    Notes: We recently found that alternative transcripts of tissue transglutaminase (tTG or TG2) were present in hippocampal brain regions of Alzheimer's disease (AD), but not in control, non-demented, age-matched brains. Since antecedent non-severe trauma has been implicated in AD and other neurodegenerative diseases, such as Parkinson's disease (PD) and amyotrophic lateral sclerosis (ALS), we were interested in whether alternative transcripts might be detected in a model of neurotrauma, controlled-contusion spinal cord injury (SCI) in the rat. Implicated in diverse roles from growth and differentiation to apoptotic cell death, only bifunctional tTG, of the nine member TG family, has dual catalytic activities: guanine trinucleotide (GTP) hydrolyzing activity (GTPase), as well as protein cross-linking. These functions imply two physiological functions: programmed cell life and death. These may have profound roles in the nervous system since studies in cultured astrocytes found tTG short (S) mRNA transcripts induced by treatment with injury-related cytokines. In the developing rat spinal cord, tTG activity is concentrated in ventral horn alpha motoneurons, but neither studies of spinal cord tTG gene expression, nor evaluation of the GTP-regulated isoforms in tissues, have been reported. We now report increased tTG protein and gene expression occurring rapidly after SCI. In parallel, novel appearance of a second, short form transcript, in addition to the normal long (L) isoform, occurs by 8 h of injury. Up-regulation of tTG message and activity following neural injury. with appearance of a truncated GTP-unregulated S form, may represent new approaches to drug targets in neurotrauma.

    Notes: Abstract: To investigate the regulation of posttranslational modifications of τ that might be pertinent to the production of the paired helical filament (PHF) of Alzheimer's disease, we incubated human neuroblastoma cells with the protein phosphatase inhibitor okadaic acid. This treatment results in increased immunoreactivity of τ with the monoclonal antibodies Alz-50, PHF-1, T3P, and NP8, a reduction in Tau-1 immunoreactivity, and an elevation in apparent molecular weight of τ. Moreover, our data demonstrate that accumulation of phosphates in τ leads to a decrease in the turnover rate of τ in the neuroblastoma cells. It is suggested that similar build-up of hyperphosphorylated τ in the neuronal perikarya may represent an early event in PHF formation. The present system facilitates the investigation of regulatory mechanisms governing the occurrence of PHF epitopes, their effects on neuronal cell metabolism, and possible pharmacological intervention.

    Notes: Abstract: Microtubules and their associated proteins play a prominent role in many physiological and morphological aspects of brain function. Abnormal deposition of the microtubule-associated proteins (MAPs), MAP2 and γ, is a prominent aspect of Alzheimer's disease. MAP2 and γ are heat-stable phosphoproteins subject to high rates of phosphorylation/dephosphorylation. The phosphorylation state of these proteins modulates their affinity for tubulin and thereby affects the structure of the neuronal cytoskeleton. The dinoflagellate toxin okadaic acid is a potent and specific inhibitor of protein phosphatases 1 and 2A. In cultured rat cortical neurons and a human neuroblastoma cell line (MSN), okadaic acid induces increased phosphorylation of MAP2 and γ concomitant with early changes in the neuronal cytoskeleton and ultimately leads to cell death. These results suggest that the diminished rate of MAP2 and γ dephosphorylation affects the stability of the neuronal cytoskeleton. The effect of okadaic acid was not restricted to neurons. Astrocytes stained with antibodies to glial fibrillary acidic protein (GFAP) showed increased GFAP staining and changes in astrocyte morphology from a flat shape to a stellate appearance with long processes.

    Notes: (Na1/2La1/2)(Mg1/3Nb2/3)O3 undergoes a series of phase transitions that involve cation order on the A- and B-sites of the parent perovskite structure. At high temperatures both sites contain a random distribution of cations; below 1275°C a 〈111〉 layering of Mg and Nb leads to the formation of a 1:2 ordered structure with a monoclinic supercell. A second transition was observed at 925°C, where the Na and La cations order onto alternate A-site positions along the 〈001〉 direction of the parent subcell. By quenching samples from above 1275°C to preserve the disorder on the B-site, a fourth variant of this compound was obtained by inducing A-site order through a subsequent anneal at 900°C. Although the changes in structure do not produce significant alterations in the relative permittivity (ɛr∼ 35), they do have a significant effect on the value of the temperature coefficient of the capacitance.

    Notes: The effect of the B-site cation chemistry and ordering on the dielectric properties of solid solutions in the (1−x)Pb(Mg1/3Nb2/3)O3–Pb(Sc1/2Nb1/2)O3 (PMN-PSN) perovskite system was examined in samples with 0.1 ≤x≤ 0.9. Thermal annealing treatments were effective in inducing long-range B-site order in the samples within this compositional range. The well-ordered, large chemical domain ceramics exhibit relaxor behavior up to x=∼0.5; for higher values of x, normal ferroelectric behavior was observed. For x≤ 0.5 reductions in the chemical domain, size had no significant effect on the weak-field dielectric properties, but induced a transition to relaxor behavior for x 〉 ∼0.6. The disordered PSN-rich samples undergo a spontaneous zero-field relaxor to ferroelectric transition similar to that reported previously for PSN. The field-dependent properties of compositions lying closest to the relaxor to ferroelectric crossover exhibited the highest sensitivity to alterations in the chemical order. The properties of this system are consistent with a “random site” description of the 1:1 ordered Pb(β′1/2β1/2)O3 structure with β′= (Mg(2−2x)/3Nb(1−x)/3Scx) and β″= Nb.

    Notes: Single-phase perovskites were formed in the (1−x)Ba(Zn1/3Nb2/3)O3-(x)La(Zn2/3Nb1/3)O3 system for compositions with 0.0≤x≤0.6. Although the stability of the trigonal “1:2” ordered structure of the Ba(Zn1/3Nb2/3)O3 end member is very limited (0.0≤x≤0.05), low levels of lanthanum induce a transformation to a cubic, “1:1” ordered structure that has a broad range of homogeneity (0.05≤x≤0.6). Samples with x 〉 0.6 were comprised of La3NbO7, ZnO, and a perovskite with x = 0.6. The cubic 1:1 phases were fully ordered and no evidence was found for a compositionally segregated microstructure. These observations could not be reconciled in terms of a “space-charge” model; rather, they supported a charge-balanced, “random-site” structure for the 1:1 cation-ordered Ba(β1/2′β1/2″)O3 phases.
