    Description: Ion channels have played a substantial role in the evolution of novel traits across all of the domains of life. A fascinating example of a novel adaptation is the convergent evolution of electric organs in the Mormyroid and Gymnotiform electric fishes. The regulated currents that flow through ion channels directly generate the electrical signals which have evolved in these fish. Here, we investigated how the expression evolution of two sodium channel paralogs (Scn4aa and Scn4ab) influenced their convergent molecular evolution following the teleost-specific whole-genome duplication. We developed a reliable assay to accurately measure the expression stoichiometry of these genes and used this technique to analyze relative expression of the duplicate genes in a phylogenetic context. We found that before a major shift in expression from skeletal muscle and neofunctionalization in the muscle-derived electric organ, Scn4aa was first downregulated in the ancestors of both electric lineages. This indicates that underlying the convergent evolution of this gene, there was a greater propensity toward neofunctionalization due to its decreased expression relative to its paralog Scn4ab. We investigated another derived muscle tissue, the sonic organ of Porichthys notatus , and show that, as in the electric fishes, Scn4aa again shows a radical shift in expression away from the ancestral muscle cells into the evolutionarily novel muscle-derived tissue. This study presents evidence that expression downregulation facilitates neofunctionalization after gene duplication, a pattern that may often set the stage for novel trait evolution after gene duplication.

    Description: The conserved RNA-binding protein Musashi1 (MSI1) has emerged as a key oncogenic factor in numerous solid tumors, including glioblastoma. However, its mechanism of action has not yet been established comprehensively. To identify its target genes comprehensively and determine the main routes by which it influences glioblastoma phenotypes, we conducted individual-nucleotide resolution cross-linking and immunoprecipitation (iCLIP) experiments. We confirmed that MSI1 has a preference for UAG sequences contained in a particular structural context, especially in 3' untranslated regions. Although numerous binding sites were also identified in intronic sequences, our RNA transcriptome sequencing analysis does not favor the idea that MSI1 is a major regulator of splicing in glioblastoma cells. MSI1 target mRNAs encode proteins that function in multiple pathways of cell proliferation and cell adhesion. Since these associations indicate potentially new roles for MSI1, we investigated its impact on glioblastoma cell adhesion, morphology, migration, and invasion. These processes are known to underpin the spread and relapse of glioblastoma, in contrast to other tumors where metastasis is the main driver of recurrence and progression.

    Description: Regional assessments of trends in climate extremes are necessary for countries to make informed decisions about adaptation strategies and to put these changes into a global context. A workshop bringing together several Southeast Asian countries has delivered a new set of daily weather observations suitable to analyse the changes in temperature and precipitation extremes between 1972 and 2010. The use of a consistent and widely tested methodology in this study allows a direct comparison with results from other parts of the world. Trends in a range of climate extremes indices were assessed focusing on spatial variation in these trends. For most locations temperature trends obtained in this study appear broadly consistent with previous assessments; for some locations stronger trends have been detected through the inclusion of new data. In contrast to earlier studies, evidence of trends in precipitation extremes is emerging, with significant increasing trends in both regional and subregional data. In addition, large correlations between regional rainfall extremes and large-scale features such as El Niño-Southern Oscillation and the Indian Ocean Dipole were identified. Finally, the observed trends are compared with a regional climate model reconstruction of the historical period. It was found that the model captures very well the trends and spatial variation of temperature extremes across the region, albeit with an underestimation of the more extreme indices. In contrast, the trends in precipitation extremes are largely overestimated, particularly in the western side of Southeast Asia.     Description: Three spatial structure groups of radionuclides in U and Th series, 210Pb-excess and 137Cs, and 40K were found based on analyzing temporal and spatial datum of their content by factor analysis with oblique rotation in Nhatrang bay.U and Th spatial structure with their contours decreased toward the offshore, ran longshore and divided seawater of bay into two parts with strong gradient on both sides. Inside part located from center of Nhatrang bay toward the seashore with three main deposit centers of their contents higher than 23 Bq/kg.dry for 238U and 40 Bq/kg.dry for 232Th, indicated unstability of shoreline. Almost sediments coming from river extended toward the offshore, were stopped and transported toward southeastern. The outside part was less than abovementioned content. The boundary line between two parts superposed with the constantly limit line of turbid plume in the rainy season. Direct influence of the continental runoff was limited by the 9 Bq/kg.dry contour of 238U, 19 Bq/kg.dry contour of 232Th. Longshore current was a predominant process whereas lateral transport as sifting and winnowing process of finer grains in sediments of Nhatrang bay. Areas that had very low content of 137Cs and 210Pb-excess adjoining shoreline showed areas being eroded. Accumulation of 137Cs and 210 Pbexcess nearby river mouth characterized for fine compositions of sediments controlled by seasonal plumes and sites further toward the south indicated finer materials transported from river and accumulated in lack of hydrodynamic process. Near shore accumulation of 40K revealed the sediments there originated from bed erosion.
