    Notes: Summary The authors report their preliminary experience with the use of radiolabelled monoclonal antibodies (MAb) as an adjuvant treatment for 33 malignant gliomas. MAbs employed in this study are raised against Tenascin (TN) which is an antigen of the extracellular matrix of the tumour. It has also been found in neoplastic cells but never in normal brain tissue. This therapy is aimed to give a local high dose radiation (boost) while sparing healthy brain structures. This treatment has always been well tolerated and no adverse reactions at the level of CNS or major extraneural organs has been observed. Significant improvement of median survival has been obtained but this result should be cautiously evaluate since the study is non-randomized. Comparison with other current adjuvant technique is briefly discussed.

    Notes: Summary  The site of leakage in a patients with rhinorrhea of various origin may be difficult to identify. The aim of our paper is to evaluate the contribution of cisternography in combination with single photon emission tomography (SPECT) to identify the fistulous track. From 1/1/1992 to 30/11/1997 we studied 20 patients with rhinorrhea posing a challanging diagnostic problem as to identification of the leakage site. Two mls of Indium DTPA (In 111) were injected into the subarachnoid space by the lumbar route. The tracer was followed by planar scintigraphy until it reached the cranial base and subsequently the SPECT acquisition started. A fistula was demonstrated in all of our cases including patients with no active leakage at the time of examination, patients with no bone defects on thin sliced CT scanning or patients with a normal MRI. At surgery the fistulous track was confirmed in all but two cases when a bilateral fistula was operatively identified only on one side. In conclusion whenever a CT scanning fails to demonstrate significant bone defects and MRI does not localize a fistulous track, SPECT cisternography via the lumbar route proved in our experience to be a reliable examination for a precise diagnosis.

    Notes: Abstract. The high-grade malignant gliomas (anaplastic astrocytomas and glioblastoma) have a very bad prognosis since the available methods of treatment (surgery, radiotherapy and chemotherapy) are unable to control the progression of the disease for long. The use of specific monoclonal antibodies labelled with a suitable isotope (iodine-131 or yttrium-90) represents an effective approach to hamper tumour regrowth. Some authors have injected the antibodies intravenously, or have tried to increase the tumour/background ratio with the avidin/ biotin system. In many cases the labelled monoclonal antibodies were injected directly into the tumoral bed after the operation. The authors’ experiences concern a quite large locoregional radioimmunotherapy study which was performed by using antitenascin antibodies labelled initially with 131I and more recently with 90Y. The clinical results demonstrate the ability of this technique to control, for a long time, the growth of these tumours. The glioblastoma median survival was prolonged to 25 months (131I group) or 31 months (90Y group). The response rate (which comprises PR, CR and NED) was 47.1% (glioblastoma 131I group) or 40% ( glioblastoma 90Y group). In many cases a significant tumour shrinking effect was radiologically demonstrated. The use of 90Y proved more favourable in bulky lesions, and reduced the radioprotection problems.

    Notes: Abstract This study centers on marker chromosomes carrying expanded chromosomal regions which were observed in two independent derivatives of the AA12 murine fibrosarcoma line, the 10−3 M MTX-res H2 and the 5×10−7 M MTX-res E. Previous characterization of the marker chromosomes of MTX-res variants showed their common derivation from a marker chromosome (m) of the parental line, endowed with two interstitial C-bands. Cytogenetic evidence pointed to one C-band ofm as the site involved in the chromosomal rearrangements leading to the HSR/ASR chromosomes. ISH of a3H-labeled satellite DNA probe allowed satellite sequences flanking the HSR/ASR in the marker chromosomes, where the C-band was no longer visible, to be detected. FISH experiments using biotinylated DHFR and satellite DNA probes showed that the respective target sequences are contiguous in new marker chromosomes. They also allowed inter- and intrachromosomal rearrangements to be seen at DHFR amplicons and satellite sequences. Double-color FISH using digoxygenated satellite DNA and biotinylated pDHFR7 showed that in a marker chromosome from the H2 cell line the two target sequences are not only adjacent, but closer than 3 Mb, as indicated by overlapping of the different fluorescence signals given by the two probes. Another marker chromosome in the E variant was shown to display a mixed ladder structure consisting of a head-to-head tandem of irregularly-sized satellite DNA blocks, with two symmetrical interspersed DHFR clusters.

    Description: A crucial problenl for seismic archeology is how to recognize seismic effects and how to date them. On an experimental basis. we proposed that the problem be reversed, and that we begin at the other end: i.e. by analyzing already known seismic effects on ancient structures, testified by written sources. to be able to .calibrate〉〉 the types or possible observations and any subsequent elaborations. The choice of the walls of Noto was suggested by the fact that Noto was abandoned following the earthquake of l693 (I,= XI MCS. Me 7.5) which had already been studied in depth as part of an ING research programme (1988-92). Moreover, just after recent research, this event proved to be reconstructed with a high quality standard. Photogrammetric measurements were made on several parts of the town walls to plot a numerical model aimed at ascertaining specific aspects of the earthquake damage. An estimate of the ground acceleration during the earthquake has been attempted via non-linear finite-element analyses of a building located by the main city gate. The analyses show that. in order to obtain the building vault collapse, a ground acceleration of 0.5 to 0.7 g had to be reached during the earthquake. This result, typical of a strong earthquake such as the one of 1693, proves that an approach based on finite element analysis and a sound engineering judgment Inay be systematically applied to historical earthquake sites to obtain some estimates of ground acceleration in historical earthquakes. On the whole, this work aimed at starting up the second development phase of the great event of 1693 of which the macroseismic erfects are known. In the meantime, some possibilities of tackling structural analyses in seismic archaeology are being explored.

    Description: This paper investigates the use of switching converters for the power supply distribution to calorimeters in the ATLAS experiment when the Large Hadron Collider (LHC) will be upgraded beyond the nominal luminosity. Due to the highly hostile environment the converters must operate in, all the main aspects are considered in the investigation, from the selection of the switching converter topologies to the thermal analysis of components and PCBs, with attention to reliability issues of power devices subject to ionizing radiations. The analysis focuses on the particular, but crucial, case of the power supplies for calorimeters, though several outcomes of the research can profitably be applied to other detectors like muon chambers.

    Description: Little is known regarding the post-transcriptional networks that control gene expression in eukaryotes. Additionally, we still need to understand how these networks evolve, and the relative role played in them by their sequence-dependent regulatory factors, non-coding RNAs (ncRNAs) and RNA-binding proteins (RBPs). Here, we used an approach that relied on both phylogenetic sequence sharing and conservation in the whole mapped 3'-untranslated regions (3'-UTRs) of vertebrate species to gain knowledge on core post-transcriptional networks. The identified human hyper conserved elements (HCEs) were predicted to be preferred binding sites for RBPs and not for ncRNAs, namely microRNAs and long ncRNAs. We found that the HCE map identified a well-known network that post-transcriptionally regulates histone mRNAs. We were then able to discover and experimentally confirm a translational network composed of RNA Recognition Motif (RRM)-type RBP mRNAs that are positively controlled by HuR, another RRM-type RBP. HuR shows a preference for these RBP mRNAs bound in stem–loop motifs, confirming its role as a ‘regulator of regulators’. Analysis of the transcriptome-wide HCE distribution revealed a profile of prevalently small clusters separated by unconserved intercluster RNA stretches, which predicts the formation of discrete small ribonucleoprotein complexes in the 3'-UTRs.

    Description: Essential tremor (ET) is the most prevalent movement disorder, affecting millions of people in the USA. Although a positive family history is one of the most important risk factors for ET, the genetic causes of ET remain unknown. In an attempt to identify genetic causes for ET, we performed whole-exome sequencing analyses in a large Spanish family with ET, in which two patients also developed epilepsy. To further assess pathogenicity, site-directed mutagenesis, mouse and human brain expression analyses, and patch clamp techniques were performed. A disease-segregating mutation (p.Gly1537Ser) in the SCN4A gene was identified. Posterior functional analyses demonstrated that more rapid kinetics at near-threshold potentials altered ion selectivity and facilitated the conductance of both potassium and ammonium ions, which could contribute to tremor and increase susceptibility to epilepsy, respectively. In this report, for the first time, we associated the genetic variability of SCN4A with the development of essential tremor, which adds ET to the growing list of neurological channelopathies.

    Description: The focus of the study is optimizing the technique for estimating geocenter motion and variations in J 2 by combining data from the Gravity Recovery and Climate Experiment (GRACE) satellite mission with output from an Ocean Bottom Pressure (OBP) model and a Glacial Isostatic Adjustment (GIA) model [ Swenson et al. , 2008; Sun et al. , 2016]. First, we conduct an end-to-end numerical simulation study. We generate input time-variable gravity field observations by perturbing a synthetic Earth model with realistically simulated errors. We show that it is important to avoid large errors at short wavelengths and signal leakage from land to ocean, as well as to account for self-attraction and loading (SAL) effects. Second, the optimal implementation strategy is applied to real GRACE data. We show that the estimates of annual amplitude in geocenter motion are in line with estimates from other techniques, such as SLR and global GPS inversion. At the same time, annual amplitudes of C 10 and C 11 are increased by about 50% and 20%, respectively, compared to estimates based on [ Swenson et al. , 2008]. Estimates of J 2 variations are by about 15% larger than SLR results in terms of annual amplitude. Linear trend estimates are dependent on the adopted GIA model, but still comparable to some SLR results.
