    Notes: Skin-prick experiments with different concentrations of histamine (0.1 mg/ml, 1.0 mg/ml, 10.0 mg/ml) and isotonic NaCl solution for control were performed on the forearms of 10 healthy volunteers. Regular recordings of the cutaneous blood flow by laser Doppler flowmetry were performed in the centre of the weals and 5, 10, 15 and 20 mm from the centre representing the zone of perilesional flare. In the weal centre the blood flow was increased but neither different concentrations of histamine nor the control reaction could be differentiated. Recordings performed 5 and 10 mm from the centre showed, however, significant differences between the different solutions of histamine tested as well as the control reaction. The better differentiation was obtained 5 and 10 min after skin-prick. In conclusion, laser Doppler flowmetry with recordings of the perilesional flare is found useful for quantification of weal reactions.

    Notes: Skin biopsies from five healthy subjects, taken before and after UVB irradiation, were examined using immunohistological techniques for the cytokines interleukin-1 (IL-1) and tumour necrosis factor (TNF). Using polyclonat specific antibodies against IL-1 and TNF, the two cytokines appeared identically located on the epidermal cell membranes of the stratum granulosum and stratum spinosum in unexposed skin. After UVB-exposure, the staining intensity for both IL-1/epidermal cell derived thymocyte-activating factor (ETAF) and TNF was markedly increased, and the epidermal staining included the basal cell layer.Immunohistological investigation of tissue-bound epidermal cytokines may be valuable in the study of skin diseases.

    Notes: The elastosis-inducing capacity of UV-A and UV-B was studied in the skin of hairless mice. After irradiation with UV-B and UV-A applied either simultaneously, separately or sequentially, the degree of elastosis in the dermis was microscopically evaluated. This semi-quantitative method showed no significant elastosis in the UV-A irradiated mice, and moderate elastosis in the UV-B irradiated mice. Heavy elastosis was observed when the mice were exposed sequentially to UV-B and a large dose of UV-A, but if a moderate dose of UV-A was given simultaneously with UV-B, the degree of elastosis was slightly reduced compared with the elastosis induced by UV-B alone.

    Notes: Summary In eight healthy volunteers the effect of PUVA therapy on skin vessel leakiness was examined (a quadrant on one side of the abdominal skin was shielded during the UVA irradiation). The relative concentration of intravenously injected labeled albumin in suction blister fluid from irradiated skin was only slightly elevated 2 days after PUVA treatment, whereas a significant elevation (P〈0.01) was observed on non-irradiated skin. No significant changes were observed in the concentrations of four endogenous plasma proteins in serum and blister fluid. It is concluded that PUVA therapy can induce an increased vascular leakage of albumin from the skin vessels due to humoral factors. In exposed — in contrast to non-exposed — skin, the findings are influenced by an interstitial oedema.
