    Notes: A population of 108 common bean recombinant inbred lines (RILs) (F5:6-9), derived from a leafhopper (Empoasca fabae and E. kraemeri)-susceptible cultivar (‘Berna’) and a leafhopper-resistant line (EMP 419) was used to identify molecular markers genetically linked to leafhopper resistance and seed weight. Bulked segregant analysis and quantitative trait analysis identified eight markers that were associated with resistance to E. fabae, and four markers that were associated with E. kraemeri resistance. Three markers were associated with resistance to both species. A partial linkage map of the bean genome was constructed. Composite interval mapping identified quantitative trait loci (QTL) for resistance to both leaf hopper species on core-map linkage groups B1, B3 and B7. QTL for seed weight were found close to the locus controlling testa colour and an α-phaseolin gene.

    Notes: Similar to leptin, ciliary neurotrophic factor (CNTF) suppresses appetite and selectively reduces body fat in leptin-deficient ob/ob mice. To assess the relative importance of specific regions of the hypothalamus in mediating these effects, we administered a CNTF analogue (CNTFAx15) or leptin to mice made obese by administration of gold thioglucose (GTG), which destroys a well-defined portion of the medial basal hypothalamus. CNTFAx15 treatment reduced appetite and body weight in obese GTG-lesioned C57BL/6 mice, whereas leptin failed to effect similar changes regardless of whether treatment was initiated before or after the lesioned mice had become obese. Because leptin does not reduce food intake or body weight in most forms of obesity (a condition termed ‘leptin resistance’), we also investigated the actions of leptin in GTG-lesioned leptin-deficient (ob/ob) mice. By contrast to C57BL/6 mice, leptin treatment reduced food intake and body weight in GTG-lesioned ob/ob mice, although the effect was attenuated. To further compare the neural substrates mediating the anorectic actions of leptin and CNTF, we determined the patterns of neurone activation induced by these proteins in the hypothalamus of intact and GTG-lesioned mice by staining for phosphorylated signal transducer and activator of transcription 3 (pSTAT3). CNTFAx15 stimulated robust pSTAT3 signalling in neurones of the medial arcuate nucleus in both intact and lesioned C57BL/6 and ob/ob mice. Leptin administration stimulated pSTAT3 signalling in only a few neurones of the medial arcuate nucleus in intact or lesioned C57BL/6 mice, but elicited a robust response in intact or lesioned ob/ob mice. By contrast to CNTFAx15, leptin treatment also resulted in prominent activation of STAT3 in several areas of the hypothalamus outside the medial arcuate nucleus. This leptin-induced pSTAT3 signal was at least as prominent in intact and GTG-lesioned C57BL/6 mice as it was in ob/ob mice, and thus was not correlated with appetite suppression or weight loss. These results indicate that the medial arcuate nucleus is a key mediator of appetite suppression and weight loss produced by CNTF and leptin, whereas GTG-vulnerable regions play a role only in leptin-induced weight loss. Other regions of hypothalamus in which pSTAT3 signal is induced by leptin may regulate energy metabolism through mechanisms other than appetite reduction.

    Notes: Abstract The evolution of spatial pattern is a central issue in developmental biology. Turing's (Phil. Trans. R. Soc. Lond. B237, 37–72, 1952) chemical theory of morphogenesis is a seminal contribution. In this talk I give a personal and necessarily limited view of its impact on mathematical and developmental biology. I briefly describe some of the interesting mathematical aspects of Turing's reaction-diffusion mechanism and discuss some of the different models which Turing's vision inspired. The emphasis throughout is on the practical biological applications of the various theories.

    Notes: Abstract A non-linear stability analysis using a multi-scale perturbation procedure is carried out on the practical Thomas reaction-diffusion mechanism which exhibits bifurcation to non-uniform states. The analytical results compare favourably with the numerical solutions. The sequential patterns generated by this model by variations in a parameter related to the reaction-diffusion domain indicate its capacity to represent certain key morphogenetic features required in a recent model by Kauffman for pattern formation in theDrosophila embryo.

    Notes: Abstract We consider a simple cell-chemotaxis model for spatial pattern formation on two-dimensional domains proposed by Oster and Murray (1989,J. exp. Zool. 251, 186–202). We determine finite-amplitude, steady-state, spatially heterogeneous solutions and study the effect of domain growth on the resulting patterns. We also investigate in-depth bifurcating solutions as the chemotactic parameter varies. This numerical study shows that this deceptively simple-chemotaxis model can produce a surprisingly rich spectrum of complex spatial patterns.

    Notes: Abstract We consider a cell-chemotaxis model which can generate spatially heterogeneous patterns in cell density and chemoattractant. A local perturbation about the uniform steady state propagates across the domain leaving behind a steady state pattern of standing peaks and troughs. We investigate this patterning process analytically and obtain estimates for the pattern wavelength and speed of spread.     Notes: Abstract We propose a model mechanism for the initiation and spatial positioning of teeth primordia in the alligator, Alligator mississippiensis. Detailed embryological studies by Westergaard and Ferguson (1986, 1987, 1990) have shown that jaw growth plays a crucial role in the developmental patterning of the tooth initiation process. Based on biological data we develop a dynamic patterning mechanism, which crucially includes domain growth. The mechanism can reproduce the spatial pattern development of the first seven teeth primordia in each half jaw of A. mississippiensis. The results for the precise spatio-temporal sequence compare well with experiment. Simulation of the model also predicts that certain transplantations can alter the spatial sequence of teeth primordia initiation.
