    Notes: A new white beam single-crystal Laue diffraction (LD) and energy-dispersive powder diffraction (EDPD) facility has been constructed on station 9.7 of the SRS. This station receives radiation from a 5-T superconducting wiggler magnet with useful photon flux from 5 to 60 keV and a peak flux on the sample of 7×1011 photons/s/mm2 in a 0.1% bandwidth at 10 keV. The novel design of this facility allows rapid changeover between the two experiments without compromising either. Components have been built in a modular manner for ease of optimization for a particular set of data capture conditions.

    Notes: We reviewed our experience with six T-cell-rich B-cell lymphomas (TRBL) presenting in skin. Immunohistochemical studies were performed on all biopsies. The lymphoid population consisted mainly of CDS and/or UCHL-1 (CD45RO) positive T cells. 5 to 15% of the lymphoid cells stained for the B-cell marker L26 (CD20). Monoclonality of the B-cell component was demonstrated in all cases, utilizing either light chain restriction (5 cases) or clonal immunoglobulin heavy chain gene rearrangement by polymerase chain reaction (PCR) (2 cases). One case was confirmed to be monoclonal by both techniques. Additionally, no clonal rearrangements of the T-cell receptor gamma gene were observed. There was considerable morphological variety in these cases. In H&E stained sections, the differential diagnosis included pseudolymphoma, peripheral T-cell lymphoma, Hodgkin's disease, Lennert's lymphoma and a MALT lymphoma. A significant component of monoclonal plasma cells was present in 3 of 6 cases, suggesting a possible origin from cutaneous immunocytoma. In fact, one of our cases was a biphasic lymphoma displaying TRBL with a small focus of immunocytoma. We conclude that immunophenotypic analysis is necessary for the diagnosis of TRBL. Pathologists should be aware of this type of cutaneous B-cell lymphoma to avoid misinterpretation as a pseudolymphoma.

    Notes: Twelve patients with large plaque parapsoriasis (LPP) were investigated for the presence of predominant T-cell clones, analyzing the T-cell receptor (TCR) γ-chain gene. The diagnostic and prognostic significance of TCR gene rearrangement status was assessed by a correlation with the long-term clinical follow-up. Six out of 12 patients showed a clonal T-cell population. Clinically, among the patients with clonal disease one developed clearcut mycosis fungoides (MF) after a follow-up of 8 years, in the other 5 patients no such diagnosis could be made after follow-up of 2–21 years (median: 9 years). In patients with polyclonal infiltrates the lesions remained virtually unchanged. These findings indicate that in LPP TCR gene rearrangement status has no prognostic significance and does not allow distinction of LPP and early MF. Both conditions show a clonal T-cell infiltrate with similar frequency, are very similar in clinical and histologic presentation and according to recent studies share the same low risk to develop overt MF. Therefore both terms refer to the identical clinical situation. This should be designated as early MF and efforts should concentrate on identifying those patients that are at risk to develop aggressive disease.

    Notes: Abstract. Paramecium primaurelia expresses three major types of surface antigens. We report here the identification of the gene for serotype S, which completes the sequence data of expressed serotypes of P. primaurelia. The complete open reading frame of surface antigen S was identified using a novel technique, based upon the presence of conservative regions in the non-coding areas of the multigene family. We were able to isolate the 7194-bp-long open reading frame from the macronuclear DNA for Serotype 156S. The corresponding mRNA was detected in the two serotype S-expressing stocks, 60 and 156, of P. primaurelia, which clarifies that both stocks are using the same S allele. Comparisons of the nucleic acid and the deduced amino-acid sequence showed high identity to surface antigen 51B of P. tetraurelia, sufficient to cause an immunological cross-reaction in vivo. Immunologically relevant epitopes in vivo were identified in the central regions of the genes, constructed of nearly perfect tandem repeats.

    Notes: Abstract A constructivist-oriented inservice program provided teachers of mathematics (K-12) with intensive two-week summer institutes and weekly classroom follow-up. Pre- and post-program data on student outcomes indicate that, along with transformations in the nature and quality of mathematics activity in the classroom, students' beliefs about learning mathematics changed and elementary students' attitudes toward mathematics improved. Although the instruction they now received focused more on conceptual understanding and less on computational skill, students' scores on standardized tests assessing routine knowledge were maintained.

    Description / Table of Contents: Summary The Clarke-Howel-Evans-McConnell-Syndrome is a rare hereditary disease characterized by palmoplantar keratoses, squamous cell carcinoma of the esophagus and oral leukoplakia. According to a new classification recently proposed by Stevens and colleagues, the syndrome can also be classified as Palmoplantar Ectodermal Dysplasia Type III. We report a family from the Black Forest region of Germany afflicted with the syndrome. The family was traced through five generation. 27 of 46 family members showed tylotic skin changes. In addition, 8 patients showed oral leukoplakia and 5 died from squamous cell carcinoma of the esophagus. Using screening examinations, early changes of the esophageal mucosa could be detected. The responsible gene has been mapped in the family. It is located at 17q23-qter, telomeric to the keratin II gene cluster. Therefore a defect in one of the well known keratin genes can be excluded as a cause of the syndrome.
    Notes: Zusammenfassung Das Clarke-Howel-Evans-McConnell-Syndrom ist ein seltenes hereditäres Krankheitsbild, das durch die Symptome palmoplantare Hyperkeratosen, Ösophaguskarzinom und orale Leukoplakien gekennzeichnet ist. Nach einer kürzlich von Stevens vorgeschlagenen Klassifikation der Palmoplantarkeratosen wird der Symptomenkomplex auch unter dem Begriff ektodermale Dysplasie Typ III subsumiert. In der vorliegenden Arbeit berichten wir über eine betroffene Familie aus dem Schwarzwald, bei der Beobachtungen über 5 Generationen vorliegen. Zusammenfassend traten bei 27 der 46 erfaßten Familienmitglieder tylosisartige Hautveränderungen auf, bei 8 Patienten bestanden zusätzlich orale Leukoplakien. Insgesamt 5 Patienten verstarben an einem Ösophaguskarzinom. Anläßlich von Screeninguntersuchungen konnten endoskopisch frühe Schleimhautveränderungen des Ösophagus diagnostiziert werden. Der dem Syndrom zugrundeliegende Gendefekt wurde kürzlich auch in der von uns beschriebenen Familie auf Chromosom 17q23-qter lokalisiert, telomer gelegen zum Keratin-II-Gencluster. Somit scheidet ein Defekt in einem der bekannten Keratingene als Ursache für die Erkrankung aus. Weitere wichtige Aspekte zu diesem Krankheitsbild werden in der Besprechung erörtert.
