    Notes: Therapy of chronic hepatitis C non- responders to interferon monotherapy with standard doses of interferon plus ribavirin is usually ineffective.〈section xml:id="abs1-2"〉〈title type="main"〉Aim:To evaluate the efficacy and tolerability of high-dose prolonged combination retreatment in non- responder patients.〈section xml:id="abs1-3"〉〈title type="main"〉Methods:Patients were retreated for 6 months with 6 MU αIFN on alternate days and 1000 or 1200 mg/day ribavirin. HCV-RNA negative patients continued therapy for an additional 6 months.〈section xml:id="abs1-4"〉〈title type="main"〉Results:Forty patients (29 males, mean age 49.7 years, 34 genotype 1b, 11 with F3 fibrosis) were treated. At 6 months, 20 (50%) patients were HCV-RNA negative but six of them discontinued therapy because of adverse events. A sustained response was achieved in 28% of patients (11/40). A sustained response was more frequent among patients with genotype non-1b than in those with genotype 1b (67 vs. 21%, P=0.005) and clearance of HCV-RNA in the first 3 months had a high predictive value for sustained response (100% of sustained responders vs. 24% of non-responders, P=0.0001).〈section xml:id="abs1-5"〉〈title type="main"〉Conclusions:High-dose prolonged combination therapy in non-responders to IFN monotherapy leads to a higher rate of sustained response than the standard combination regimen. Tolerability may be a rate-limiting factor. Maximal effectiveness can be predicted in patients with non-1b genotype and in those who clear HCV-RNA soon after starting retreatment.

    Notes: Abstract Background. Pelizaeus-Merzbacher disease (PMD) is a rare dysmyelinating disorder characterised by early pendular nystagmus, often rotatory and muscular hypotonia with subsequent ataxia, spasticity and mental retardation. Various point mutations or duplications in the PLP gene on the X chromosome are responsible for PMD in the majority of patients. Autosomal recessive inheritance, particularly in the connatal form, cannot be excluded. Three different forms of the disease have been identified based on their onset, progression and severity of myelin pathology indicated by MRI features. Objective. To determine if MR spectroscopy is useful in the diagnosis of the connatal form of PMD. Materials and methods. Proton MR spectroscopy was performed on two children with connatal PMD. Results. Our patients showed a markedly decreased peak of Cho. This alteration is well represented by quantitative analysis of the NAA-to-Cho ratio, which is the most important ratio affected. A significant decrease of the Cho-to-Cr ratio is also present. In the connatal form of PMD, global lack of myelination may be relevant, as demonstrated by a significant Cho peak reduction. Conclusions. Proton MR spectroscopy may be of diagnostic value in metabolic and destructive disorders of the brain. A greater number of patients with connatal PMD is needed in order to elucidate the significance of reduction of the Cho peak.

    Notes: Abstract Hear-transfer rates from an axially enclosed transferred arc to a surrounding water-cooled cylindrical sleeve, 15 cm high, were measured. The arc (argon or nitrogen) was struck between a movable cathode within the sleeve and a bath of molten copper below the sleeve, serving as anode. The distance from the bottom of the sleeve to the surface of the molten copper (L o) was constant. Variables studied were the diameterD of the sleeve (5, 7.5. and 10 cm), the length of the arc within the sleeveL (5, 10, and 15 cm), the currentI (200, 250, and 300 A) and a tangential flow of gas or vortex within the sleeve (0, ?0, and 50 liters/min). The total power transferred to the sleeve,P s was measured caloronetrically and was the sure ofP r the effective power radiated by the arc of lengthL within the sleeve.P a, the power radiated into the sleeve from the arc of length Lo below the sleeve, andP o , the power radiated from the melt surface (a constant of small value), minusP a , the power lost by convection from the sleeve (negligible, except for a strong vortex). BothP r andP o were found to be equal to the product of the Joule heat released within their respective arc lengths, IVgL and IVg0L0 (where Vg and Vg0 are the voltage gradients), and dimenonless efliciency factors, ηr and η0. which are functions ofL/D andL 0 /D, respectively, for each gas, regardless of the geometry of the sleeve, the current, and the strength of the vortex.

    Description: Vertebrate-like T 2 AG 3 telomeres in tlc1-h yeast consist of short double-stranded regions and long single-stranded overhang (G-tails) and, although based on Tbf1-capping activity, they are capping deficient. Consistent with this idea, we observe Y’ amplification because of homologous recombination, even in the presence of an active telomerase. In these cells, Y’ amplification occurs by different pathways: in Tel1 + tlc1h cells, it is Rad51-dependent, whereas in the absence of Tel1, it depends on Rad50. Generation of telomeric G-tail, which is cell cycle regulated, depends on the MRX (Mre11-Rad50-Xrs2) complex in tlc1h cells or is MRX-independent in tlc1h tel1 mutants. Unexpectedly, we observe telomere elongation in tlc1h lacking Rad51 that seems to act as a telomerase competitor for binding to telomeric G-tails. Overall, our results show that Tel1 and Rad51 have multiple roles in the maintenance of vertebrate-like telomeres in yeast, supporting the idea that they may participate to evolutionary conserved telomere protection mechanism/s acting at uncapped telomeres.

    Description: Symbiotic stars are long-orbital-period interacting binaries characterized by extended emission over the whole electromagnetic range and by complex photometric and spectroscopic variability. In this paper, the first of a series, we present Optical Gravitational Lensing Experiment (OGLE) light curves of all the confirmed symbiotic stars in the Large Magellanic Cloud, with one exception. By careful visual inspection and combined time series analysis techniques, we investigate for the first time in a systematic way the photometric properties of these astrophysical objects, trying in particular to distinguish the nature of the cool component (e.g. semiregular Variable versus OGLE small-amplitude red giant), to provide its first-order pulsational ephemerides and to link all this information with the physical parameters of the binary system as a whole. Among the most interesting results, there is the discovery of a 20 yr-long steady fading of Sanduleak's star, a peculiar symbiotic star known to produce the largest stellar jet ever discovered. We discuss by means of direct examples the crucial need for long-term multi-band observations to get a real understanding of symbiotic and other interacting binary stars. We eventually introduce BOMBOLO, a multi-band simultaneous imager for the Southern Astrophysical Research 4 m Telescope, whose design and construction we are currently leading.
