    Notes: Abstract: Rat brain opioid receptors were solubilized with digitonin and a zwitterionic detergent, 3-[(3-cholamido-propyl)-dimethylammonio]-1-propanesulfonate (CHAPS). The yield of solubilization was 70–75% with digitonin and 30–35% with CHAPS. Kinetic and equilibrium studies performed from digitonin extracts resulted in KD values comparable with those of the membrane fractions. Two [3H]naloxone binding sites were obtained in the extracts similarly to membrane fractions. The rank order potency of drugs used in the competition experiments did not change during solubilization. The distributions of μ, δ, and κ opioid receptor binding sites were similar in membrane and digitonin-solubilized fractions (48–50%μ, 35–37%κ, and 13–17%δ subtypes). The hydrodynamic properties of digitonin- and CHAPS-solubilized preparations were studied by sucrose density gradient centrifugation and Sepharose-6B chromatography. In all cases, two receptor populations were identified with the following parameters: sedimentation coefficients for the digitonin extracts were 9.2S and 13.2S and for CHAPS extract 8S and 15.6S; the Stokes radii were 45Å and 65Å for the digitonin extract and 31Å and 76Å for the CHAPS-solubilized preparation.

    Notes: Ca2+/calmodulin dependent protein kinase (CaMPK) II is a key enzyme in many physiological processes. The enzyme is inactive unless Ca2+/CaM binds to it. In this inactive form CaMPK-II does not bind ATP suggesting that the ATP-binding domain is involved in an intramolecular interaction. We show here that F12, a 12 amino acid long peptide fragment of the ATP-binding domain (CaMPK-II23–34, GAFSVVRRCVKV) can inhibit the Ca2+/CaM-dependent activity (IC50 of 3 µm) but has no effect on the Ca2+/CaM-independent activity of CaMPK-II. Kinetic analysis exhibited mixed inhibition with respect to autocamtide-2 and ATP. The inhibition by F12 showed specificity towards CaMPK-II, but also inhibited CaMPK-I (IC50 = 12.5 µm), while CaMPK-IV (IC50 = 85 µm) was inhibited poorly and cAMP-dependent protein kinase (PKA) was not inhibited. Substitution of phenylalanine at position 25 to alanine (A12), had little effect on the inhibition of different Ca2+/CaM-dependent protein kinases, suggesting that phenylalanine 25 does not play a crucial role in the interactions involving F12. Thus the molecular interactions involving the ATP-binding domain appears to play a role in the regulation of nonphosphorylated CaMPK-II activity.

    Notes: An increasing incidence of cutaneous malignant melanoma had been observed in the Netherlands.Therefore, between 1989 and 1992 a preventive skin cancer campaign was organized in part of western Netherlands. The effect of this screening campaign has yet to be evaluated by investigating the distribution of all in situ and invasive primary cutaneous melanoma according to anatomical subsite, tumour thickness, and histological type in a population-based study during 1980–92. A total number of 2236 (1361 female; 875 male) invasive and 580 (395 female; 185 male) in situ melanomas were diagnosed. The median age of patients with thin lesions was 46 years, intermediate lesions 54 years, and thick lesions 65 years, which may be indicative of slow tumour progression. The median thickness of the invasive lesions was 0·9 mm for women and 1·1 mm for men and the invasive to in situ ratio was 3·4 for female and 4·7 for male subjects, suggesting a reasonable awareness of melanoma in this population. The relative tumour density (RTD) was remarkably high for melanomas in the head/neck area for both sexes and invasive melanoma on the male trunk and the lower legs of females. The low RTD of invasive melanomas on the lower limbs of men and the high density in the head/neck area for both sexes are remarkable and support the aetiological role of ultraviolet exposure. It is concluded that our findings are in many ways similar to those of other descriptive studies.

    Notes: A number of new synthetic nociceptin ligands were studied in receptor binding and functional tests in rat brain membranes and in cloned systems. Ligand binding experiments were performed with three different radioprobes developed in our lab. The nociceptin derivatives exhibited high affinity in competition experiments. Receptor-mediated G-protein activation was determined in [35S]GTPgS binding assays. Among the new structures examined, Ac-RYYRIK-ol was found to be only a weak stimulator by itself, whereas this compound inhibited receptor-mediated G-protein activation.     Notes: Experimental studies are presented of a binary wire based on the quantum-dot cellular automata computational paradigm. The binary wire consists of capacitively coupled double-dot cells charged with single electrons. The polarization switch caused by an applied input signal in one cell leads to the change in polarization of the adjacent cell and so on down the line, as in falling dominos. Wire polarization was measured using single islands as electrometers. Experimental results are in very good agreement with the theory and confirm there are no metastable states in the wire. © 1999 American Institute of Physics.

    Notes: The influenza virus haemagglutinin has an important role in the infectious cycle of the virus and carries multiple B and T cell epitopes. It is synthesized as a single polypeptide chain but viral infectivity depends on its post-translational enzymatic cleavage. The cleavage site of a trypsin-like enzyme responsible for this modification is found in the most conserved intersubunit region of the molecule. In this study the role of this region in antibody recognition was investigated. Synthetic peptides comprising the intact and cleaved forms of the intersubunit segment were used to examine the specificity of virus- or peptide- induced antibodies. The immune response elicited by viral infection resulted in the appearance of antibodies capable of neutralizing the virus without interfering with its binding to the receptor. A monoclonal antibody (MoAb) of such functional properties was shown to recognize the intact intersubunit region both in the uncleaved haemagglutinin molecule and in a 25-mer synthetic peptide comprising the intact intersubunit region. Specificity and functional studies revealed the conformation dependent recognition of the C-terminal segment of the haemagglutinin 1 subunit by this MoAb. The binding of the antibody was shown to inhibit the trypsin-mediated cleavage of the haemagglutinin molecule and the membrane fusion event. The enzymatic cleavage of the haemagglutinin was demonstrated to abolish antibody recognition of the infective virus suggesting an escape mechanism mediated by the functional destruction of this highly conserved region. The synthetic peptide corresponding to the intact intersubunit region is characterized by an ordered structure and is able to elicit an antibody response in BALB/c mice while its subfragments are nonimmunogenic. Furthermore, this peptide elicited a protective immune response demonstrated by in vivo experiments.

    Notes: A 62-year-old female patient is described who developed toxic epidermal necrolysis (TEN) after medication with phenytoin and oxazepam. Initially phenytoin was discontinued and dexamethasone pulse therapy (1.5 mg/kg on 3 consecutive days) was initiated on the tenth day of skin disease. This resulted in clinical improvement. Histologically re-epithelialization could be demonstrated below the necrotic epidermis. However, on the eighteenth day of skin disease (10 days after discontinuation of phenytoin and 8 days after the start of dexamethasone pulse therapy), a histologically verified rebound-TEN developed with a detachment of 95%. Oxazepam was stopped and a second series of dexamethasone pulse therapy was given. Re-epithelialization began within 24 h of the start of the second series of dexamethasone pulse therapy, and continued to almost complete recovery within 1 week.

    Notes: Sperm motility variables from the milt of the common carp Cyprinus carpio were assessed using a computer-assisted sperm analysis (CASA) system across several months (March-August 1992) known to encompass the natural spawning period. Two-year-old pond-raised males obtained each month were primed with an exogeneous hormone injection of carp pituitary extract and stripped of milt 18–24h later. The milt was diluted, activated and videotaped using a high-speed (200 Hz) videocamera and recorder. Videotaped samples were subsequently analysed using the CellTrak/S CASA system (Motion Analysis Corp.) for percent motility, curvilinear and straight-line velocity. In addition to assessing changes in motility parameters across several months, a comparison was made between two predilution/ activation media combinations (homologous seminal plasma/NaCl+HEPES v. 2-h incubation in 200 mM KCl+Tris/Tris). The percentage of motile cells assessed immediately after activation was significantly greater in the summer months (May, June, July) when compared to a spring sampling point (March); when assessed 1 min after activation, cells prediluted in seminal plasma maintained a higher percentage of motility than those prediluted in KC1. Curvilinear and straight-line velocities exhibited a slight seasonal trend; variations in response to the predilution treatments were observed with these measurements. Sperm count gradually increased through April and May (9–63 × 109 to 2–38 × lO10ml– 1 milt), declined in June and July (to 1–83 × lO10ml– 1 milt), and was followed by a steep increase in August (2–74 × 1010 ml– 1 milt). Mean seminal plasma osmolality remained relatively constant (250–265 mOsmol kg – 1) throughout the sampling period.
