    Notes: Abstract The relationship between each of two pharmacologic effects (tachycardia and psychological “high”) of delta-9-tetrahydrocannabinol (THC) and plasma THC concentration was investigated in three male and three female experienced marihuana smokers. Each subject smoked one 1% THC cigarette on two occasions separated by 2 h. Heart rate and subjective psychological self-rating were determined frequently throughout the 4 h study period. Data were analyzed by calculating the area under the parameter versus time curves, constructing hysteresis plots, and calculating the decay rate constants from pharmacologic effect versus time plots. In both males and females, dose inhaled and psychological response were apparently equivalent for the first and second cigarettes. While all subjects exhibited marked tachycardia in response to the first cigarette, heart rate in both male and female subjects was not increased as markedly during the second cigarette. Interestingly, female subjects had less tachycardiac response than males during the second cigarette. Hysteresis plots revealed that both heart rate and subjective psychological effects were elicited in an effect compartment which was “deep” relative to the reference plasma compartment. The time courses of tachycardiac and psychological responses lagged behind the plasma THC concentration-time profile. Zero-order decay rate constants for subjective psychological rating did not change substantially from the first to second cigarette. This study suggests that plasma THC concentration is a poor predictor of simultaneously occurring physiological and psychological pharmacologic effects.

    Notes: Abstract. As the prevalence of childhood obesity increases, exercise testing of obese children is likely to increase as well. This article discusses the implications of pediatric obesity for exercise testing and provides some recommendations for conducting tests and evaluating results. Studies comparing obese and nonobese children during exercise testing indicate that obese children are capable of meeting the challenges of exercise testing to nearly the same extent as their nonobese peers. Their physiologic responses, at least for the levels of obesity reported in the literature, are not sufficiently different from their nonobese counterparts to necessitate major changes in test protocols. Laboratory staff should pay special attention to fostering confidence in the obese child during the pretest routine.

    Notes: Polycapillary optics, i.e. shaped arrays consisting of hundreds of thousands of hollow glass capillary tubes, can be used to redirect, collimate or focus X-ray beams. X-rays emitted over a large angular range from conventional laboratory-based sources can be transformed into a beam with a small angular divergence or focused onto a small sample or sample area. Convergent beams of X-rays, with convergence angles as high as 15°, have been produced using polycapillary X-ray optics. Focused-spot sizes as small as 20 µm have been achieved, with flux densities two orders of magnitude larger than that produced by pinhole collimation. This results in a comparable decrease in data collection times because of the increase in direct-beam intensity and reciprocal-space coverage. In addition, the optics can be employed to reduce background and provide more convenient alignment geometries. The inverse dependence of the critical angle for total external reflection on photon energy results in suppression of high-energy photons. This effect can be employed to allow the use of higher tube potentials to increase the characteristic line emission and has also been employed to increase significantly the Kα/Kβ ratio in Cu radiation. Measurements of X-ray diffraction data and crystallographic analyses have been performed for systems ranging from elemental crystals to proteins. Data from a lysozyme protein `standard' with a slightly convergent beam, taken in 3 min per frame with 2° oscillation with a 2.8 kW source, refined to an intensity variance of 5% compared to a standard data set. High-quality data were also obtained with a 0.03 kW fixed-anode source and a 2° convergent lens in 5 min per frame.

    Notes: Summary The blood and urinary clearances of cis-diammine 1,1-cyclobutane dicarboxylate platinum(II) (CBDCA, JM8) were determined in four patients with malignancy. A 40 μCi iv injection of 191Pt/193Pt (3:1)-labelled CBDCA was followed by serial blood and urine sampling to 72h. The blood clearance was triphasic, mean values for the fast, intermediate, and slow phases being 10.8 min, 2.5 h, and 125 h, respectively. The fraction of activity excreted in the urine within the first 6 h had a mean value of 66.7%, contrasting with 26.4% for cisplatin. There was only a small fraction of CBDCA excreted by the slow phase (1.5%) as against an average of 20% for CHIP and 27% for cisplatin. The early and rapid renal clearance of CBDCA may account for reduced nephrotoxicity.

    Notes: Summary The in vivo distribution in man of the four platinum derivatives cis-dichlorodiammine platinum(II) (DDP), cis-trans-dichlorodihydroxy-bis-(isopropylamine) platinum (IV) (CHIP), cis-dichloro-bis-cyclopropylamine platinum (II) (CP), and cis-diammine 1,1-cyclobutanedicarboxylate platinum(II) (CBDCA) has been observed. The availability of these compounds labelled with the radioactive isotope of platinum, platinum-191, has made serial in vivo imaging of their distribution possible. Injection of 17–35 MBq (5–28 mg) of the labelled compound IV was followed by imaging, using a gamma camera, with particular reference to the kidneys, liver, and tumour site. Hepatic and renal clearances were observed in all nine patients, but no unequivocal evidence of tumour uptake was found. The left kidney uptake was estimated at times up to 7 days after injection. Mathematical analysis of some of the uptake curves failed to show any significant difference between the clearance times observed. However, the two patients who received CBDCA did show a higher initial renal uptake, falling within the 1st day to levels comparable with those of the other compounds, and the three patients who received DDP showed consistently liver uptake.

    Notes: Summary The stigmas of species inAneilema andCommelina are trifid and comprise elongate papillae. Progressive degeneration of papular cells is observed in stigmas from open flowers and at anthesis papillae may be moribund and collapsed. Fluid emanating from the hollow style flows onto the surface through ruptures in the cuticle at the interpapillar junctions into the interstices at maturity. This secretion stains positively for protein. Stigmas are of the “wet” type. The cuticle overlying the papillar cells is ridged and at the final stages prior to flowering this cuticle becomes detached from the underlying cellulosic wall. The sub-cuticular space so formed is filled with secretion. InAneilema species detachment of cuticle is at the papillar tip and along the lateral walls. InCommelina species the anticlinal walls of adjacent papillae are strongly attached for much of their length and thus detachment of cuticle is restricted to the papillar tip. The cell wall at the tip in both genera may proliferate forming a rudimentary transfer-cell type wall. The secretion is considered to be produced by the papillar cells. It is PAS positive but fails to stain for protein and in both the light and electron microscopes appears heterogenous. Pollen attachment, hydration, germination and early tube growth are very rapid following self-pollination, the pollen tubes entering the neck of the style within ten minutes of attachment. A unique character combination involving pollen and stigmas in these genera indicates a monophyletic origin.
