    Notes: To elucidate the molecular basis of cognitive memory formation in the primate, transcriptional activation during learning of a visual pair-association (PA) task was evaluated systematically along the occipito-temporo-hippocampal pathway in the macaque monkey brain. Split-brain monkeys were used for intra-animal comparison, which enables elimination of animal-to-animal variation in gene expression. We found that the expression of the mRNA of an immediate-early gene (IEG), zif268, was up-regulated selectively in the perirhinal cortex (area 36) during the formation of PA memory compared to that during learning of a visual control task. The mRNA expression levels of another IEG, c-jun, were not up-regulated during the PA learning in any cortical areas examined. We also showed that cells strongly expressing zif 268 mRNA accumulated in patches in area 36 during learning of the PA task. As the zif 268 gene encodes a transcription factor, these results suggest that the activation of zif 268 mRNA in area 36 may function as a trigger of the cascade of gene activation that leads to cellular events underlying neuronal reorganization for visual long-term memory formation.

    Notes: In humans and monkeys, memory consists of various components which were initially revealed through neuropsychological studies of amnesic patients. One memory component, the long-term memory about facts and events (declarative memory), has been shown to require the integrity of the medial temporal lobe and the neocortex. To investigate a map of gene expression during declarative memory formation in the primate, we monitored expression of immediate early genes in the temporal lobe of macaque monkeys. We trained the monkeys to learn visual associative memory tasks, and immunohistochemically analysed the expression of protein products of immediate early genes. We found that Zif268, a transcription factor regulated by neuronal activity, accumulated in patches in the anterior temporal lobe during visual stimulus-stimulus association learning, whereas other transcription factors, c-Fos and JunD, were not. By contrast, the patchy expression of Zif268 was not observed during another type of learning, visual discrimination learning. These results suggest that Zif268 activates a gene cascade related to the formation of long-term memory in the primate.

    Notes: Summary 4-Vinyl [2.2]paracyclophane (1) was successfully prepared as white crystals in 45 % yield via three step reactions. Monomer 1 was homopolymerizable with 2,2′-azobis(isobutyronitrile) (AIBN), boron trifluoride etherate, and butyllithium. Monomer 1 was copolymerizable with methyl acrylate and p-chlorostyrene in the presence of AIBN in benzene at 60 °C. It was found that monomer 1 was less conjugative and more electron-donating than styrene.
