    Notes: Abstract We investigated the influence of tumour depth and differences in blood concentration and autofluorescence between tumour and normal tissue on the fluorescence of a tumour-localising agent. Carotenoporphyrin, CP(Me)3, was injected into rats and nude mice with intradermal tumours. On the tumours an incision was made, uncovering 2 mm2 of the tumour, and fluorescence measurements, including excitation-emission maps and fluorescence ratios, were made on skin, covered and uncovered tumour. The measured fluorescence ratio in the uncovered tumour showed a three- to tenfold increase compared to the covered tumour. We used a one-dimensional layered tissue model to analyse the data. In conclusion, even with a high tumour-selectivity deeper lying tumours cannot always be detected, particularly if the tumour has a high blood concentration or low autofluorescence intensity.

    Description: Stable isotope and ice-rafted debris records from three core sites in the mid-latitude North Atlantic (IODP Site U1313, MD01-2446, MD03-2699) are combined with records of ODP Sites 1056/1058 and 980 to reconstruct hydrographic conditions during the middle Pleistocene spanning Marine Isotope Stages (MIS) 9–14 (300–540 ka). Core MD03-2699 is the first high-resolution mid-Brunhes record from the North Atlantic's eastern boundary upwelling system covering the complete MIS 11c interval and MIS 13. The array of sites reflect western and eastern basin boundary current as well as north to south transect sampling of subpolar and transitional water masses and allow the reconstruction of transport pathways in the upper limb of the North Atlantic's circulation. Hydrographic conditions in the surface and deep ocean during peak interglacial MIS 9 and 11 were similar among all the sites with relative stable conditions and confirm prolonged warmth during MIS 11c also for the mid-latitudes. Sea surface temperature (SST) reconstructions further reveal that in the mid-latitude North Atlantic MIS 11c is associated with two plateaus, the younger one of which is slightly warmer. Enhanced subsurface northward heat transport in the eastern boundary current system, especially during early MIS 11c, is denoted by the presence of tropical planktic foraminifer species and raises the question how strongly it impacted the Portuguese upwelling system. Deep water ventilation at the onset of MIS 11c significantly preceded surface water ventilation. Although MIS 13 was generally colder and more variable than the younger interglacials the surface water circulation scheme was the same. The greatest differences between the sites existed during the glacial inceptions and glacials. Then a north – south trending hydrographic front separated the nearshore and offshore waters off Portugal. While offshore waters originated from the North Atlantic Current as indicated by the similarities between the records of IODP Site U1313, ODP Site 980 and MD01-2446, nearshore waters as recorded in core MD03-2699 derived from the Azores Current and thus the subtropical gyre. Except for MIS 12, Azores Current influence seems to be related to eastern boundary system dynamics and not to changes in the Atlantic overturning circulation.

    Description: The mechanisms leading to the emergence of strong 100,000-year ice age cycles at the end of the Middle Pleistocene Transition (MPT), ~670-800 ka, encompassing Marine Isotope Stage (MIS) 19, 18 and 17, are still not fully understood. This period is associated with anomalously warm North Atlantic sea surface temperatures enhancing moisture production and transport to northern hemisphere ice-sheets. However, the most relevant processes explaining the progressive ice accumulation during the end of MPT remain unclear yet, partly due to the poor knowledge of the Eurasian environments and atmospheric climate. New marine-terrestrial climatic records from the IODP Site U1385 (south-west Iberian margin) and their comparison with loess records from the Chinese Plateau reveal a nearly synchronous, long-term warming and wetting trend in the two subtropical regions from MIS 19 to 17. Mediterranean forest cover and winter precipitation were stronger during the MIS 18 glacial compared to the MIS 19 interglacial and significantly correlated with oceanic moisture production off Iberia, also suggested by iLOVECLIM climate model simulation. Likewise, the strengthening of the East Asian Summer monsoon was associated with the increasing thermal gradient between the tropical and polar West North Pacific Ocean enhancing moisture production off eastern Eurasia from MIS 19 to 17. These findings provide supporting evidence for a nearly-continuous supply of oceanic moisture from the mid-latitude North Atlantic and tropical West North Pacific to higher latitudes through the westerlies, which might have critically contributed to the accelerated expansion of Eurasian and North American ice-sheets at the end of the MPT.

    Description: This study aimed at investigating the in vitro activities of amoxicillin-clavulanate, doxycycline, ceftazidime, imipenem, and trimethoprim-sulfamethoxazole against Burkholderia pseudomallei in planktonic and biofilm forms, through broth microdilution and resazurin-based viability staining, respectively. In planktonic growth, the strains were susceptible to the drugs, while in biofilm growth, significantly higher antimicrobial concentrations were required, especially for ceftazidime and imipenem, surpassing the resistance breakpoints. These results highlight the importance of the routine evaluation of biofilm antimicrobial susceptibility.

    Description: Proteins containing repetitive amino acid domains are widespread in all life forms. In parasitic organisms, proteins containing repeats play important roles such as cell adhesion and invasion and immune evasion. Therefore, extracellular and intracellular parasites are expected to be under different selective pressures regarding the repetitive content in their genomes. Here, we investigated whether there is a bias in the repetitive content found in the predicted proteomes of 6 exclusively extracellular and 17 obligate intracellular protozoan parasites, as well as 4 free-living protists. We also attempted to correlate the results with the distinct ecological niches they occupy and with distinct protein functions. We found that intracellular parasites have higher repetitive content in their proteomes than do extracellular parasites and free-living protists. In intracellular parasites, these repetitive proteins are located mainly at the parasite surface or are secreted and are enriched in amino acids known to be part of N- and O-glycosylation sites. Furthermore, in intracellular parasites, the developmental stages that are able to invade host cells express a higher proportion of proteins with perfect repeats relative to other life cycle stages, and these proteins have molecular functions associated with cell invasion. In contrast, in extracellular parasites, degenerate repetitive motifs are enriched in proteins that are likely to play roles in evading host immune response. Altogether, our results support the hypothesis that both the ability to invade host cells and to escape the host immune response may have shaped the expansion and maintenance of perfect and degenerate repeats in the genomes of intra- and extracellular parasites.

    Description: Cell death is an important target for imaging the early response of tumors to treatment. We describe here the validation of a phosphatidylserine-binding agent for detecting tumor cell death in vivo based on the C2A domain of synaptotagmin-I. Methods: The capability of near-infrared fluorophore-labeled and 99m Tc- and 111 In-labeled derivatives of C2Am for imaging tumor cell death, using planar near-infrared fluorescence imaging and SPECT, respectively, was evaluated in implanted and genetically engineered mouse models of lymphoma and in a human colorectal xenograft. Results: The fluorophore-labeled C2Am derivative showed predominantly renal clearance and high specificity and sensitivity for detecting low levels of tumor cell death (2%–5%). There was a significant correlation ( R 〉 0.9, P 〈 0.05) between fluorescently labeled C2Am binding and histologic markers of cell death, including cleaved caspase-3, whereas there was no such correlation with a site-directed mutant of C2Am (iC2Am) that does not bind phosphatidylserine. 99m Tc-C2Am and 111 In-C2Am also showed favorable biodistribution profiles, with predominantly renal clearance and low nonspecific retention in the liver and spleen at 24 h after probe administration. 99m Tc-C2Am and 111 In-C2Am generated tumor-to-muscle ratios in drug-treated tumors of 4.3 x and 2.2 x , respectively, at 2 h and 7.3 x and 4.1 x , respectively, at 24 h after administration. Conclusion: Given the favorable biodistribution profile of 99m Tc- and 111 In-labeled C2Am, and their ability to produce rapid and cell death–specific image contrast, these agents have potential for clinical translation.

    Description: Fossil eggs and embryos that provide unique information about the reproduction and early growth of vertebrates are exceedingly rare, particularly for pterosaurs. Here we report on hundreds of three-dimensional (3D) eggs of the species Hamipterus tianshanensis from a Lower Cretaceous site in China, 16 of which contain embryonic remains. Computed tomography scanning, osteohistology, and micropreparation reveal that some bones lack extensive ossification in potentially late-term embryos, suggesting that hatchlings might have been flightless and less precocious than previously assumed. The geological context, including at least four levels with embryos and eggs, indicates that this deposit was formed by a rare combination of events, with storms acting on a nesting ground. This discovery supports colonial nesting behavior and potential nesting site fidelity in the Pterosauria.

    Description: ABSTRACT The Araguari River is located in the central uplands of Brazil. Over the past 40 years, the region has undergone considerable change in its economy because of the green revolution and urban development that followed the construction of Brasilia in the 1960s, the new capital of Brazil. The main changes in the landscape were the construction of two hydroelectric power plants across the main valley, resulting in ~95 per cent loss in the natural features of the lower 200 km of the Araguari course. The inundation of the river valley has dramatically modified the landscape, with submersion of many locations, especially gorges, islands as well as tributary valleys and historic sites. The concept of geodiversity implies that abiotic elements of nature have unique qualities that are not uniformly distributed and include geomorphic, geologic and pedologic qualities within the context of their location. The concept of geodiversity is used to understand and assess losses from the Araguari River Valley landscapes. In this research, we present the total quantity of geosite types occurring for our Araguari River Valley study site. Twelve complex sites were identified, with six transformed to simple geosites because of inundation associated with formation of two reservoirs. The major changes caused by river impoundment resulted in a reduction in the richness of geosites and a reduction in their complexity by 50 per cent. The study results find that 14.3 per cent of the original geodiversity was lost after valley inundation. Quantification of changes in geodiversity is essential to comprehensively characterize the environmental impact of dams. Copyright © 2012 John Wiley & Sons, Ltd.
