    Notes: Summary A case of pituitary apoplexy occurring after Gd-DTPA-administration for contrast enhanced MRI in a patient with an hGH-producing macro-adenoma is presented. Within days the initially increased hGH level fell to the normal range, the oral glucose tolerance test (OGTT) showed a normal suppression of hGH and complete anterior pituitary insufficiency developed. At this time repeated MRI suggested a haemorrhagic infarction of the macroadenoma. Fourteen months later re-examination confirmed spontaneous cure of the acromegaly, improvement of adenopituitary function and shrinkage of the sellar content. The causal linkage between the pituitary adenoma apoplexy and Gd-DTPA-administration is unclear. It might be due to contrast induced blood pressure and endothelial permeability changes, possibly promoted by pre-existing diabetes mellitus associated vasculopathy.

    Notes: Abstract Background: In this study we tried to estimate the local surgical trauma in patients undergoing endoscopic or conventional hernia repair via the changes in peripheral blood T cell subpopulations (i.e., T-helper 1 (TH1) and TH2 cells), recently shown to be recruited differentially to inflammatory sites. Methods: Cells were identified flow-cytometrically by intracellular cytokine staining on a single cell level in 30 patients undergoing conventional (Shouldice) or total extraperitoneal patch (TEPP) hernia repair. Results: The TH1 cells decreased postoperatively in Shouldice patients on an average of 20.8–31.4%, whereas in TEPP patients only a minor decline (mean, 7.8–9.2%) was observed. The TH2 cells did not change significantly in TEPP patients, and a small increase (mean, 7.7%) was detected in Shouldice patients. Conclusions: Our results suggest that the postoperative reduction in TH1 cells reflects local surgical trauma and can be helpful in evaluating different surgical procedures. When conventional and endoscopic hernia repair were compared, the latter proved less traumatizing.

    Notes: Summary The synthesis and secretion of the toxic exoprotein α-haemolysin of E. coli PM152 is coded by the transmissible plasmid pHly152 (41×106 dalton) as shown by the transformation of the plasmid DNA and the isolation of mutants that are specifically altered in the synthesis and transport of haemolysin. These mutants were obtained by chemical mutagenesis and insertion of the ampicillin transposon (Tn3) into pHly152. Tn3 transposition was also used for the identification and the location of the cistrons on pHly152 essential for haemolysis. The EcoRI and HindIII fragments of the haemolytic plasmid pHly152 were cloned and used for the complementation of the haemolysis negative Tn3 insertion mutants. A DNA segment of 3.2×106 dalton could be thus identified which consists of at least three clustered cistrons necessary for haemolysis. Two of these cistrons are required for the formation of active haemolysin. At least one other cistron seems to be involved in the secretion of active haemolysin through the outer membrane of E. coli. The gene products determined by these cistrons were identified in minicells of E. coli. Their molecular properties were determined and their possible function in the formation and secretion of haemolysin will be discussed.

    Notes: Summary The tissue origin, growth patterns and organization ofin vitro tissue cultures initiated from immature maize (Zea mays L.) embryos were cytologically examined. Examination of thick sections from initiating cultures indicated that the scutellum is the tissue of origin. During culture initiation the scutellum, especially in the region of the epithelium, becomes highly meristematic and proliferates rapidly. These cultures were characterized by a cambial zone which provided for extendedin vitro proliferation and by apical meristems developing on the culture surface. These meristems developed to the two leaf primordia stage under the cultural conditions of this study. Differentiation of root primordia associated with the apical meristems was not observed. The apical meristems appeared to arisede novo at the tissue surface and were probably multicellular in origin. The observations are discussed in relation to their significance concerning mutation and selection.

    Description / Table of Contents: Zusammenfassung Leuchtbakterienteste stellen eine elegante Methode zur schnellen, reproduzierbaren Bestimmung bakterientoxischer Effekte dar. Einer allgemeinen Anwendung steht allerdings entgegen, daß die Luminescenzeigenschaften auf relativ wenige Bakterienfamilien mit zum Teil extremen Standortansprüchen beschränkt sind. Die Übertragbarkeit der Meßergebnisse auf interessierende Anwendungen ist deshalb in vielen Fällen nicht gegeben. In der vorliegenden Arbeit wird am Beispiel eines gentechnologisch konstruierten luminescenten Abwasserstammes gezeigt, daß durch Verwendung von anwendungsspezifischen Testorganismen das Anwendungsspektrum von Leuchtbakterientesten erweitert werden kann.
    Notes: Summary Luminescent bacteria bioassays represent an elegant method for fast and reproducible determination of microbial toxicity. A general application of this method is however prevented by the fact that the capability for bioluminescence is restricted to a relatively small group of bacteria with extreme environmental demands. Therefore it seems questionable whether results obtained with those organisms can be used for all interesting applications. It is shown in this paper, that the field of applications for bacterial luminescent assays can be widened using gentechnologically constructed luminescent bacteria.

    Description: Trans-activation element DNA-binding protein of 43 kDa (TDP-43) characterizes insoluble protein aggregates in distinct subtypes of frontotemporal lobar degeneration and amyotrophic lateral sclerosis. TDP-43 mediates many RNA processing steps within distinct protein complexes. Here we identify novel TDP-43 protein interactors found in a yeast two-hybrid screen using an adult human brain cDNA library. We confirmed the TDP-43 interaction of seven hits by co-immunoprecipitation and assessed their co-localization in HEK293E cells. As pathological TDP-43 is ubiquitinated, we focused on the ubiquitin-conjugating enzyme UBE2E3 and the ubiquitin isopeptidase Y (UBPY). When cells were treated with proteasome inhibitor, ubiquitinated and insoluble TDP-43 species accumulated. All three UBE2E family members could enhance the ubiquitination of TDP-43, whereas catalytically inactive UBE2E3C145S was much less efficient. Conversely, silencing of UBE2E3 reduced TDP-43 ubiquitination. We examined 15 of the 48 known disease-associated TDP-43 mutants and found that one was excessively ubiquitinated. This strong TDP-43K263E ubiquitination was further enhanced by proteasomal inhibition as well as UBE2E3 expression. Conversely, UBE2E3 silencing and expression of UBPY reduced TDP-43K263E ubiquitination. Moreover, wild-type but not active site mutant UBPY reduced ubiquitination of TDP-43 C-terminal fragments and of a nuclear import-impaired mutant. In Drosophila melanogaster, UBPY silencing enhanced neurodegenerative TDP-43 phenotypes and the accumulation of insoluble high molecular weight TDP-43 and ubiquitin species. Thus, UBE2E3 and UBPY participate in the regulation of TDP-43 ubiquitination, solubility, and neurodegeneration.
