prescribed therapy in its entirety. The factors found operative in nonadherence to therapy were presented under four headings: (1) the patient's attitude toward the diagnosis of ulcer; (2) the patient's attitude toward the recommended treatment; (3) the patient's attitude toward the physician; and (4) the response of guilt in the patient who does not adhere to therapy. Cases were presented illustrating the various components in each category, and methods of handling were described.

    Description: Spatially extensive and persistent drought episodes have repeatedly influenced human history, including the 'Strange Parallels' drought event in monsoon Asia during the mid-18th century. Here we explore the dynamics of sustained monsoon failure using observed and tree-ring reconstructed drought patterns and a 1300-year pre-industrial community earth system model control run. Both modern observational and climate model drought patterns during years with extremely weakened South Asian monsoon resemble those reconstructed for the Strange Parallels drought. Model analysis reveals that this pattern arises during boreal spring over Southeast Asia, with decreased precipitation and moisture flux, while related summertime climate anomalies are confined to the Indian subcontinent. Years with simulated South Asian drying exhibit canonical El Niño conditions over the Pacific and associated shifts in the Walker circulation. In contrast, multi-year drought periods, resembling those sustained during the Strange Parallels drought, feature anomalous Pacific warming around the dateline, typical of El Niño Modoki events.

    Description: Transforming mature tourism resorts has evolved toward a greater involvement of public authorities and away from the mere renovation of public spaces. Authorities today are required to lead the reorganization of tourism activities through the development of co-operative networks between all stakeholders involved. In this paper, a participatory integrated approach has been designed and implemented in collaboration with Spanish authorities and the tourism sector to propose a strategy to achieve the renovation of tourism resorts. This methodology was applied to Puerto de la Cruz, the oldest tourism destination in the Canary Islands and a clear paradigm of a consolidated resort. The objective is to define and implement policies to transform Puerto de la Cruz into a more sustainable tourism destination.

    Description: Effect of severe drought events in combination with organic amendments (municipal solid waste, MSW, sheep manure, SM, and cow manure, CM) on soil dehydrogenase, urease, β-glucosidase and phosphatase activities and microbial community by analyzing phospholipid fatty acids was studied under controlled laboratory conditions for one year. Two levels of irrigation were used: (1) watered soils, where the soils were maintained at 60% of their water holding capacity through the experiment, and (2) non-watered soils, without irrigation through the experiment. The severe drought conditions negatively affected the soil enzymatic activities and total bacterial and fungal PLFA concentrations. The application of organic amendments to the soil subjected to severe drought, increased soil water retention and encouraged the growth and activity of soil microbial populations. However, the chemical composition of the organic matter applied to the soil also strongly influenced soil moisture. In non-watered soils and compared with the unamended soil, the dehydrogenase activity was 71.3 %, 60.9% and 38.6% higher in the soil with SM, CM and MSW, respectively. Urease activity was 60.6%, 51.5% and 37% higher in the soil with SM, CM and MSW, respectively. β-glucosidase and phosphatase activities had a similar trend. Water retention was higher when the organic wastes applied to the soils had a higher content of humic acids than fulvic acid contents.

    Description: Although much research has taken place in WiFi indoor localization systems, their accuracy can still be improved. When designing this kind of system, fingerprint-based methods are a common choice. The problem with fingerprint-based methods comes with the need of site surveying the environment, which is effort consuming. In this work, we propose an approach, based on support vector regression, to estimate the received signal strength at non-site-surveyed positions of the environment. Experiments, performed in a real environment, show that the proposed method could be used to improve the resolution of fingerprint-based indoor WiFi localization systems without increasing the site survey effort.

    Description: We propose in this work a technique for determining the amplitude distribution of a wave packet containing a large number of short optical pulses with different amplitudes. The technique takes advantage of the fast response of the optical Kerr effect in a fiber nonlinear optical loop mirror (NOLM). Under some assumptions, the statistics of the pulses can be determined from the energy transfer characteristic of the packet through the NOLM, which can be measured with a low-frequency detection setup. The statistical distribution is retrieved numerically by approximating the solution of a system of nonlinear algebraic equations using the least squares method. The technique is demonstrated numerically in the case of a packet of solitons.

    Description: Brown adipose tissue (BAT) is specialized in non-shivering thermogenesis through the expression of the mitochondrial uncoupling protein-1 (UCP1). In this paper, we describe the relationship between UCP1 and proteins involved in ATP synthesis. By the use of BATIRKO mice, which have enhanced UCP1 expression in BAT, an increase in ATP synthase as well as in ubiquinol cytochrome c reductase levels was observed. Alterations in mitochondrial mass or variations in ATP levels were not observed in BAT of these mice. In addition, using a protocol of brown adipocyte differentiation, the concerted expression of UCP1 with ATP synthase was found. These two scenarios revealed that increases in the uncoupling machinery of brown adypocites must be concomitantly followed by an enhancement of proteins involved in ATP synthesis. These concerted changes reflect the need to maintain ATP production in an essentially uncoupling cell type. J. Cell. Biochem. © 2013 Wiley Periodicals, Inc.
