    Beschreibung: To investigate transient dynamics of soil water redistribution during infiltration, we conducted horizontal borehole and surface ground penetrating radar measurements during a 4-day infiltration experiment at the rhizontron facility in Selhausen, Germany. Zero-offset ground penetrating radar profiling in horizontal boreholes was used to obtain soil water content information at specific depths (0.2, 0.4, 0.6, 0.8 and 1.2 m). However, horizontal borehole ground penetrating radar measurements do not provide accurate soil water content estimates of the top soil (0–0.1 m depth) because of interference between direct and critically refracted waves. Therefore, surface ground penetrating radar data were additionally acquired to estimate soil water content of the top soil. Due to the generation of electromagnetic waveguides in the top soil caused by infiltration, a strong dispersion in the ground penetrating radar data was observed in 500 MHz surface ground penetrating radar data. A dispersion inversion was thus performed with these surface ground penetrating radar data to obtain soil water content information for the top 0.1 m of the soil. By combining the complementary borehole and surface ground penetrating radar data, vertical soil water content profiles were obtained, which were used to investigate vertical soil water redistribution. Reasonable consistency was found between the ground penetrating radar results and independent soil water content data derived from time domain reflectometry measurements. Because of the improved spatial representativeness of the ground penetrating radar measurements, the soil water content profiles obtained by ground penetrating radar better matched the known water storage changes during the infiltration experiment. It was concluded that the combined use of borehole and surface ground penetrating radar data convincingly revealed spatiotemporal soil water content variation during infiltration. In addition, this setup allowed a better quantification of water storage, which is a prerequisite for future applications, where, for example, the soil hydraulic properties will be estimated from ground penetrating radar data.

    Notizen: Postsynaptic densities (PSDs) isolated from porcine cerebral cortices are large aggregates consisting of more than 30 different proteins. Inductively coupled plasma–mass spectrometric analyses revealed that isolated PSDs contained zinc at a concentration of 4.1 nmol per mg protein. Treatment with 8 m urea lead to dissociation of the PSDs into small components and, concomitantly, depletion of most of their bound zinc. After removal of the urea by dialysis, urea-dissociated PSD proteins did not reassemble into aggregates by themselves. Adding ZnCl2 to urea-treated PSD samples resulted in the assembly of urea-dissociated proteins into large aggregates with morphology and protein composition closely resembling those of the original PSDs. Mg2+, Ca2+, Co2+, Cd2+, Cu2+, Mn2+, Fe3+, K+ and Na+ ions at higher concentrations also induced the aggregation of urea-dissociated PSD protein. The structures of the K+-, Na+-, Mg2+- and Ca2+-induced aggregates were distinct from that of the original PSDs. Our results indicate that the structure of the PSD could be disassembled and reassembled under in vitro conditions. They further suggest that Zn2+ ions, by binding to certain zinc-binding proteins, play an important role in the formation and maintenance of the structure of the PSD.

    Notizen: A kinetic model is developed to study the process of phase separation that occurs when a polymer-dispersed liquid-crystal diffraction grating is produced by a process of photopolymerization. We find that the type of system produced depends sensitively on the rate of polymerization and on whether the spatially periodic illumination used to produce the grating is followed by a period of uniform illumination. While the average liquid-crystal concentration is always increased in the region of least irradiation, in some cases small inclusions of liquid-crystal form by spinodal decomposition at the minima in the irradiating intensity, while in others they form at the maxima. The simplest version of the theory combines the Flory-Huggins and Hillert models of phase separation. In some circumstances it must be augmented by including the effects of interdiffusion of polymer molecules having different degrees of polymerization. © 1996 American Institute of Physics.

    Notizen: A kinetic approach is used to examine the structure of the approximations that lead to the Flory–Huggins expression for the free energy of mixing of a polymer solution. The kinetic model that we study can be solved exactly in one dimension, and also on an infinite-dimensional lattice, in which case it reduces to the Flory–Huggins formula. An approximate solution that interpolates between these limits gives results for a three-dimensional lattice that differ rather little from those of the original Flory–Huggins theory. © 1996 American Institute of Physics.

    Notizen: Apomorphine, the catechol-derived dopamine D1/D2 receptor agonist, is currently in use as an antiparkinsonian drug. It has previously been reported that apomorphine was able to elicit expression of the enzyme tyrosine hydroxylase, a marker for DA neurons, in the fetal rat cerebrocortical cultures whilst in the presence of brain-derived neurotrophic factor. The present study demonstrated that treatment of fetal rat ventral mesencephalic cultures with apomorphine caused a marked increase in the number of dopaminergic neurons. The action of apomorphine can be mimicked by dopamine receptor (D1 and D2) agonists or blocked by preincubation with D1/D2 receptor antagonists. Incubation of recipient mesencephalic cultures with the conditioned medium derived from apomorphine-stimulated donor mesencephalic cultures elicited a 3.72-fold increase in the number of TH-positive neurons. Increased mRNA expression levels of brain-derived neurotrophic factor and glial cell line-derived neurotrophic factor were also found in the apomorphine-treated mesencephalic cells along with concomitant protein expression increases in the conditioned medium. Moreover, the trophic activity observed could be partially neutralized by antibodies against either brain-derived neurotrophic factor or glial cell line-derived neurotrophic factor. Cultured fetal striatal cells, but not hippocampal cells, also responded to apomorphine treatment. The membrane filtration studies revealed that both 〈30 kDa and 〉50 kDa fractions contained trophic activities. The latter characterization distinguishes them from most known neurotrophic factors. These results suggest that the apomorphine-modulated development of dopaminergic neurons may be mediated by activation of the dopamine receptor subtypes D1 and D2 thereby increasing the production of multiple growth factors.

    Notizen: Bystin has been identified as a protein which mediates cellular interactions between trophoblastic and endometrial epithelial cells by forming complexes with two partners, trophinin and tastin, during embryo implantation. However, the presence of bystin in the central nervous system has not been demonstrated. Here, we report the cloning of the full-length cDNA of the rat bystin gene from adult brain. Immunohistochemical and RT-PCR analysis showed that the levels of bystin expression were markedly up-regulated in the both 6-hydrodopamine-lesioned rat nigrostriatum and stab-lesioned cerebral cortex in adult rats. Double immunofluorescence staining revealed that most bystin-expressing glial cells were astrocytes (immature or mature). To determine the mechanisms for the up-regulation of bystin expression in glial cells, primary cultures of postnatal cortical astrocytes were employed. Western blot analysis showed that the expression of bystin was elevated by treatment with pro-inflammatory mediators lipopolysaccharide and interleukin-1β. Nerve growth factor known to be released after brain injury also induced bystin expression in the cultures. Exposure of astrocyte cultures to the differentiating agent forskolin resulted in up-regulation of bystin followed by a pronounced astrocytic stellation. The results suggest that the injury in the adult brain induces spatiotemporal up-regulation of bystin and it could be influenced, at least in part, by elevation of intracellular cAMP level. Bystin expressed by reactive astrocytes may be involved in their differentiation during the inflammatory processes following brain injury. The reappearance of bystin may also indicate that some reactive astrocytes have the capacity to recapitulate early developmental stages.

    Notizen: During neurodegenerative processes, cascades of degeneration and subsequent regeneration are triggered. However, the molecular nature of the factors involved in the neurodegeneration of the CNS remains largely unknown. In this study, the variations of protein expression in the striatum of adult Sprague–Dawley rats following 6-hydroxydopamine lesions were investigated, in order to better understand the molecular events occurring in the denervated target tissue. The rat striatum, ipsilateral to the lesion was analysed by two-dimensional gel electrophoresis followed by matrix assisted laser desorption/ionization-time of flight mass spectrometry. Seven proteins were up-regulated (188.1–750% compared to control) in response to the lesion: amyloid precursor-like protein 2 (APLP2), kininogen, glucokinase, tropomyosin α chain, type brain-1 and calpactin I light chain; whilst four proteins, neural epidermal growth factor-like 2, minichromosome maintenance 6, and thyroid hormone receptor β-2, were down-regulated (to between 36% and 59% of levels in sham-operated controls). Three proteins that did not match with available data in the SWISS-PROT protein database were also determined. Immunohistochemical analysis demonstrated colocalization of APLP2 and tyrosine hydroxylase in the nigral neurons. Moreover, reduction of APLP2-positive neurons in the substantia nigra pars compacta as well as the increases in the substantia nigra pars reticulata and in the striatum were observed. Furthermore, the conditioned medium of the Chinese hamster ovary cells over-expressing APLP2-751 (chondroitin sulphate-modified), but not APLP2-763 (nonchondroitin sulphate-modified), was able to increase the number of the tyrosine hydroxylase-positive neurons in fetal mesencephalic cultures. These results suggest that the expression of APLP2, a protein that has been thought to be associated with Alzheimer's disease, is up-regulated in the striatum following dopaminergic denervation. They also support the view that chondroitin sulphate-modified APLP2 protein may play an important role in the dopaminergic nigrostriatal system.
