    Notes: Acid invertases play a key role in sugar metabolism, and the plant hormone abscisic acid (ABA) enhances sugar accumulation in crop sink organs, but information about the relationship between ABA and acid invertases has been limited. The present experiments were done with both in vivo pre-incubation of the grape (Vitis vinifera × V. labrusca L.) berry tissues in ABA-containing medium and in vivo infiltration of ABA into the intact berries. The results show that ABA activates both the soluble and cell wall-bound acid invertases during fruit development by enhancing their activities and amounts as assessed by immunoblotting or enzyme-linked immunosorbent assay. This activation was pH, time course and ABA dose dependent. The serine/threonine protein kinase inhibitors K252a, staurosporine and H7 and acid phosphatase increased the activation of ABA-induced acid invertase, but the tyrosine protein kinase inhibitor quercetin strongly suppressed the ABA-induced effects, suggesting that a complex reversible protein phosphorylation is involved in the ABA-induced activation of acid invertases. The effects of the protein kinase inhibitors were dependent on the in vivo state of the tissues but independent of the expression of acid invertases. Two ABA analogues, (–)-ABA and trans-ABA, had no effect on acid invertases, showing that the ABA-induced activation of acid invertases is specific to the physiologically active form of ABA. These data suggest that ABA may be involved in fruit development by activating acid invertases.

    Description: Background: Patients with chronic obstructive pulmonary disease (COPD) often have organ dysfunction and resulting poor quality of life; however, in China little information is available regarding factors that affect their health. Here, the relationship between risk factors, activities and psychological disorders and health of patients with COPD in rural areas of Xuzhou, China was assessed. Methods: A cross-sectional study of 7597 COPD patients randomly selected by place of residence from 24,641 COPD patients who had been identified by screening of the 1.10 million health records of all residents of the target area was carried out to evaluate the relationships between risk factors, activities, psychological disorders and the ADO index (age, dyspnea, and airflow obstruction). The participants were assessed by spirometry and by administering a newly designed face-to-face questionnaire, which included items on general factors, risk factors, activities and psychological disorders. Correlations between the ADO index and the items addressed by the questionnaire were calculated. Results: The mean score of the ADO index was 3.7 +/- 1.6. The ADO indices of current smokers, ex-smokers, and non-smokers were 3.9 +/- 2.1, 3.7 +/- 1.9, and 3.2 +/- 1.5, respectively (P 〈 0.001). The ADO indices of cooks and non-cooks were 4.0 +/- 2.2 and 3.5 +/- 1.7, respectively (P 〈 0.001). The correlation coefficient between self-assessment of health status and ADO index was 0.976 (P 〈 0.001). Only 5.7% of patients reported no limitation of their daily living activities. The correlation coefficient between daily living activities and ADO index was 0.981 (P 〈 0.001). Only 5.5% of patients reported no limitation of social activities. The correlation coefficient between social activities and ADO index was 0.989 (P 〈 0.001), between the assessed anxiety score and ADO index 0.972 (P 〈 0.001), and between the assessed depression score and ADO index 0.989 (P 〈 0.001). Conclusions: COPD severity was significantly correlated with behavior (especially regarding smoking and cooking with biofuel in confined spaces), physical strength, daily living activities, social activities, anxiety and depression. Comprehensive approaches are required for the prevention and treatment of COPD.

    Description: Although the mapping of spatiotemporal patterns of urban expansion has been widely studied, relatively little attention has been paid to detailed comparative studies on spatiotemporal patterns of urban growth at the regional level over a relatively longer timeframe. This paper was based on multi-sensor remote sensing image data and employs several landscape metrics and the centroid shift model to conduct a multi-angle quantitative analysis on urban expansion in Beijing, Tianjin and Tangshan (Jing-Jin-Tang) in the period from 1970–2013. In addition, the impact analysis of urban growth on land use was adopted in this research. The results showed that Beijing, Tianjin and Tangshan all experienced rapid urbanization, with an average annual urban growth rate of 7.28%, 3.9%, and 0.97%, respectively. Beijing has especially presented a single choropleth map pattern, whereas Tianjin and Tangshan have presented a double surface network pattern in orientation analysis. Furthermore, urban expansion in Beijing was mainly concentrated in Ring 4 to Ring 6 in the northwest and southeast directions, whereas the major expansion was observed in the southeast in Tianjin, primarily affected by dramatic development of Binhai New Area and Tianjin South Railway Station. Naturally, the urban expansion in Tangshan was significantly influenced by the expansion of Beijing and was primarily southwestward. The hot-zones of urbanization were observed within the ranges of 7–25 km, 6–18 km, and 0–15 km, accounting for 93.49%, 89.44% and 72.44% of the total expansion area in Beijing, Tianjin and Tangshan, respectively. The majority of the newly developed urban land was converted from cultivated land and integrated from other built-up land over the past four decades. Of all new urban land in the Beijing, Tianjin and Tangshan, more than 50% was converted from cultivated land, and there was a general tendency for smaller cities to have higher percentages of converted land, accounting for 50.84%, 51.19%, and 51.58%, respectively. The study revealed significant details of the temporal and spatial distributions of urban expansion in Beijing, Tianjin and Tangshan and provided scientific support for the collaborative development of the Beijing, Tianjin and Hebei (Jing-Jin-Ji) regions.

    Description: The relationship between calcium intake and colorectal cancer (CRC) risk remains inconclusive. We conducted this study to evaluate whether the association between calcium intake and CRC risk differs by anatomic subsite and determine the dose-response relationship for this association, as well as assess when in carcinogenesis calcium may play a role. We assessed calcium intake every 4 years and followed 88,509 women (1980-2012) in the Nurses' Health Study and 47,740 men (1986-2012) in the Health Professionals Follow-Up Study. We documented 3,078 incident CRC cases. Total calcium intake (≥1400 vs. 〈600 mg/d) was associated with a statistically significant lower risk of colon cancer (multivariable relative risk: 0.78, 95%CI: 0.65-0.95). Similar results were observed by different sources of calcium (from all foods or dairy products only). The inverse association was linear and suggestively stronger for distal colon cancer (0.65, 0.43-0.99) than for proximal colon cancer (0.94, 0.72-1.22, P - common effects =0.14). Additionally, when comparing different latencies, the overall pattern suggested that the inverse association appeared to be stronger with increasing latency and was strongest for intakes 12-16 years before diagnosis. Comparing total calcium intakes of ≥1400 vs. 〈600 mg/d for intake 12–16 y before diagnosis, the pooled RR (95% CIs) of CRC was 0.76 (0.64-0.91). Higher calcium intake was associated with a lower risk of developing colon cancer, especially for distal colon cancer. Overall inverse association was linear and did not differ by intake source. Additionally, calcium intake approximately 10 years before diagnosis appeared to be associated with a lower risk of CRC. This article is protected by copyright. All rights reserved.

    Description: Leucine-rich glioma inactivated 1 (LGI1) is a secreted protein that interacts with ADAM transmembrane proteins, and its mutations are linked to human epilepsy. The function of LGI1 in CNS development remains undefined. Here, we report novel functions of LGI1 in the generation of cerebellar granule precursors (CGPs) and differentiation of radial glial cells (RGCs) in the cerebellum. A reduction in external granule layer thickness and defects in foliation were seen in embryonic and new-born LGI1 knockout (KO) mice. BrdU staining showed an inhibited proliferation of CGPs in KO embryos, which might be explained by the reduced Sonic hedgehog in embryos. In addition, the differentiation of RGCs into Bergmann glias was suppressed in KO mice. Enhanced Jagged1-Notch1 signaling in KO mice via reduced β-secretase proteolysis suggests that altered phenotype of RGCs is due to abnormal Notch1 signaling.     Description: Despite the absence of any homologs of Tannerella forsythia KLIKK proteases in Tannerella sp.6_1_58FAA_CT1, the strain possesses a putative cysteine protease (G9S4N1) closely related to RgpB of Porphyromonas gingivalis . G9S4N1 lacks obvious propeptide that behaves as inhibitor of proteases and was proven to be a propeptide-independent protease. Unlike RgpB, which exclusively cleaves Arg Xaa bonds, G9S4N1 exhibits both arginine- and citrulline-specific activities. Mutations of Asp177, a potential P1-Arg binding site, to uncharged or positively charged residues did not alter the substrate specificity of G9S4N1 significantly. Moreover, a group of arginine-specific proteases from different species including porcine trypsin, bovine thrombin, and a trypsin-like serine protease of dengue 2 virus CF40-Gly-NS3pro185 also display different specificity toward citrulline residue, suggesting that citrulline-modified protein might have different roles and destiny in biological processes involving various proteases.

    Description: Introduction: Senior managers’ attitudes towards safety are very important regarding the safety practices in an organization. The study is to describe the current situation of senior managers′ attitudes towards safety in the Chinese coal industry. Method: We evaluated the changing trends as well as the reasons for these changes in the Chinese coal industry in 2009 and in 2014 with 168 senior manager samples from large Chinese state-owned coal enterprises. Evaluations of 15 safety concepts were performed by means of a questionnaire. Results and Conclusions: Results indicate that, in 2014, three concepts were at a very high level (mean > 4.5), and six were at a relatively high level (4.5 > mean > 4.0). Analyses of changing trends revealed that nine concepts improved significantly, while four greatly declined in 2014 compared to those in 2009. The data reported here suggest that the reasons for the significant improvement with respect to the nine concepts include the improvement in social and legal environments, the improvement of the culture of social safety, workers′ safety demands being met, and scientific and technical advances in the coal industry. The decline of the four concepts seemed to be caused by a poor awareness of managers in the coal industry that safety creates economic benefits, insufficient information on safety, inadequate attention to the development of a safety culture and safety management methods, and safety organizations and workers′ unions not playing their role effectively. Practical Applications: We therefore recommend strengthening the evidence that safety creates economic benefits, providing incentives for employees to encourage their participation in safety management, and paying more attention to the prevention of accidents in coal mines via safety organizations and unions. These results can provide guidelines for workers, industrialists, and government regarding occupational safety in the whole coal industry.

We previously found that nicotine inhibited 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) metabolism by CYP2A13, but its influence on other components of cigarette smoke remains unclear. The nicotine component of cigarette smoke extract (CSE) was separated, purified, and identified using high-performance liquid chromatography (HPLC) and ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS), splitting CSE into a nicotine section (CSE-N) and nicotine-free section (CSE-O). Cell viability and apoptosis by Cell Counting Kit-8 (CCK-8) and flow cytometry assays were conducted on immortalized human bronchial epithelial (BEAS-2B) cells stably expressing CYP2A13 (B-2A13) or vector (B-V), respectively. Interestingly, CSE and CSE-O were toxic to BEAS-2B cells whereas CSE-N showed less cytotoxicity. CSE-O was more toxic to B-2A13 cells than to B-V cells (IC50 of 2.49% vs. 7.06%), which was flatted by 8-methoxypsoralen (8-MOP), a CYP inhibitor. CSE-O rather than CSE or CSE-N increased apoptosis of B-2A13 cells rather than B-V cells. Accordingly, compared to CSE-N and CSE, CSE-O significantly changed the expression of three pairs of pro- and anti-apoptotic proteins, Bcl-2 Associated X Protein/B cell lymphoma-2 (Bax/Bcl-2), Cleaved Poly (Adenosine Diphosphate-Ribose) Polymerase/Poly (Adenosine Diphosphate-Ribose) Polymerase (C-PARP/PARP), and C-caspase-3/caspase-3, in B-2A13 cells. In addition, recombination of CSE-N and CSE-O (CSE-O/N) showed similar cytotoxicity and apoptosis to the original CSE. These results demonstrate that the nicotine component decreases the metabolic activation of CYP2A13 to CSE and aids in understanding the critical role of CYP2A13 in human respiratory diseases caused by cigarette smoking.

Three ensemble techniques, a simple average ensemble, a weighted average ensemble, and an ANN-based ensemble, were used to combine the outputs of the individual ANN models. The objective was to highlight the performance of the general regression neural network-based ensemble technique (GNE) through an improvement of monthly streamflow forecasting accuracy. Before the construction of an ANN model, data preanalysis techniques, such as empirical wavelet transform (EWT), were exploited to eliminate the oscillations of the streamflow series. Additionally, a theory of chaos phase space reconstruction was used to select the most relevant and important input variables for forecasting. The proposed GNE ensemble model has been applied for the mean monthly streamflow observation data from the Wudongde hydrological station in the Jinsha River Basin, China. Comparisons and analysis of this study have demonstrated that the denoised streamflow time series was less disordered and unsystematic than was suggested by the original time series according to chaos theory. Thus, EWT can be adopted as an effective data preanalysis technique for the prediction of monthly streamflow. Concurrently, the GNE performed better when compared with other ensemble techniques.
