    Notes: Abstract The consecutive structural genes for the iron-sulfur flavoenzyme sulfide dehydrogenase, sudB and sudA, have been identified in the genome of Pyrococcus furiosus. The translated sequences encode a heterodimeric protein with an α-subunit, SudA, of 52598 Da and a β-subunit, SudB, of 30686 Da. The α-subunit carries a FAD, a putative nucleotide binding site for NADPH, and a [2Fe-2S]2+,+ prosthetic group. The latter exhibit EPR g-values, 2.035, 1.908, 1.786, and reduction potential, E m,8=+80 mV, reminiscent of Rieske-type clusters; however, comparative sequence analysis indicates that this cluster is coordinated by a novel motif of one Asp and three Cys ligands. The motif is not only found in the genome of hyperthermophilic archaea and hyperthermophilic bacteria, but also in that of mesophilic Treponema pallidum. The β-subunit of sulfide dehydrogenase contains another FAD, another putative binding site for NADPH, a [3Fe-4S]+,0 cluster, and a [4Fe-4S]2+,+ cluster. The 3Fe cluster has an unusually high reduction potential, E m,8=+230 mV. The reduced 4Fe cluster exhibits a complex EPR signal, presumably resulting from magnetic interaction of its S=1/2 spin with the S=2 spin of the reduced 3Fe cluster. The 4Fe cluster can be reduced with deazaflavin/EDTA/light but not with sodium dithionite; however, it is readily reduced with NADPH. SudA is highly homologous to KOD1-GOGAT (or KOD1-GltA), a single-gene encoded protein in Pyrococcus kodakaraensis, which has been putatively identified as hyperthermophilic glutamate synthase. However, P. furiosus sulfide dehydrogenase does not have glutamate synthase activity. SudB is highly homologous to HydG, the γ-subunit of P. furiosus NiFe hydrogenase. The latter enzyme also has sulfide dehydrogenase activity. The P. furiosus genome contains a second set of consecutive genes, sudY and sudX, with very high homology to the sudB and sudA genes, and possibly encoding a sulfide dehydrogenase isoenzyme. Each subunit of sulfide dehydrogenase is a primary structural paradigm for a different class of iron-sulfur flavoproteins.

    Description / Table of Contents: Samenvatting De iepen langs de grachten in de stad Amsterdam worden periodiek aangetast door ringelrupsen (Malacosoma neustrium). Jaarlijks worden de bomen bestoven met een Derris-stuifmengsel. Deze bestrijding heeft nadelen, daar Derrispoeder giftig is voor vissen, nuttige insekten en andere levende organismen. Vergelijkende proeven in het laboratorium met Derris-stuifmengsel en preparaten op basis vanBacillus thuringiensis hadden tot resultaat, dat bij de gegeven concentraties de bacterie-poeders een iets betere werkzaamheid vertoonden dan Derris (tabel 1). Daar deze poeders bovendien geen gevaar opleveren voor mensen en huisdieren, en zelfs tegenover insekten zeer specifiek werken, verdient het gebruik van deze poeders de voorkeur boven dat van Derris. Vooral uit een oogpunt van natuurbescherming zijn in deze omstandigheden de preparaten, gebaseerd opBacillus thuringiensis, de beste bestrijding van deze insektenplaag in een grote stad. Proeven in de straten van Amsterdam metBacillus thuringiensis-poeder zijn tevens reeds enige jaren genomen. Zij worden in de komende jaren op grotere schaal voortgezet. Dit onderzoek geschiedde onder aspiciën van de „Werkgroep voor Harmonische Insektenbestrijding T.N.O.”
    Notes: Summary Laboratory experiments comparing the effect of dusts of Derris powder with dusts of products, based onBacillus thuringiensis, indicated a higher toxicity to Tent Caterpillars (Malacosoma neustrium) of the latter group. As, moreover, these dusts are harmless to man and higher animals, and even have a very specific action to insects, the use of the products is to be preferred above the use of Derris dust. The bacterial dusts are already used for some time in outdoor experiments, and it is proposed to use these dusts on a large scale in coming years. The insecticides are killing through their specific action the nuisant caterpillars only, and are therefore appreciable also because of recreation and nature conservation.

    Description / Table of Contents: Samenvatting Onbehandelde rupsen vanEphestia kuehniella andPieris brassicae werden samengebracht met een aantal rupsen van dezelfde soort, die kort tevoren anaal, resp. oraal geïnfecteerd waren met een suspensie vanB. thuringiensis. Van de laatste categorie stierven de meeste na enkele dagen. Ofschoon de gezonde, zieke en dode rupsen daarna nog enige weken werden samengehouden, trad er onder de onbehandelde rupsen praktisch geen sterfte op, die veroorzaakt was door bacteriën.
    Notes: Abstract A number of untreated larvae ofEphestia kuehniella andPieris brassicae were confined with some larvae of the same species that had recently been infected withBacillus thuringiensis. The majority of the last category died within a few days. Although healthy, diseased and dead specimens were kept together for several days, scarcely any larva of the healthy group became diseased or died by the action of the bacterium. It was concluded thatB. thuringiensis does not spread easily.
