    Notes: Carbon-13 (C-13) solid-state NMR spectroscopy was used to investigate the chemical nature of organic C in mineral topsoil sampled under grassland and adjacent recently established (10–17 years old) coniferous forest (Douglas fir, Pseudotsuga menziesii; Ponderosa pine, Pinus ponderosa; Corsican pine, Pinus nigra) at two sites (Craigieburn, Cave Stream) in the South Island of New Zealand. This involved using a Cross-Polarized/Magic-Angle Spinning (CP/MAS) technique to identify different chemical forms of soil organic C, whilst Proton Spin Relaxation Editing (PSRE) was used to determine different ‘pools’ of soil organic C. Results obtained from the Craigieburn soils (0–5 cm) were more promising than those obtained from the Cave Stream soils (0–10 cm) because the total Fe content was smaller, and indicated a shift towards more recalcitrant forms of organic C in soil under trees compared with grassland, which might reflect reduced inputs of fresh organic matter to the soil under trees.

    Notes: Abstract We examined the effects of conifers on the forms of P in low-fertility tussock grassland soils using 31P nuclear magnetic resonance (NMR) and soil P fractionation. Results from field and glasshouse experiments clearly demonstrated that conifers enhanced the mineralization of labile (and to a lesser extent more resistant) forms of soil organic P which, in turn, increased amounts of labile inorganic P in the soil. These findings have important implications for P availability and long-term sustainable management of grassland soils in New Zealand.

    Notes: Abstract The use of instrumental neutron activation analysis (INAA) in determining selected elements in biological materials has been reported by numerous authors. Some of these have detailed the use of INAA to determine an elemental analysis in tobaccos of various geographical origins. This paper describes the first use of INAA to measure the distribution of selected inorganic elements within the tobacco leaf at a single plant stalk position for one tobacco curing routine. Three replicate plots of a common bright tobacco cultivar were grown under normal cultural conditions characteristic for the bright variety. The tobacco leaves were sampled at selected positions in the leaf. The brigh tobacco was cured in a conventional flue-curing barn using standard practices. Immediately after collection, each individual sample was freeze-dried, crushed and sampled. The leaf midrib samples were prepared using a similar procedure. A subsample of about 100 mg was taken from each sample, sealed in a polyethylene bag, irradiated in a thermal neutron flux of 2×1017 n·m−2·s−1 in a pneumatic rabbit system, and subsequently counted to obtain the reported data. A standard reference material was used as a comparator to yield relative elemental concentrations for Ca, Mg, Mn, Na, K, Cl and Br. The data show that chlorine, potassium, sodium and calcium have definite concentration trends within the tobacco leaf. The data also show that some elements, e.g. Mn, were more uniformly distributed throughout the leaf.

    Notes: Silicon-29 n.m.r. was used to study the products from trimethylsilylation of minerals. Chemical shifts are reported for four model compounds, viz. the monomer, dimer, linear trimer (from natrolite) and cyclic tetramer (from laumontite). Information is then obtained from the products of trimethylsilylation of the anhydrous calcium silicates wollastonite and pseudo-wollastonite. In the latter case it is shown that the principal products are the four model compounds, and percentage yields are obtained for these.

    Notes: Substituent effects on the 19F chemical shifts of a series of 3- and 4-XCH2-substituted fluorobenzenes have been studied in five different solvents (dimethyl sulphoxide, acetone, chloroform, carbon tetrachloride and benzene). The results show that in both series substituent effects (other than inductive effects) contribute in at least some cases to the overall shifts, but that these showed little solvent dependence. The meta series showed a proportionately greater solvent dependence than the para series, and this is attributed to the greater importance of ‘through solvent’ direct field effects in the former.

    Notes: Selective pulsed excitation was used in a technique to suppress the resonance due to a per-12C-isotopomer so as to overcome dynamic range problems when observing ‘13C satellite’ lines. The technique was used to obtain nuclear spin parameters from the 13C satellites in 31P resonance for the racemic isomer of P,P′-dimethyl-P,P′-diphenyldiphosphine disulphide, [MePhP(:S)]2.

    Description: : Phosphorylation plays an important role in cellular signal transduction. Current phosphorylation-related databases often focus on the phosphorylation sites, which are mainly determined by mass spectrometry. Here, we present PhosphoNetworks, a phosphorylation database built on a high-resolution map of phosphorylation networks. This high-resolution map of phosphorylation networks provides not only the kinase–substrate relationships (KSRs), but also the specific phosphorylation sites on which the kinases act on the substrates. The database contains the most comprehensive dataset for KSRs, including the relationships from a recent high-throughput project for identification of KSRs using protein microarrays, as well as known KSRs curated from the literature. In addition, the database also includes several analytical tools for dissecting phosphorylation networks. PhosphoNetworks is expected to play a prominent role in proteomics and phosphorylation-related disease research.     Description: Meprin metalloproteases are abundantly expressed in the brush-border membranes of kidney proximal tubules. Meprins are implicated in ischemia-reperfusion (IR)-induced renal injury and diabetic nephropathy. The protein kinase A (PKA) signaling pathway modulates extracellular matrix metabolism in diabetic kidneys. The present study evaluated isoform-specific interactions between the catalytic subunit of PKA (PKA C) and meprins. To this end, cytosolic-enriched kidney proteins from meprin αβ double knockout mice, and purified forms of recombinant mouse PKA Cα, Cβ1, and Cβ2, were incubated with activated forms of either homomeric meprin A or meprin B. The cleaved protein products were subjected to SDS-PAGE and analyzed by Coomassie staining and Western blot analysis. While meprin A only cleaved PKA Cβ1, meprin B cleaved all three PKA C isoforms. Analysis of the proteolytic fragments by mass spectrometry revealed that meprin A and B cleave the PKA C isoforms at defined sites, resulting in unique cleavage products. Michaelis-Menten enzyme kinetics demonstrated that meprin B-mediated cleavage of PKA Cα occurs at a rate consistent with that of other physiologically relevant meprin substrates. Meprin cleavage decreased the kinase activity of PKA Cα, Cβ1, and Cβ2. PKA C levels were higher in diabetic kidneys, with evidence of in vivo fragmentation in wild-type diabetic kidneys. Confocal microscopy showed localization of meprin A in the glomeruli of diabetic kidneys. At 3 h post-IR, PKA C levels in proximal tubules decreased compared with distal tubules, which lack meprins. These data suggest that meprins may impact kidney injury, in part, via modulation of PKA signaling pathways.
