    Notes: Summary Formalin-fixed paraffin-embedded sections of fetal and adult human lungs were examined for the localization of gastrin releasing peptide (GRP) and calcitonin (CT) in bronchial endocrine-type cells with the indirect immunoperoxidase method. In fetal lungs, the appearance of CT was much later than that of GRP, and CT-containing cells were less frequent than GRP-containing cells which, in later fetal life, formed “neuroepithelial bodies (NEB)”. NEB revealed little CT immunoreactivity. The serial section technique demonstrated that all CT immunoreactants in fetal and neonatal lungs were present within GRP-containing cells. An increase of CT immunoreactivity in GRP-containing cells was observed in perinatal lungs. The lung of a neonate who died of hyaline membrane disease contained the most abundant CT immunoreactants. In adult lungs, CT immunoreactivity was identified in some GRP-containing cells but cells containing only GRP or CT were also present. Cells containing both hormones occasionally formed hyperplastic foci in the bronchi of fibrotic lungs. Most cells of pulmonary tumorlets consistently showed GRP immunoreactivity, but the number of CT immunoreactive cells in them varied greatly.

    Notes: Summary Immunohistochemical localization of theN-terminal fragment (1–76) (NTF) of human pro-opiomelanocortin (POMC) was studied in human adult and fetal pituitary glands, as well as in pituitary adenomas associated with Cushing's syndrome and in ectopic ACTH-producing tumors. Comparison of localization between NTF and ACTH was performed using mirror sections. Our results indicated concomitant localization of NTF and ACTH in the same cells, not only in normal adult and fetal pituitaries but also in pituitary adenomas and ectopic ACTH producing tumours. Specificity of the NTF staining was confirmed by immunoabsorption. Negative staining of the bovine pituitary gland indicated the immunohistochemical localization ofN-terminal (1–45) of human POMC as there is a known species difference in the sequence 1–45 between human and the bovineN-terminal fragment. Presence of NTF in cisterna of rough endoplasmic reticulum indicates its production by small cell carcinoma. These findings, together with the previous studies, suggest that the complete form of POMC is produced in the tumours as well as in normal pituitaries.

    Notes: Summary This light microscopically pleomorphic tumor, which occurred in the renal capsule of 35-year-old female was diagnosed as a malignant fibrous histiocytoma. Ultrastructurally, the tumor showed two major components; fibroblast-like and histiocyte-like cells. This report stresses the rarity of such neoplasm in the kidney and also the usefulness of electron microscopy in establishing the diagnosis.

    Notes: Summary Ten FSH producing pituitary adenomas were studied immunohistochemically. 9 cases were in males, and 7 showed elevated serum FSH levels. Immunohistochemically, all cases showed the presence of α-subunit and FSH-β subunits in many tumour cells. These two subunits were frequently colocalized in the same cells. However, the expression of LH-β subunit was extremely low (1 of 10 cases exhibiting occasional LH-β positive tumour cells), although it has been reported that FSH-β and LH-β subunits are colocalized in the same cells of the normal adult pituitary gland. Immunoelectron microscopically, α-subunits and FSH-β were present in the secretory granules and suggested the co-release of subunits or secretion of combined form of FSH. In 7 cases, TSH-β was positive, and in some cases, TSH-β was colocalized in the same tumour cells which contained α-subunit and FSH-β subunit. A few cases also demonstrated immunoreactivity for PRL and ACTH. Our immunohistochemical studies suggest that FSH adenomas are multihormonal and that there is abnormal gene expression in FSH cells with loss of LH-β appearance and co-expression of TSH-β.

    Notes: Summary An autopsy case of thymic carcinoma is reported. The surgically excised cystic tumour from a 38-year-old Japanese male mimicked a thymic cyst pathologically. After a clinical course of 6 years, postmortem examination disclosed extensive metastases, including cystic lesions lined by benign-loocking squamous epithelium. This evidence suggested that the present tumour was one of the specific variants of thymic carcinoma and not a thymic cyst with malignant transformation.

    Notes: Abstract The expression and distribution of prolactin (PRL) mRNA and their alterations induced by estrogen and bromocriptine were investigated using non-radioisotopic in situ hybridization (ISH) at the electron microscopic (EM) level. Our EM-ISH studies using biotinylated oligonucleotide probes showed that estrogen induced whirling changes of the rough endoplasmic reticulum (RER) of female rat PRL cells and increased transcription of PRL genes located on the polysomes of the whirling RER. The presence of mammosomatotroph cells in the rat pituitary gland was also verified in our EM-ISH studies. After bromocriptine administration, PRL cells contained many secretory granules due to the inhibition of secretion. Pre- and post-embedding EM-ISH and northern hybridization studies revealed that bromocriptine induced the distorted, vesiculated, and dilated RER, and also the suppressed PRL mRNA expression. The activity of protein kinase C (PKC), which mediates PRL gene expression, tended to be elevated by estrogen and suppressed by bromocriptine. Therefore, it is considered that the ultrastructural and quantitative changes in PRL mRNA expression evoked by estrogen and bromocriptine may be mediated by the intracellular signal transduction system, including PKC.

    Notes: Abstract  Adrenomedullin (AM) is a novel vasorelaxant peptide isolated from pheochromocytoma. Proadrenomedullin N-terminal 20 peptide (PAMP) is a hypotensive peptide generated by posttranslational enzymatic processing of a 185-amino acid pro-AM molecule, the same precursor as AM. In this study, we investigated localizations of these peptides by immunocytochemistry and AM mRNA by non-radioisotopic in situ hybridization followed by the streptavidin and biotin complex (ABC) method and catalyzed signal amplification (CSA) in the rat adrenal medulla and gastric mucosa. In the gastric mucosa, both AM- and PAMP-like immunoreactivities were found in the neuroendocrine cells, but PAMP-positive cells were more abundant than AM-positive ones. By immunoelectron microscopy, AM and PAMP were localized exclusively in the secretory granules. The distribution pattern of AM mRNA-positive cells, only a limited portion of which had AM and/or PAMP, was also similar to that of the two peptides. But AM mRNA was detected also in a few epithelial cells as well as neuroendocrine cells. The two peptides might play an important role in the control of local circulation in the rat stomach.

    Notes: Summary This immunohistochemical study disclosed that 9 of 15 GH secreting pituitary adenomas contained α subunit positive cells. These cases also contained PRL positive adenoma cells, but LHβ was negative. Of these 9 cases, 4 cases showed occasional FSHβ containing cells, 2 of these also contained a few TSHβ positive cells. By mirror section technique, variable numbers of adenoma cells were found to contain both GH and α subunit. Immunoelectron microscopically, both GH and α subunit were localized in secretory granules which suggested their co-release from the tumour cells. The presence of GH and α subunit in rough endoplasmic reticulum indicated their active production in the tumour. In the normal adult anterior pituitary gland, about 10% of GH cells contain FSH α,β and LHβ subunits and had appearances suggesting the co-production of GH and FSH as well as LH. The colocalization of GH and FSH α is considered to be associated with the neoplastic transformation GH cells which possess the intrinsic potentiality of differentiation toward α subunit. However, the mechanism for the lack or deficiency ofβ subunits in the neoplastic condition remains to be further investigated.

    Notes: Summary Combined immunohistochemical staining (IHCS) and enzyme histochemical staining (EHCS) methods for light microscopy (LM) and electron microscopy (EM) are reported, using oestrogeninduced rat pituitary tumours. For LM, combined staining for alkaline phosphatase and acid phosphatase by EHCS, using the azo dye method, and for prolactin and ACTH by IHCS, using the enzyme-labelled antibody method, gave the best results on 1 μm glycol methacrylate sections. For EM, combined staining by EHCS on 30 μm tissue sections followed by IHCS for prolactin on ultrathin Epon sections (enzyme-labelled antibody method) provided acceptable results. By these combined staining methods, the neoplastic prolactin cells were shown to have close affinity to rich alkaline phosphatase-positive capillaries and to possess an alkaline phosphatase-positive cell membrane. Furthermore, they revealed acid phosphatase-positive lysosomal and secretory granules. These combined staining methods may be valuable in studies on the actual functional status of cells.

    Notes: Summary There have been many reports on radioisotopicin situ hybridization (ISH) studies for the demonstration of pituitary hormone mRNAs in normal pituitary gland and pituitary adenomas. Recent studies have revealed that non-radioisotopic ISH has several advantages over the radioisotopic method. Using ISH with biotinylated oligonucleotide probes, we have been able to localize various pituitary hormone mRNAs in paraffin wax or frozen sections of rat normal pituitary gland and human pituitary adenomas. For control studies we used ISH with sense probes, ISH without probes, pretreatment with ribonuclease, ISH with a probe for β-actin and Northern blot hybridization. Using biotinylated probes, gene transcripts of rat growth hormone and prolactin were detected by Northern blot hybridization. The same biotinylated probes were used not only for light microscope ISH but also for the electron microscopical demonstration of rat growth hormone and prolactin mRNAs on the polysomes of the rough endoplasmic reticula. It is emphasized that biotinylated oligonucleotide probes are useful for the analysis of pituitary endocrine function because they are applicable to the three hybridization methods, namely, Northern blot hybridization and ISH at the light and electron microscope levels.
