    Notes: Abstract. Planar asymmetric glycolipid/phospholipid bilayer membranes were used as a reconstitution model of the lipid matrix of the outer membrane of Gram-negative bacteria to study complement (C) activation by various bacterial surface glycolipids with the aim of defining the C activation pathway. As glycolipids the lipopolysaccharides of Salmonella enterica serovar Minnesota R mutant strains R595 (Re LPS) and R4 (Rd2 LPS), pentaacyl lipid A from the LPS of the Escherichia coli Re mutant F515, and glycosphingolipid GSL-1 of Sphingomonas paucimobilis IAM 12576 were used. Methylester and carboxyl-reduced derivatives of GSL-1 were used to elucidate the role of the carboxyl group as common functional group of LPS and GSL-1 for C activation. The formation of lytic pores was monitored via the measurement of changes in membrane current. For all glycolipids we observed a considerable increase in membrane current soon after addition of whole human serum due to the formation of lytic pores in the membranes. Pore formation was dependent on the presence of C9, indicating that the observed current changes were due to C activation. We found that in our reconstitution system of the outer membrane lipid A, Re LPS, and Rd2 LPS activated the classical pathway, the activation being independent of specific anti-LPS antibodies. In contrast, GSL-1 and the methylester derivative of GSL-1 activated the alternative pathway even at the low serum concentrations used in this study (about 0.2% v/v). Interestingly, the carboxyl reduced GSL-1 activated the classical pathway.

    Notes: Abstract. We have studied the interaction of the polycationic peptide antibiotic polymyxin B (PMB) with asymmetric planar bilayer membranes via electrical measurements. The bilayers were of different compositions, including those of the lipid matrices of the outer membranes of various species of Gram-negative bacteria. One leaflet, representing the bacterial inner leaflet, consisted of a phospholipid mixture (PL; phosphatidylethanolamine, -glycerol, and diphosphatidylglycerol in a molar ratio of 81:17:2). The other (outer) leaflet consisted either of lipopolysaccharide (LPS) from deep rough mutants of PMB-sensitive (Escherichia coli F515) or -resistant strains (Proteus mirabilis R45), glycosphingolipid (GSL-1) from Sphingomonas paucimobilis IAM 12576, or phospholipids (phosphatidylglycerol, diphytanoylphosphatidylcholine). In all membrane systems, the addition of PMB to the outer leaflet led to the induction of current fluctuations due to transient membrane lesions. The minimal PMB concentration required for the induction of the lesions and their size correlated with the charge of the lipid molecules. In the membrane system resembling the lipid matrix of a PMB-sensitive strain (F515 LPS/PL), the diameters of the lesions were large enough (d= 2.4 nm ± 8%) to allow PMB molecules to permeate (self-promoted transport), but in all other systems they were too small. A comparison of these phenomena with membrane effects induced by detergents (dodecyltriphenylphosphonium bromide, dodecyltrimethylammonium bromide, sodiumdodecylsulfate) revealed a detergent-like mechanism of the PMB-membrane interaction.

We have investigated the mechanism of action of the cationic antimicrobial protein (18 kDa) CAP18 on liposomes and monolayers made from phospholipids and enterobacterial lipopolysaccharides (LPS). CAP18 intercalates into lipid matrices composed of LPS from sensitive strains, weaker into those made of LPS from a resistant strain (Proteus mirabilis strain R45) or negatively charged phospholipids, but not into those composed of neutral phosphatidylcholine. From the combination of data obtained with fluorescence resonance energy transfer and Fourier-transform infrared spectroscopy and film balance measurements, it can be concluded that structural differences in the LPS determine the depth of intercalation of CAP18 into the respective lipid matrices. Thus, we identified the L-Arap4N linked to the first Kdo of the LPS of P. mirabilis strain R45 to be responsible for the CAP18 resistance of this strain. These data provide insight into CAP18-mediated effects on the integrity of the outer membrane of Gram-negative bacteria and led to an improved model for rabbit CAP18 membrane interaction.

    Notes: Abstract With its antiinflammatory and immunosuppressive properties interleukin-10 (IL-10) plays a dominant role in several immune reactions including regulatory mechanisms in the skin. The overexpression of this mediator has been reported in some inflammatory dermatoses as well as in various skin tumors. These observations are of importance since they may explain the limitation of hyperinflammatory conditions as in eczemas and erythemas on the one hand and the suppression of an adequate antitumor response and thereby the progression of malignant tumors on the other hand. Moreover, elevated IL-10 expression might contribute to an enhanced risk of development of microbacterial superinfections, a frequent finding in several dermatoses, and might also be involved in the pathogenesis of connective tissue diseases. In contrast, recent studies indicate a relative IL-10 deficiency in psoriasis. Early clinical data from psoriatic patients treated with recombinant human IL-10 suggest the therapeutic potential of this cytokine and underline its impact on the regulation of the skin immune system.

    Description / Table of Contents: Zusammenfassung 1. Die antimikrobielle Wirkung des Zinkoxid wurde in verschiedenen Zubereitungen von Externa in in vitro-Versuchen geprüft. 2. Für die Testung dienten folgende als Infektionserreger in Haut- und Schleimhautbereichen häufig anzutreffenden Keime: Streptococcus pyogenes, Micrococcus pyogenes, Pseudomonas aeruginosa sowie Candida albicans und Candida tropicalis. 3. Im Röhrchentest konnte, in dem in Aqua dest. aufgeschwemmten Zincum oxydatum crudum, übereinstimmend für alle Keimarten eine kurze Überlebensdauer festgestellt werden. Streptokokken und Staphylokokken hielten sich nur wenige Stunden, Bact. pyocyaneum und Candida bis zu 24 Std. Durch Zugabe von Nährbouillon wurde der Zinkeffekt bei Streptokokken und Staphylokokken kaum beeinflußt, bei Bact. pyocyaneum und Candida aufgehoben. 4. Bei der unter gleichen Voraussetzungen erfolgten Prüfung der Wirkung des ZnO auf den Stoffwechsel der Keime, gemessen am Sauerstoffverbrauch im Warburg-Apparat, war eine deutliche Reduktion der Atmung bei allen getesteten Stämmen — sowohl in der Ruheals auch in der Proliferationsphase — zu registrieren. 5. In den ZnO enthaltenden Externa war ein Absterben der Testkeime früher zu beobachten als in den übrigen Grundstoffen dieser Zubereitungen. 6. Mit dem nachgewiesenen Zinkeffekt wird ein Beitrag zu dem viel diskutierten Problem der Konservierungsstoffe in den Externa geleistet.
    Notes: Summary 1. The antimicrobic effect of zinkoxyde in various topical preparations was studied in vitro. 2. Common infectious agents for skin and mucosa such as streptococcus pyogenes, micrococcus pyogenes, pseudomonas aeruginosa as well as candida albicans and candida tropicalis were used as test germs. 3. In an aqueous suspension of zinkum oxydatum crudum a short survival time could be demonstrated for all germs. Streptococci and staphylococci survived for only a few hours, bacterium pyocyaneum and candida for up to 24 hrs. Addition of nutrient bouillon was of almost no influence on the zink effect on streptococci and staphylococci while it averted the effect of ZnO on bact. pyocyaneum and candida. 4. Using identical test concentrations ZnO caused a distinct reduction of respiration in the Warburg apparatus for all germs in the rest period as well as in the proliferation phase. 5. In ZnO containing topical preparation the germs died earlier as compared to preparations without ZnO. 6. Our study of the zink effect might render a contribution to the problem of preservatives in topical preparations.
