    Description: Marine sediments host an unexpectedly large microbial biosphere, suggesting unique microbial mechanisms for surviving burial and slow metabolic turnover. Although dormancy is generally considered an important survival strategy, its specific role in subsurface sediments remains unclear. We quantified dormant bacterial endospores in 331 marine sediment samples from diverse depositional types and geographical origins. The abundance of endospores relative to vegetative cells increased with burial depth and endospores became dominant below 25 m, with an estimated population of 2.5 × 1028 to 1.9 × 1029 endospores in the uppermost kilometer of sediment and a corresponding biomass carbon of 4.6 to 35 Pg surpassing that of vegetative cells. Our data further identify distinct endospore subgroups with divergent resistance to burial and aging. Endospores may shape the deep biosphere by providing a core population for colonization of new habitats and/or through low-frequency germination to sustain slow growth in this environment.

    Description: A new linear accelerator system, called the SSC-Linac injector, is being designed at HIRFL (the heavy ion research facility of Lanzhou). As part of the SSC-Linac, the medium energy beam transport (MEBT) consists of seven magnetic quadrupoles, a re-buncher and a diagnose box. The total length of this segment is about 1.75 m. The beam dynamics simulation in MEBT has been studied using the TRACK 3D particle-in-cell code, and the simulation result shows that the beam accelerated from the radio frequency quadrupole (RFQ) matches well with the acceptance of the following drift tube linac (DTL) in both the transverse and longitudinal phase spaces, and that most of the particles can be captured by the final sector focusing cyclotron for further acceleration.     Description: A re-buncher with spiral arms for a heavy ion linear accelerator named as SSC-LINAC at HIRFL (the heavy ion research facility of Lanzhou) has been constructed. The re-buncher, which is used for beam longitudinal modulation and matching between the RFQ and DTL, is designed to be operated in continuous wave (CW) mode at the Medium-Energy Beam-Transport (MEBT) line to maintain the beam intensity and quality. Because of the longitudinal space limitation, the re-buncher has to be very compact and will be built with four gaps. We determined the key parameters of the re-buncher cavity from the simulations using Microwave Studio software, such as the resonant frequency, the quality factor Q and the shunt impedance. The detailed design of a 53.667 MHz spiral cavity and measurement results of its prototype will be presented.

    Description: Background: Hepatocellular carcinoma (HCC) exhibits strong intrinsic and acquired drug resistance which is the main obstacle to chemotherapy. Overexpression of ATP binding cassette (ABC) proteins correlates with activation of mitogen activated protein kinase (MAPK) pathway in HCC. Here, we systematically investigated the inhibition of MAPK pathway and its role in regulating HCC cell growth as well as ABC proteins MRP1 and MRP3 expression. Methods: The Raf1 kinase inhibitor (GW5074) and different MEK inhibitors (U0126 and AZD6244) were used to treat HCC cells to identify their effects on HCC cell growth and ABC proteins expression in vitro. Cell viability tests were performed after the treatment of MAPK pathway inhibitors and in combination with gemcitabine or doxorubicin. Western blot was applied to assess the changes of MAPK pathway and protein expression of MRP1 and MRP3. Flow cytometry was used to measure intracellular doxorubicin accumulation after the treatment of MEK inhibitors. Results: Both Raf1 inhibitor (GW5074) and MEK inhibitors (U0126 and AZD6244) suppressed HCC cell growth in a dose dependent manner. Pre-treatment of MEK inhibitor U0126 or AZD6244 sensitized HCC cells to gemcitabine or doxorubicin based chemotherapy. Raf1 inhibitor GW5074 had no effect on MRP1 and MRP3 protein expression. Treatment of gemcitabine or doxorubicin activated phosphorylated ERK and induced the upregulation of MRP1 and MRP3. MEK inhibitors U0126 and AZD6244 deactivated phosphorylated ERK, decreased endogenous MRP1 expression, reversed gemcitabine or doxorubicin induced MRP1 and MRP3 upregulation, and increased the intracellular doxorubicin accumulation. Conclusion: This study provides evidence that MEK inhibitors sensitize HCC cells to chemotherapy by increasing intracellular chemodrug accumulation. MEK inhibirors U0126 and AZD6244 reduced MRP1 as well as MRP3 expression, and may contribute partially to the sensitization. The combination of MEK inhibitor and conventional chemotherapy may offer new therapeutic option for the treatment of resistant HCC.

    Description: Background: B-cell lymphoma 6 (BCL6) protein, an evolutionarily conserved zinc finger transcription factor, showed to be highly expressed in various human cancers in addition to malignancies in the lymphoid system. This study investigated the role of BCL6 expression in breast cancer and its clinical significance in breast cancer patients. Methods: Expression of BCL6 protein was assessed using in situ hybridization and immunohistochemistry in 127 breast cancer patients and 50 patients with breast benign disease as well as in breast cell lines. Expression of BCL6 was restored or knocked down in two breast cancer cell lines (MCF-7 and T47D) using BCL6 cDNA and siRNA, respectively. The phenotypic change of these breast cancer cell lines was assessed using cell viability MTT, Transwell invasion, colony formation, and flow cytometry assays and in a xenograft mice model. Luciferase reporter gene, immunoblot, and qRT-PCR were used to investigate the molecular events after manipulated BCL6 expression in breast cancer cells. Results: BCL6 protein was highly expressed in breast cancer cell lines and tissue specimens and expression of BCL6 protein was associated with disease progression and poor survival of breast cancer patients. In vitro, the forced expression of BCL6 results in increased proliferation, anchorage-independent growth, migration, invasion and survival of breast cancer cell lines, whereas knockdown of BCL6 expression reduced these oncogenic properties of breast cancer cells. Moreover, forced expression of BCL6 increased tumor growth and invasiveness in a nude mouse xenograft model. At the gene level, BCL6 was a target gene of miR-339-5p. Expression of BCL6 induced expression of CXCR4 and cyclinD1 proteins. Conclusions: The current study demonstrated the oncogenic property of BCL6 in breast cancer and further study could target BCL6 as a novel potential therapeutic strategy for breast cancer.

    Description: A new compact accelerating structure named Hybrid RFQ is proposed to accelerate a high-intensity low-energy heavy ion beam in HISCL (High Intensive heavy ion SuperConducting Linear accelerator), which is an injector of HIAF (Heavy Ion Advanced Research Facility). It is combined by an alternative series of acceleration gaps and RFQ sections. The proposed structure has a high accelerating ability compared with a conventional RFQ and is more compact than traditional DTLs. A Hybrid RFQ is designed to accelerate 238 U 34+ from 0.38 MeV/u to 1.33 MeV/u. The operation frequency is described to be 81.25 MHz at CW (continuous wave) mode. The design beam current is 1.0 mA. The results of beam dynamics and RF simulation of the Hybrid RFQ show that the structure has a good performance at the energy range for ion acceleration.
