    Notes: Summary A study was conducted to analyse the causes of death during the period January 1985 to December 1989, in pashmina kids and goats thought to be acclimatised to a medium altitude climate. In general, mortality was higher during the rainy season (up to 62 percent) than in the winter (17 percent) and summer seasons (21 percent). An exception was the year 1987 when mortality was lowest in the rainy season, presumably due to the effects of a drought. During the rainy season the majority of 6–12 month-old kids became dull, off-feed, weak, emaciated and anaemic. Occasionally these animals showed nasal discharge and diarrhoea. Haematological examination of randomly selected animals during rainy, post-rainy and early-winter seasons in 1985, 1986 and 1989 showed very low red blood cell counts (RBC), haemoglobin (Hb) and packed cell volume (PCV); with elevated erythrocytic sedimentation rates (ESR), neutrophilia, relative lymphopaenia, eosinopaenia, hypoproteinaemia, hypoalbuminaemia, hypogamaglobinaemia, increased A:G ratio and hypoglycemia. Red blood cells showed piokilocytosis, anisocytosis, punched out or ring shapes, presence of erythroblasts, basophilic stippling and Anaplasma marginale.Post-mortem examination revealed lesions of parasites either singly or in combination. These included lice, coccidia, Moniezia expansa, Haemonchus contortus,and Trichuris ovis.Poor or slightly improved health state, anaemic blood picture and moderated mortality were witnessed 45 days post-treatment or during the post-rainy season. Early-winter studies indicated that the goats were in an excellent health condition with normal blood picture and negligible mortality. It was concluded that the higher mortality in pashmina goats during the rainy season was association with high rainfall, humidity, partial or zero grazing, confinement of goats in sheds (all environmental stresses) and parasitism. This was supported in 1987, when the lowest mortality occurred during the rainy season due to the effect of drought and the absence of environmental stresses.

    Description: Autonomic dysreflexia (AD), which describes episodic hypertension, is highly prevalent in people with spinal cord injury (SCI). In non-SCI, primary hypertension depresses cardiac contractile reserve via β-adrenergic mechanisms. In this study, we investigated whether AD contributes to the impairment in cardiac contractile function that accompanies SCI. We induced SCI in rodents and stratified them into sham, SCI, or SCI plus repetitive induction of AD. At 6-week post-SCI, we assessed cardiac function using in vivo (speckle-tracking echocardiography), ex vivo (working heart), and molecular approaches (Western blot). We also provide unique translational insight by comparing the relationship between the number of daily AD events and cardiac function in 14 individuals with cervical SCI. We found SCI and SCI plus repetitive induction of AD exhibited a reduction in left ventricular dimensions at 6-week post-SCI versus preinjury ( P 〈0.049). Compared with sham, SCI exhibited a reduction in peak radial strain along with a down and rightward shift in the Starling curve ( P 〈0.037), both of which were further depressed in SCI plus repetitive induction of AD ( P 〈0.042). In response to β-adrenergic stimulation, SCI plus repetitive induction of AD exhibited an attenuated increase in contractile indices ( P 〈0.001), despite no differences in β-receptor expression within the left ventricle. Our clinical data confirm our experimental findings by demonstrating significant associations between the number of daily AD events and markers of systolic and diastolic function along with left ventricular mechanics. Here, we provide the first evidence from a translational perspective that AD exerts insidious effects on cardiac function in rodents and humans with SCI.

    Description: A chemically diverse family of small-molecule signals, the ascarosides, control developmental diapause (dauer), olfactory learning, and social behaviors of the nematode model organism, Caenorhabditis elegans. The ascarosides act upstream of conserved signaling pathways, including the insulin, TGF-β, serotonin, and guanylyl cyclase pathways; however, the sensory processes underlying ascaroside function are poorly understood. Because ascarosides often are multifunctional and show strongly synergistic effects, characterization of their receptors will be essential for understanding ascaroside biology and may provide insight into molecular mechanisms that produce synergistic outcomes in small-molecule sensing. Based on DAF-8 immunoprecipitation, we here identify two G-protein–coupled receptors, DAF-37 and DAF-38, which cooperatively mediate ascaroside perception. daf-37 mutants are defective in all responses to ascr#2, one of the most potent dauer-inducing ascarosides, although this mutant responds normally to other ascarosides. In contrast, daf-38 mutants are partially defective in responses to several different ascarosides. Through cell-specific overexpression, we show that DAF-37 regulates dauer when expressed in ASI neurons and adult behavior when expressed in ASK neurons. Using a photoaffinity-labeled ascr#2 probe and amplified luminescence assays (AlphaScreen), we demonstrate that ascr#2 binds to DAF-37. Photobleaching fluorescent energy transfer assays revealed that DAF-37 and DAF-38 form heterodimers, and we show that heterodimerization strongly increases cAMP inhibition in response to ascr#2. These results suggest that that the ascarosides' intricate signaling properties result in part from the interaction of highly structure-specific G-protein–coupled receptors such as DAF-37 with more promiscuous G-protein–coupled receptors such as DAF-38.
