    Notes: The crystal structure of the high Tc superconducting phase in a Pb-doped Bi-Sr-Ca-Cu-O system (nominal composition Pb0.3Bi1.7Sr2Ca3Cu4OY) with a zero-resistance temperature of 107 K was determined with transmission electron microscopy. The investigation shows that the crystal structure, which is different from the Pb-free one, is monocline with dimensions a=0.532 nm, b=0.383 nm, c=3.71 nm, α=β=90° and γ=134.8°. A distinct change of wave vector of incommensurate modulation was also observed.

    Notes: The crystal structure of monoclinic magnesium astrophyllite has been redetermined. The samples were from Khibina alkaline massifs, Russia. K2NaNa(Fe,Mn)4Mg2Ti2[Si4O12]2(OH)4(OH,F)2, Mr = 1202.31, monoclinic, A2, a = 10.370 (3), b = 23.129 (5), c = 5.322 (1) Å, β = 99.55 (2)°, V = 1258.8 (5) Å3, Z = 2, Dx = 3.173 Mg m−3, Mo Kα, λ = 0.71069 Å, μ = 3.74 mm−1, F(000) = 1179, T = room temperature, R = 0.057 for 1720 reflections. The determination indicated that the coordination number of Ti is five. In tetrahedra of astrophyllite the open-branched Si—O chain connected with Ti—O semi-octahedra by corner-sharing formed a continuous sheet. Na atoms have two types of coordination. Their coordination numbers are eight and six. The Na atoms with coordination number eight are situated between layers. Other types of Na atoms are situated within the layers and form Na—O octahedra with partial occupancies.

    Notes: Abstract Respiratory and fermentative pathways co-exist to support growth and product formation in Pichia stipitis. This yeast grows rapidly without ethanol production under fully aerobic conditions, and it ferments glucose or xylose under oxygen-limited conditions, but it stops growing within one generation under anaerobic conditions. Expression of Saccharomyces cerevisiaeURA1 (ScURA1) in P. stipitis enabled rapid anaerobic growth in minimal defined medium containing glucose when essential lipids were present. ScURA1 encodes a dihydroorotate dehydrogenase that uses fumarate as an alternative electron acceptor to confer anaerobic growth. Initial P. stipitis transformants grew and produced 32 g/l ethanol from 78 g/l glucose. Cells produced even more ethanol faster following two anaerobic serial subcultures. Control strains without ScURA1 were incapable of growing anaerobically and showed only limited fermentation. P. stipitis cells bearing ScURA1 were viable in anaerobic xylose medium for long periods, and supplemental glucose allowed cell growth, but xylose alone could not support anaerobic growth even after serial anaerobic subculture on glucose. These data imply that P. stipitis can grow anaerobically using metabolic energy generated through fermentation but that it exhibits fundamental differences in cofactor selection and electron transport with glucose and xylose metabolism. This is the first report of genetic engineering to enable anaerobic growth of a eukaryote.

    Notes: Further spectroscopic studies of gallstones are reviewed with an emphasis on the formation of black pigment gallstones. This type of gallstone appears mainly in Western countries, with only 3% of the cholelithiasis patients in China having black gallstones. Fourier transform infrared absorption and electron paramagnetic resonance are used as spectroscopic probes of gallstones and their metal bilirubinate components. Nonlinear phenomena in gallstone formation were investigated through the appearance of ring structure in gallstones and fractal patterns in the formation in the precipitates of bile salt systems. Although a complete understanding of gallstone formation has not yet been achieved, interesting progress toward this goal has been made recently. © 1997 John Wiley & Sons, Inc.     Description: Smooth muscle (SM) development consists of several processes, including cell fate determination, differentiation, and maturation. The molecular mechanisms controlling SM early differentiation have been studied extensively. However, little is known about the mechanism underlying SM maturation. Cell division cycle 7 (Cdc7) has been shown to regulate cell fate determination in the initial phase of transforming growth factor-β (TGF-β)-induced SM differentiation. Our present study indicates that Cdc7 also regulates SM maturation. Knockdown of Cdc7 suppresses TGF-β-induced expression of SM myosin heavy chain, a late marker of SM differentiation. Cdc7 overexpression, on the other hand, enhances SM myosin heavy chain expression. Interestingly, Cdc7 activates the mRNA expression and promoter activity of myocardin (Myocd), a master regulator of SM differentiation, whose transcription is blocked in the initial phase of the differentiation because TGF-β does not induce Myocd mRNA until after the early SM markers are induced. These data suggest that Cdc7 mediates TGF-β-induced SM maturation via activation of Myocd transcription. Mechanistically, Cdc7 physically and functionally interacts with Nkx2.5 to regulate Myocd promoter activity. Cdc7 appears to enhance Nkx2.5 binding to Myocd promoter, leading to Myocd activation. Taken together, our studies demonstrate that Cdc7 regulates the initial and late phase of SM differentiation through distinct mechanisms.

    Description: Selective ion conduction across ion channel pores is central to cellular physiology. To understand the underlying principles of ion selectivity in tetrameric cation channels, we engineered a set of cation channel pores based on the nonselective NaK channel and determined their structures to high resolution. These structures showcase an ensemble of selectivity filters with a various number of contiguous ion binding sites ranging from 2 to 4, with each individual site maintaining a geometry and ligand environment virtually identical to that of equivalent sites in K+ channel selectivity filters. Combined with single channel electrophysiology, we show that only the channel with four ion binding sites is K+ selective, whereas those with two or three are nonselective and permeate Na+ and K+ equally well. These observations strongly suggest that the number of contiguous ion binding sites in a single file is the key determinant of the channel’s selectivity properties and the presence of four sites in K+ channels is essential for highly selective and efficient permeation of K+ ions.

    Description: Atopic dermatitis (AD) is a chronic inflammatory skin disease associated with defective epidermal barrier function, characterized by decreased stratum corneum (SC) hydration, and increased TEWL (transepidermal water loss) and pH. Staphylococcus aureus (SA) skin colonization is present in nearly all cases of AD exacerbation and correlates with disease severity, 1 suggesting an infection – inflammation cycle. The use of sodium hypochlorite (bleach) bath alone and as an adjunct to intranasal mupirocin ointment is associated with reduction in AD severity, 2,3 and decreases annual skin infection cases by ten-fold. 4 In a Cochrane review on various interventions to reduce SA colonization, including oral antibiotics, topical steroids and antibiotic ointments, only bleach bath showed a significant improvement in AD severity. 5 This article is protected by copyright. A. Soluyanov, and F. Baumberger The authors present here several advances towards a comprehensive understanding of WTe 2 . Using microfocus laser ARPES, they resolve for the first time the distinct electronic structure of both inequivalent top and bottom (001) surfaces of WTe 2 . Moreover, they demonstrate for the first time the presence of large surface state Fermi arcs on both surfaces. Using surface electronic structure calculations, they further demonstrate that these Fermi arcs are topologically trivial and that their existence is independent of the presence of type-II Weyl points in the bulk band structure. Contrary to common belief, the observation of surface state Fermi arcs is thus not suitable to robustly identify a type-II Weyl semimetal. Finally, the authors use the observation of Fermi arcs and distinct top and bottom surfaces to clarify the controversial bulk electronic structure of WTe 2 . They show that the bulk Fermi surface is formed by three-dimensional electron and hole pockets with areas that are found to be in good agreement with transport experiments, with the exception of small hole pockets that have not been observed in quantum oscillation experiments.
